Shapeshifting as Infrastructural Storytelling: Comics about the Taxibot’s Conflicting Narratives

What are the stories we tell about infrastructures and what stories do infrastructures tell (about) us? We propose a paper in a hybrid verbo-visual format, including comic-pages created by Giada Peterle and based on Tina Harris’s keynote at the 2022 GMHC conference, autoethnographic notes, and visuals collected during fieldwork. Through experimenting with graphic storytelling, we highlight examples of infrastructural revelation and concealment, drawing on the figure of the shapeshifter as both a metaphor and a method for mobilising infrastructural imagination. What unites shapeshifters in many of the stories and myths we read is how they are taken up in different ways; how they simultaneously present both the potential to improve human lives as well as produce fear due to their unpredictability. By focusing specifically on the narrative of one shapeshifting infrastructure—the Taxibot, a vehicle designed to cut down on carbon emissions and improve efficiency at airports—we use comics as a research practice for exploring this metaphor and developing a broader understanding of how mobile lives and imaginaries shape infrastructure (and vice versa). We argue that paying closer attention to storytelling can generate new understandings of the uneven nexus between infrastructures and mobile lives, weaving in our understanding of infrastructural im/mobilities.
Exchange: Explaining the Passage of Universal Healthcare in Thailand

What explains the passage of Thailand's landmark universal healthcare (UHC) policy? In separate contributions, Selway and Harris emphasized the role of electoral rules and political parties, on one hand, and “professional movements” of developmentally minded state bureaucrats on the other. Which is correct? In this article, Selway and Harris respond to each other's work. While Selway agrees that the actions of the professional movement constitute an underappreciated necessary condition for universal healthcare in Thailand, he argues that Harris overstates the role of the movement in implementation. Harris defends his position and maintains that an institution-focused account is insufficient, arguing that the actions of Thailand's Rural Doctors’ Movement not only explain universal healthcare but also gave rise to the very electoral rule changes that Selway argues were so critical to facilitating universal coverage. Selway responds to these criticisms, and the two researchers jointly consider implications for causation, qualitative research, and policymaking theory.
Determinants of muscle carnosine content

<P>The main determinant of muscle carnosine (M-Carn) content is undoubtedly species, with, for example, aerobically trained female vegetarian athletes [with <I>circa</I> 13 mmol/kg dry muscle (dm)] having just 1/10th of that found in trained thoroughbred horses. Muscle fibre type is another key determinant, as type II fibres have a higher M-Carn or muscle histidine containing dipeptide (M-HCD) content than type I fibres. In vegetarians, M-Carn is limited by hepatic synthesis of β-alanine, whereas in omnivores this is augmented by the hydrolysis of dietary supplied HCD’s resulting in muscle levels two or more times higher. β-alanine supplementation will increase M-Carn. The same increase in M-Carn occurs with administration of an equal molar quantity of carnosine as an alternative source of β-alanine. Following the cessation of supplementation, M-Carn returns to pre-supplementation levels, with an estimated t<SUB>1/2</SUB> of 5–9 weeks. Higher than normal M-Carn contents have been noted in some chronically weight-trained subjects, but it is unclear if this is due to the training <I>per se</I>, or secondary to changes in muscle fibre composition, an increase in β-alanine intake or even anabolic steroid use. There is no measureable loss of M-Carn with acute exercise, although exercise-induced muscle damage may result in raised plasma concentrations in equines. Animal studies indicate effects of gender and age, but human studies lack sufficient control of the effects of diet and changes in muscle fibre composition.</P>

The blood-brain barrier (BBB) is composed of the brain capillaries, which are lined by endothelial cells displaying extremely tight intercellular junctions. Several attempts at creating an in vitro model of the BBB have been met with moderate success as brain capillary endothelial cells lose their barrier properties when isolated in cell culture. This may be due to a lack of recreation of the in vivo endothelial cellular environment in these models, including nearly constant contact with astrocyte foot processes. This work is motivated by the hypothesis that growing endothelial cells on one side of an ultra-thin, highly porous membrane and differentiating astrocyte or astrogliomal cells on the opposite side will lead to a higher degree of interaction between the two cell types and therefore to an improved model. Here we describe our initial efforts towards testing this hypothesis including a procedure for membrane fabrication and methods for culturing endothelial cells on these membranes. We have fabricated a 1 $\mu\textrm{m}$ thick, 2.0 $\mu\textrm{m}$ pore size, and 55% porous membrane with a very narrow pore size distribution from low-stress silicon nitride (SiN) utilizing techniques from the microelectronics industry. We have developed a base, acid, autoclave routine that prepares the membranes for cell culture both by cleaning residual fabrication chemicals from the surface and by increasing the hydrophilicity of the membranes (confirmed by contact angle measurements). Gelatin, fibronectin, and a 50/50 mixture of the two proteins were evaluated as potential basement membrane protein treatments prior to membrane cell seeding. All three treatments support adequate attachment and growth on the membranes compared to the control.
FIXED POINT THEOREMS FOR INFINITE DIMENSIONAL HOLOMORPHIC FUNCTIONS

This talk discusses conditions on the numerical range of a holomorphic function defined on a bounded convex domain in a complex Banach space that imply that the function has a unique fixed point. In particular, extensions of the Earle-Hamilton Theorem are given for such domains. The theorems are applied to obtain a quantitative version of the inverse function theorem for holomorphic functions and a distortion form of Cartan's unique-ness theorem.
Cellular Recording Using CRISPR Technologies

Robust recording of environmental and biological information over time has been a key challenge limiting the development of useful cell-based sentinels that can monitor environments and cellular states in open settings. In this talk, I describe a new framework to store environmental and biological temporal information directly into the genomes of a bacterial population. Using this framework, we recently developed a “biological tape recorder” in which biological signals trigger intracellular DNA production that is then recorded by the CRISPR-Cas adaptation system. This approach enables stable recording of temporal and lineage information over multiple days, which can be accurately reconstructed by sequencing CRISPR arrays. Using CRISPR-based cellular recording in complex microbial communities, we track and dissect horizontal gene transfer events in real time across the human gut microbiome and in vivo colonization of bacteria. Robust temporal recording of environmental and biological signals using engineered cells enables new applications for chronicling cellular events on a large scale, at a higher resolution, and in otherwise inaccessible settings.
