        Alterations of mRNA and lncRNA profiles associated with the extracellular matrix and spermatogenesis in goats

        Objective: Spermatozoa are produced within the seminiferous tubules after sexual maturity. The expression levels of mRNAs and lncRNAs in testicular tissues are different at each stage of testicular development and are closely related to formation of the extracellular matrix (ECM) and spermatogenesis. Therefore, we set out to study the expression of lncRNAs and mRNAs during the different developmental stages of the goat testis.Methods: We constructed 12 RNA libraries using testicular tissues from goats aged 3, 6, and 12 months, and studied the functions of mRNAs and lncRNAs using the gene ontogeny (GO) and Kyoto encyclopedia of genes and genomes (KEGG) databases. Relationships between differentially expressed genes (DEGs) were analyzed by lncRNA-mRNA coexpression network and protein-protein interaction network (PPI). Finally, the protein expression levels of matrix metalloproteinase 2 (MMP2), insulin-like growth factor 2 (IGF2), and insulin-like growth factor-binding protein 6 (IGFBP6) were detected by western blotting.Results: We found 23, 8, and 135 differentially expressed lncRNAs and 161, 12, and 665 differentially expressed mRNAs that were identified between 3 vs 6, 6 vs 12, and 3 vs 12 months, respectively. GO, KEGG, and PPI analyses showed that the differential genes were mainly related to the ECM. Moreover, MMP2 was a hub gene and co-expressed with the lncRNA TCONS-0002139 and TCONS-00093342. The results of quantitative reverse-transcription polymerase chain reaction verification were consistent with those of RNA-seq sequencing. The expression trends of MMP2, IGF2, and IGFBP6 protein were the same as that of mRNA, which all decreased with age. IGF2 and MMP2 were significantly different in the 3 vs 6-month-old group (p<0.05).Conclusion: These results improve our understanding of the molecular mechanisms involved in sexual maturation of the goat testis.         TRAJECTORY PLANNING ALGORITHM USING GAUSS PSEUDOSPECTRAL METHOD BASED ON VEHICLE-INFRASTRUCTURE COOPERATIVE SYSTEM

        Vehicle-infrastructure cooperative systems can potentially enhance both traffic safety and efficiency by conducting coordinated control through the interactive strategy between the vehicles and the infrastructure. In this study, the interactive strategy of a vehicle infrastructure cooperative system is designed. Lane change maneuver is a conventional behavior in driving. Thus, this paper proposes a trajectory planning algorithm based on a Gauss pseudo-spectral method that is applied to the intelligent vehicle-infrastructure cooperative system in the lane change scenario. A road side unit calculates the planning trajectory using collected vehicle information and sensor data and then sends the trajectory planning advice to the designated vehicle. The Gauss pseudo-spectral method is used to obtain the planning trajectory, which effectively helps solve the discontinuous optimization problems in partial conditions. It transforms the optimal control problem of dynamic systems into a nonlinear programming problem using the orthogonal collocation method to discretize the objective function and various constraints of the optimization problem. Furthermore, the ssuential quadratic programming method is used to solve the problem numerically. The effectiveness of the proposed method and interactive strategy are demonstrated through simulations and experimental results.

        Influence of structure coupling effect on damping coefficient of offshore wind turbine blades

        The aim of this study was to explore the influence of structure coupling effect on structural damping of blade based on the blade vibration characteristic. For this purpose, the scaled blade model of NREL 5 MW offshore wind turbine was processed and employed in the wind tunnel test to validate the reliability of theoretical and numerical models. The attenuation curves of maximum displacement and the varying curves of equivalent damping coefficient of the blade under the rated condition were respectively compared and analyzed by constructing single blade model and whole machine model. The attenuation law of blade dynamic response was obtained and the structure coupling effect was proved to exert a significant influence on the equivalent damping coefficient. The results indicate that the attenuation trend of the maximum displacement response curve of the single blade varies more obviously with the increase of elastic modulus as compared to that under the structure coupling effect. In contrast to the single blade model, the varying curve of equivalent damping coefficient with the period is relatively steep for the whole machine model. The findings are of great significance to guide the structure design and material selection for wind turbine blades.

        Local damage detection of a fan blade under ambient excitation by three-dimensional digital image correlation

        Damage detection based on dynamic characteristics of a structure is one of important roles in structural damage identification. It is difficult to detect local structural damage using traditional dynamic experimental methods due to a limited number of sensors used in an experiment. In this work, a non-contact test stand of fan blades is established, and a full-field noncontact test method, combined with three-dimensional digital image correlation, Bayesian operational modal analysis, and damage indices, is used to detect local damage of a fan blade under ambient excitation without use of baseline information before structural damage. The methodology is applied to detect invisible local damage on the fan blade. Such a method has a seemingly high potential as an alternative to detect local damage of blades with complex high-precision surfaces under extreme working conditions because it is a noncontact test method and can be used under ambient excitation without human participation.

        Estimation Algorithm of Minimum Dwell Time in Precision Cylindrical Plunge Grinding Using Acoustic Emission Signal

        This work presents an investigation of the minimum dwell time during the precision cylindrical-plunge grinding process. Accordingto the model of acoustic emissions proposed in earlier work, two estimation algorithms of the dwell time, focused on the size errorand the surface roundness of the machined workpiece, are deduced from the models of the material removal and the surfaceroundness, respectively. Based on the proposed estimation algorithms, an on-line estimation method of the minimum dwell time isdeveloped using the measurement and analysis of the acoustic emission signal. A series of experiments of grinding C45 steel areconducted to confirm the validity of the proposed estimation algorithms and determine the model coefficient. The ability of theestimation algorithms to predict the minimum dwell time is deemed necessary for optimal control of the grinding process. In addition,the proposed model provides a new approach to estimate the machined qualities of a workpiece such as the surface roughness andsize error using the dwell time of the grinding process.
