      • 입제의 수면처리에 의한 주요수도해충의 동시방제

        본 시험은 유기인계 살충제인 Terracur P입제 및 Lebaycid 입제와 유기염소계 살충제인 상감마입제$(\gamma-BHC 입제)$를 수면시용 하였을때 수도요해충인 이화명충, 끝 동매미충, 애멸구 및 흰둥멸구에 대한 동시방제가능성과 방제적기를 구명하고 약제간의 방제효과를 비교코저 실시하였다. 얻어진 시험결과는 다음과 같다. 1) 약제처리후 부화직후 이화명충 접종 시험에 있어서 상감마와 Terracur P는 높은 살충율을 보였으나 Lebaycid의 살충율은 낮았다. 約 $50\%$ 살충력 잔효일수에 있어서 상감마는 8-14日이었고 Terracur P는 6일간이었다. 2) 당초 계획된 방제적기에 관한 정보는 얻을 수 없었다. 즉 주어진 일간격이 좁아 유의한 차를 얻을 수 없었다. 3) 1,2화기 이화명충에 대한 방제효과는 Terracur P=상감마>Lebaycid의 순이었다. 4) Terracur P와 Lebaycid 입제의 처리는 끝동매미충의 방제효과가 좋았으나 상감마$(\gamma-BHC 입제)$의 방제효과는 기대할 수 없었다. 끝동매미충에 대한 Terracur P와 Lebaycid입제의 잔효력은 30일 이상이었다. 5) 애멸구와 흰등별구에 대한 시험은 충의 서식밀도가 낮기 때문에 약제간 방제효과를 얻을 수 없었다. 6) 공시약제중 Terracur P와 Lebaycidd입제는 이화명충과 끌동매미충의 동시방제제로서 사용할 수 있을 것 같다. This experiment was conducted to find out the simultaneous control effect of granulated insecticides on the several major rice insect pests in Korea. The granulated insecticides used were Terracur P$5\%$ (one of the phosphoric insecticides), Lebaycid $5\%$, and Sang-gamma (gamma-BHC) $6\%$. Those insecticides were broadcasted by hand on the paddy field at the ratio of 3kg. in June and 4kg. in August per 10 a, and the date of insecticidal application was based on the maximum moth occurrence of the rice stem borer in 1969. The rice insect pests tested in this study were rice stem borer (Chilo suppressalis W.), green rice leafhopper (Nephotettix bipunctatus cincticeps U.), smiller brown planthopper (Laodelphax striatellus F.), and white back planthopper (Sogotella furcifera H.). For the study of residual effectiveness of the insecticides, the rice stem borer larvae just hatched from the eggs were installed on the sheath of rice plants collected from the paddy field after insecticidal treatment, and the mortality was made at 48 hours after installation. High mortality was observed in Terracur P and Sang-gamma treatments and low mortality in Lebaycid treatment. The duration of residual effects giving about 50% mortality was about 14 days in Sang-gamma and about 6 days in Terracur P treatments. The study on the effectiveness of the insecticides to the rice stem borer in the paddy field was made by the number of dead blades and white heads of the rice plants injured by the larvae, and the number of survival larvae in the straws after crop harvest. The order of insecticidal effectiveness to the rice stem borer was Terracur P=Sang-gamm>Lebaycid. The study on the effectiveness of the insecticides to the leafhoppers and planthoppers in the paddy field was made by the population density by sweeping (5 times) at the given day intervals after treatments. Terracur P was highly effective for the control of the green rice leafhoppers and Lebaycid was moderately effective, but Sang-gamma seemed not to be effective. The effectiveness of the insecticides tested to the planthoppers was not clear, and it seemed to related with the low insect population density. Study on the determination of proper timing of insecticidal application was not also made clear, and it seemed to be short in the range of day intervals in the insecticidal application. For the study on the control effects of the rice insect pests, rice yield per 100 hills per plot was observed. There was no statistically significant difference among the treatments in rice yields, but the order of yield (Terracur P>Sang-gamma>Lebaycid>Control) was paralleled with the tends of control effects above mentioned. In a view point of simultaneous control of the rice stem borers and green rice leafhoppers by the application of granulated insecticides, Terracur P and Lebaycid were more effective than Sang-gamma which have been used.

        Improved antimicrobial activity of Pediococcus acidilactici against Salmonella Gallinarum by UV mutagenesis and genome shuffling

        <P>Pediococcus acidilactici is a widely used probiotic, and Salmonella enterica serovar Gallinarum (SG) is a significant pathogen in the poultry industry. In this study, we improved the antimicrobial activity of P. acidilactici against SG using UV mutation and genome shuffling (GS). To improve antimicrobial activity against SG, UV mutagenesis was performed against wild-type P. acidilactici (WT), and five mutants showed improved antimicrobial activity. To further improve antimicrobial activity, GS was performed on five UV mutants. Following GS, four mutants showed improved antimicrobial activity compared with the UV mutants and WT. The antimicrobial activity of GS1 was highest among the mutants; however, the activity was reduced when the culture supernatant was treated with proteinase K, suggesting that the improved antimicrobial activity is due to a proteinous substance such as bacteriocin. To validate the activity of GS1 in vivo, we designed multi-species probiotics and performed broiler feeding experiments. Groups consisted of no treatment (NC), avilamycin-treated (PC), probiotic group 1 containing WT (T1), and probiotic group 2 containing GS1 (T2). In broiler feeding experiments, coliform bacteria were significantly reduced in T2 compared with NC, PC, and T1. The cecal microbiota was modulated and pathogenic bacteria were reduced by GS1 oral administration. In this study, GS1 showed improved antimicrobial activity against SG in vitro and reduced pathogenic bacteria in a broiler feeding experiment. These results suggest that GS1 can serve as an efficient probiotic, as an alternative to antibiotics in the poultry industry.</P>

        Plasma Arc Welding of 780CP High Strength Steel Sheet Lap Joint for Tensile Strength of 100% Compared to Base Metal

        Homogeneous and heterogeneous PAW (plasma arc welding) experiments were conducted to obtain weld beads with tensile strength of 100% or more compared to that of the base metal in lap joint fillet welding of CP (complex phase) 780 MPa grade high strength steel sheet. Homogeneous and heterogeneous welding mean welding technic without and with welding wire in each. Common basic welding parameters of both homogeneous and heterogeneous PAW wire were applied, including welding speed of 21 cm/min, arc length of 3.5 mm, weaving amplitude of 3 mm, work angle of 30°, and Ar 99.99% shielding gas with 12 L/min. In homogeneous PAW, the optimized additional welding conditions were a welding current of 90 and 95 A, push angle of 20°, and weaving frequency of 5 Hz. The fracture strengths of all the samples were less than that of the base metal, and all fractures occurred at the weld bead fusion zone due to insufficient reinforcement. In heterogeneous PAW, the optimized additional welding conditions were a welding current of 125 A, push angle of 0°, weaving frequency of 4 Hz, and wire feed speeds of 1.53 and 1.38 m/min. Moreover, the welding wire was fed from behind the arc. The fracture strengths of all the samples were greater than that of the base metal, and fractures occurred at the base metal or fusion zone boundary. The reason for higher tensile in heterogeneous PAW wire is deduced as the large cross-section area of reinforcement provided by the welding wire comparing to homogeneous welding. It is well known that small radius of welding heat source clearly produces narrow heat affected zone. Therefore, the PAW produces relatively narrower heat affected zone, then resulted as fully enough strength of weldment. This is another reason of the current achievement.

        <P>BACKGROUND AND PURPOSE: Intravenous (IV) thrombolysis with recombinant tissue plasminogen activator (rt-PA) has demonstrated favorable clinical outcomes in a 3-6 h window in patients selected with perfusion/diffusion mismatch. However, the advantages of combined IV and intraarterial (IA) thrombolysis after 3 h of stroke onset are unexplored. METHODS: Acute ischemic stroke patients with persistent occlusion of intracranial large arteries were screened prospectively for thrombolysis by evaluating perfusion/diffusion mismatch on MRI. The IV rt-PA was initiated within 3-6 h, and additional urokinase (UK) was then administered via the IA route after angiography. RESULTS: Four patients had middle cerebral artery occlusion and one patient had an internal carotid artery occlusion. The median time from the symptom onset to the initiation of IV therapy and to the initiation of IA treatment was 215 +/- 30 min and 292 +/- 41 min, respectively. The median National Institutes of Health Stroke Scale (NIHSS) scores were as follows: initial, 13; immediately after IA treatment, 8; at 24 h, 5; and at 7 days, 3. The Thrombolysis in Myocardial Infarction (TIMI) score after the completion of thrombolysis was 2-3. Four patients without intracerebral hemorrhage recovered completely or exhibited mild disability and one patient with hemorrhage also demonstrated a favorable outcome. CONCLUSION: This preliminary result suggests that if a significant perfusion/diffusion mismatch on MRI is identified, a sequential combination thrombolysis of IV rt-PA and IA UK is potentially beneficial in moderate to severe acute ischemic stroke patients who are treated within 3-6 h after symptom onset.</P>

        Dipeptide-functionalized polyamidoamine dendrimer-mediated apoptin gene delivery facilitates apoptosis of human primary glioma cells

        <P><B>Abstract</B></P> <P>Glioblastoma multiform (GBM) is the most frequent and aggressive form of brain tumors in adults. However, the development of more efficient and safe nonviral vector gene therapy represents a promising therapeutic approach, using a tumor-specific killer gene, named apoptin. In this study, we describe the efficacy of non-viral gene delivery vectors, the amino acid-conjugated PAMAM derivatives (PAMAM-H-R and PAMAM-H-K) in delivering a therapeutic gene, displaying affinity toward human primary glioma cells (GBL-14 cells) and dermal fibroblasts. We analyzed transfection efficiency, using luciferase (Luci) and a pDNA encoding for enhanced fluorescent protein (EGFP), and cytotoxicity in both cells. The results show that transfection efficiency of PAMAM-H-R improved compared to native PAMAM dendrimer, but cytotoxicity of PAMAM-H-R and PAMAM-H-K were very low. We treated both cells with a polyplex formation of PAMAM-H-R or PAMAM-H-K/apoptin, and analyzed their cellular uptake and localization by flow cytometry and confocal microscopy. Furthermore, we analyzed the endosomal escape effect using TEM images, and found that PAMAM-H-R showed very fast escape from endosome to the cytosol. Caspase 3 activity assay, cell cycle distribution, and JC-1 analysis showed apoptosis induced by apoptin in GBL-14 cells. This indicates that PAMAM-H-R can be a potential nonviral vector gene delivery carrier for brain tumor therapy. The present study demonstrates that PAMAM-H-R/apoptin gene polyplex can be used as an effective therapeutic candidate for GBM due to its selective induction of apoptosis in primary glioma cells as a potential nonviral gene delivery carrier for brain tumor therapy.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Novel Multiwavelength Erbium-Doped Fiber and Raman Fiber Ring Lasers With Continuous Wavelength Spacing Tunability at Room Temperature

        <P> In this paper, we investigate a flexibly tunable multiwavelength fiber ring laser with a continuous wavelength spacing tunability, incorporating a superimposed chirped fiber Bragg grating (CFBG) at room temperature. We exploit versatile gain media such as erbium-doped fiber (EDF) and Raman amplifiers in generating the multiwavelength fiber ring laser. The wavelength spacing of a superimposed CFBG can be continuously controlled by modifying the chirp ratio of the grating with the specially designed apparatus. After configuring a fiber ring cavity with the proposed multichannel filter, we realize stable multiwavelength fiber ring lasers at room temperature, which are based on an EDF amplifier incorporating a highly nonlinear dispersion-shifted fiber or a Raman amplifier using a dispersion-compensated fiber. Power fluctuation is less than <TEX>$\sim$</TEX> 0.6 dB. No lasing wavelength change is severally observed for the whole tuning range. For the multiwavelength EDF laser, stable 11-wavelength simultaneous operation spaced at 0.51 nm, with a signal-to-noise ratio (SNR) of more than <TEX>$\sim$</TEX>45 dB at room temperature, is achieved. A high-quality multiwavelength Raman laser output with stable 30 lasing wavelengths spaced at 0.51 nm with a high SNR of more than <TEX>$\sim$</TEX>45 dB is also realized. By applying the proposed tuning technique to both the multiwavelength EDF and Raman lasers, a wide and continuous tunability of lasing wavelength spacing is successfully achieved, which is measured to be <TEX>$\sim \pm$</TEX>0.033 nm/mm. By controlling the pump power in the multiwavelength Raman fiber ring laser, we can control the number of lasing channels at a wavelength spacing of 0.51 nm. </P>
