        Geometric optimization and performance analysis of radial MR valve using Taguchi orthogonal experiment method

        Aiming at the problems of increasing external dimensions and deteriorating key performance indicators in the design process of magnetorheological (MR) valve by using structural optimization method, a geometric optimization design methodology for the optimal design of a MR valve structure under specific volume constraints is proposed in this article. The optimization methodology couples the finite element model (FEM) constructed in COMSOL software with the Taguchi orthogonal experiment and response surface technology to build an approximate response surface function for the identified independent variables. Suitable optimization algorithms are then utilized to determine the optimal geometry of the MR valve, thereby maximizing the valve performance. Firstly, a radial MR valve with a single excitation coil was presented, and its structure and working principle were also elaborated. A mathematical model of the pressure drop was derived on the basis of the Bingham-Papanastasiou non-Newtonian constitutive model of MR fluid and the magnetic circuit had been analyzed with the FE analysis methodology. Then, a second-order response surface model (RSM) had been fitted for the magnetic flux density in the radial flow channel and spool region of the radial MR valve based on the Taguchi orthogonal experimental design. The fitted model was a function of the four independent variables of the radial MR valve, and the accuracy of the developed response surface function over the entire design space had also been estimated. Meanwhile, predictions made by the RSM and FE models were evaluated by analysis of variance and it was exhibited that the RSM model’s results agree with FE result fairly. Subsequently, the geometric optimization problem had been formulated for the constructed RSM exploiting the genetic algorithm to find the global optimum geometrical parameters of the radial MR valve. Furthermore, the experimental test rig was setup to explore the pressure drop and the response time characteristics of the initial and optimal radial MR valve as well as the dynamic performance of the MR valve controlled cylinder system under different excitation conditions. The experimental results show that under the applied current of 2 A, the pressure drop and adjustable coefficient of the optimal radial MR valve observably increased with values of 3.15 MPa and 5.40, respectively, when compared to 2.11 MPa and 4.22 of their respective initial values. Also, at the applied current of 1.25 A, the damping force of the MR valve controlled cylinder system enlarged by 46 %, with its optimal value being 3.65 kN and initial value as 2.50 kN, which was an excellent verification of the correctness of the RSM and the effectiveness of the optimal design.

      • Endoplasmic Reticulum Mediated Necrosis-like Apoptosis of HeLa Cells Induced by Ca<sup>2+</sup> Oscillation

        Apoptosis and necrosis are distinguished by modality primarily. Here we show an apoptosis occurred instantly, induced by $300\;{\mu}M$ W-7 ((N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide hydrochloride), inhibitor of calmodulin), which demonstrated necrotic modality. As early as 30 min after W-7 addition, apoptotic (sub-diploid) peak could be detected by fluorescence-activated cell sorter (FACS), “DNA ladders” began to emerge also at this time point, activity of caspase-3 elevated obviously within this period. Absence of mitochondrial membrane potential (MMP) reduction and cytochrome c, AIF (apoptosis inducing factor) release, verified that this rapid apoptosis did not proceed through mitochondria pathway. Activation of caspase-12 and changes of other endoplasmic reticulum (ER) located proteins ascertained that ER pathway mediated this necrosis-like apoptosis. Our findings suggest that it is not credible to judge apoptosis by modality. Elucidation of ER pathway is helpful to comprehend the pathology of diseases associated with ER stress, and may offer a new approach to the therapy of cancer and neurodegenerative diseases.

        Modeling of nanoparticles’ aggregation and sedimentation in nanofluid

        The aggregation and sedimentation of nanoparticles in nanofluid have significant influences on the stability and applicability of nanofluids. The objective of this study is to propose a model to predict the nanoparticles’aggregation and sedimentation characteristics. The characteristics are evaluated by the concentration of nanoparticles in nanofluid at different time. The concentration of nanoparticles can be calculated according to the speed and location of each nanoparticle. Then, the speed and location of each nanoparticle can be yielded when the forces on each nanoparticle are determined. For the forces on nanoparticles are related to the space structure of nanoparticle clusters, the clusters’ space structures are simulated. Case study shows that the mean deviation of predicted nanoparticle concentration from experimental data for Fullerence + H2O, Fullerence + Oil and CuO + Oil nanofluids are 25%, 16% and 13%, respectively. The model can provide quantitative prediction of the aggregation and sedimentation characteristics of nanoparticles in nanofluid.

        PRESSURE DROP CHARACTERISTICS OF R410A-OIL MIXTURE FLOW BOILING IN SMALL SMOOTH TUBES

        This study presents experimental frictional pressure drop for R410A/oil mixture flow boiling in small horizontal smooth tubes with inside diameters of 4.18 mm and 2.0 mm. Experimental conditions cover nominal oil concentrations from 0 to 5%. The test results show that the presence of oil enhances two-phase frictional pressure drop about 0–120% and 0–90% at present test conditions for 4.18 mm I.D. smooth tube and 2.0 mm I.D. smooth tube, respectively, and the enhanced effect is more evident at higher vapor qualities where the local oil concentrations are higher. A new correlation to predict the local frictional pressure drop of R410A/oil mixture flow boiling inside conventional size and small smooth tubes is developed based on local properties of refrigerant–oil mixture, and the experimental data of 4.18 mm I.D. and 2.0 mm I.D. smooth tubes and that of 6.34 mm I.D. smooth tube (Hu et al., 2008) are well-correlated with the new correlation.

        FOXK1 regulates epithelial-mesenchymal transition and radiation sensitivity in nasopharyngeal carcinoma via the JAK/STAT3 signaling pathway

        Background Nasopharyngeal carcinoma (NPC) is the most common head and neck tumor in China. Forkhead box (FOX) proteins have 19 subfamilies, which can maintain cell metabolism, regulate cell cycle and cell growth, etc. FOXK1 is a member of the FOX family, and studies have found that FOXK1 is closely related to tumors. Objective This experiment aims to study the effects of FOXK1 interference on proliferation, apoptosis, invasion, epithelial-mesenchymal transition (EMT), and radiosensitivity, by regulating the Janus kinas/signal translator and activator of the transfer 3 (JAK/STAT3) pathway. Methods The expression of FOXK1 was detected via immunohistochemistry using clinical nasopharyngeal carcinoma tissues and adjacent tissues. The relationship between FOXK1 expression and tumor stage was subsequently evaluated. The colony formation rate was calculated through the colony formation experiment. Cell apoptosis and cell cycle distribution were detected using flow cytometry, while cell invasion was detected using the Transwell method. The number of cells in the nucleus of each group after 30 min, 4 h, and 24 h of radiotherapy with the 2 Gy dose was counted using immunofluorescence under γ-H2AX focal points of a laser confocal microscope. Results FOXK1 is clearly expressed in the patients’ cancer tissues. The expression of FOXK1 was significantly correlated with the patient’s sex. FOXK1 interference or Peficitinib can upregulate the apoptosis rate of 5-8 F and CNE-2 cells; increase the G2 phase of cells; and inhibit the invasion, migration, and EMT of cells. At the same time, FOXK1 interference can downregulate the protein expression of p-JAK1, p-JAK2, and p-STAT3 in cells. Interference from FOXK1 or Peficitinib alone can reduce the rate of cell colony formation under different radiation doses, and enhance the green fluorescence intensity of γ-H2AX in the nucleus after 4 and 24 h of the 2 Gy dose of radiotherapy. These results are optimal when FOXK1 interference and Peficitinib are used together. Conclusion FOXK1 interference in NPC cells can regulate EMT through the JAK/STAT3 signal pathway, enhance the radiosensitivity of cells, and thus inhibit tumor cell progression.

        Alterations in antioxidant function and cell apoptosis in duck spleen exposed to molybdenum and/or cadmium

        To investigate the effects of molybdenum (Mo) and/or cadmium (Cd) on antioxidant function and the apoptosis-related genes in duck spleens. Sixty healthy 11-day-old ducks were randomly divided into six groups of 10 ducks (control, low Mo group, high Mo, Cd, low Mo + Cd, and high Mo + Cd groups). All were fed a basal diet containing low or high dietary doses of Mo and/or Cd. Relative spleen weight, antioxidant indices, apoptosis-related gene mRNA expression levels, and ultrastructural changes were evaluated after 120 days. The results showed that the relative spleen weight decreased significantly in the high Mo + Cd treatment group which compared with control group. Malondialdehyde levels increased and xanthine oxidase and catalase activities decreased in the Mo and/or Cd groups compared with levels in the control group. Bak-1 and Caspase-3 expressions were upregulated in the high Mo + Cd group, while Bcl-2 was downregulated. In addition, mitochondrial crest fracture, swelling, vacuolation, deformed nuclei, and karyopyknosis in both Mo + Cd treated groups were more severe than in the other groups. The results suggest that Mo and/or Cd can induce oxidative stress and apoptosis of spleen via effects on the mitochondrial intrinsic pathway. Moreover, the results indicate the two elements have a possible synergistic relationship.

        Comparative transcriptome analysis of salt-sensitive and salt-tolerant maize reveals potential mechanisms to enhance salt resistance

        Background Salt stress is a devastating environmental stress that causes plant growth inhibition and yield reduction. Objective The identification of salt-tolerant genes brings hope for the generation of salinity-tolerant crop plants through molecular breeding. Methods In this study, one salt-sensitive and one salt-tolerant maize inbred line were screened from 242 maize inbred lines. Reactive oxygen species (ROS)-related enzyme activities were detected and salt-responsive comparative transcriptome analysis was performed for control and 220 mM NaCl treated maize leaves. Results Salt-tolerant maize inbred line (L87) showed higher ROS-related enzyme (SOD, POD, APX and CAT) activities and accumulated relatively lower levels of ROS under salt stress. Of the total DEGs, 1856 upregulated DEGs were specific to L87, including stress tolerance-related members of the 70kDa family of heat shock proteins (Hsp70s) and aquaporins. The DEGs involved in the abscisic acid (ABA), ethylene, jasmonic acid (JA) and salicylic acid (SA) signal transduction pathways may determine the difference in salt tolerance between the two varieties, especially one central component SnRK2, that positively regulates ABA signaling and was only upregulated in L87. Analysis of DEGs related to ROS scavenging showed that some peroxidase (POD), glutathione S-transferase (GST), catalase (CAT) and superoxide dismutase (SOD) genes specific to L87 probably enhanced its salt tolerance. The analysis of differentially expressed transcription factors (TFs) suggested that WRKY TFs could contribute to the difference in salt tolerance between the two maize lines. Conclusion Compared with Salt-sensitive maize inbred line (L29), L87 exhibits specific regulatory mechanisms related to salt tolerance, including plant hormone interactions, ROS scavenging and the regulation of TFs. Our study identifies new candidate genes that may regulate maize tolerance to salt stress and provides useful information for breeding maize with high salt resistance.

        Elevated level of renal xanthine oxidase mRNA transcription after nephropathogenic infectious bronchitis virus infection in growing layers

        To assess relationships between xanthine oxidase (XOD) and nephropathogenic infectious bronchitis virus (NIBV) infection, 240 growing layers (35 days old) were randomly divided into two groups (infected and control) of 120 chickens each. Each chicken in the control and infected group was intranasally inoculated with 0.2 mL sterile physiological saline and virus, respectively, after which serum antioxidant parameters and renal XOD mRNA expression in growing layers were evaluated at 8, 15 and 22 days post-inoculation (dpi). The results showed that serum glutathione peroxidase and superoxide dismutase activities in the infected group were significantly lower than in the control group at 8 and 15 dpi (p < 0.01), while serum malondialdehyde concentrations were significantly higher (p < 0.01). The serum uric acid was significantly higher than that of the control group at 15 dpi (p < 0.01). In addition, the kidney mRNA transcript level and serum activity of XOD in the infected group was significantly higher than that of the control group at 8, 15 and 22 dpi (p < 0.05). The results indicated that NIBV infection could cause the increases of renal XOD gene transcription and serum XOD activity, leading to hyperuricemia and reduction of antioxidants in the body.
