        Flavonoid Glycosides and Potential Antivirus Activity of Isolated Compounds from the Leaves of Eucalyptus citriodora

        The extraction and solvent partition of the leaves of Eucalyptus citriodora, and repeated column chromatography for n-BuOH fraction yielded a new flavonoid glycoside, citrioside C (1), along with three known flavonoid glycosides (2-4). The latter were identified with kaempferol-3-O-${\beta}$-$\small{D}$-glucopyranosyl (12)-${\alpha}$-$\small{L}$-rhamnoside (2), kaempferol-3-O-${\alpha}$-$\small{L}$-rhamnoside (3), and quercetin-3-O-${\alpha}$-$\small{L}$-rhamnoside (4). Their chemical structures were identified on the basis of spectroscopic data analyses including NMR, MS, UV, and IR. All constitutents were isolated for the first time from the leaves of Eucalyptus citriodora. The potential antivirus activity of all the isolated compounds was evaluated. Compound 4 showed potent antiviral activity against respiratory syncytial virus with 50% inhibition concentration ($IC_{50}$) value of $1.9{\mu}g/mL$ and selective index value of 9.8.

      • Effects of Doctor-patient Communication on Quality of Life among Breast Cancer Patients in Southern China

        Objective: This study aimed to determine effects of doctor-patient communication on the quality of life among breast cancer survivors in 16 communities in southern China. Methods: Multistage random sampling was to use to recruit 260 females from the Guangzhou Cancer Registry Database who were diagnosed with breast cancer. A questionnaire provided data on the doctor-patient communication (including the doctor's attitude, the patient's participation with the medical decision and information about the disease) and QOL (quality of life), as measured using FACT-B. Univariate analysis, non-conditional logistic regression was used to evaluate the associations between the doctor-patient communication and QOL. Results: Females who received good attitudes from doctors demonstrated higher FACT-B (OR=4.65, 95% CI: 1.68-12.86), social well-being (OR=5.88, 95% CI: 2.16-16.05), emotional well-being (OR=4.77, 95% CI: 1.92-11.88), and functional well-being ((OR=5.26, 95% CI: 1.90-14.52) compared to the females who encountered worse attitudes from their doctor, adjusting for age, education, marriage, employment, family income, years since diagnosis, TNM stage, radiation therapy, chemotherapy and side effects, particularly when the TNM stage was 0-II and the patients exhibited no side effects. Regardless of the length of time after diagnosis, doctors' good attitudes resulted in higher QOL scores. Conclusions: This study demonstrated that the doctor-patient communication has a significant association with the QOL of breast cancer survivors, mainly dependent on the doctors' attitude. Effective intervention is required to develop optimal doctor-patient communication.

        A new phenylethanoid glycoside with antioxidant and anti-HBV activity from Tarphochlamys affinis

        A new phenylethanoid glycoside, named taraffinisoside A (1), together with five known glycosides were isolated from the stems and leaves of Tarphochlamys affinis. The structure of taraffinisoside A was identified on the basis of detailed spectral analysis. Compounds 1-4 and 6 showed potent antioxidant activities with $IC_{50}$ values of 10.36, 19.73, 43.95, 15.30 and $46.04{\mu}M$ by 1,1-diphenyl-2-picryhydrazyl radical-scavenging assay. Compounds 1, 2 and 4 showed anti-HBV activities, with $IC_{50}$ values of 0.50, 0.72 and 0.26 mM for HBsAg and 0.93, 0.42 and 0.07 mM for HBeAg, respectively.

        The world economy has obviously changed over last five decades. Growing internationalization and involving globalization have become its distinctive features. In addition, the activities of MNEs play an important role in international business activities. The increasing influence of MNEs has proved a double-edged sword. From the theoretical perspective, the jobs of Hofstede’s Cultural Dimensions have showed the culture’s inflluence in the context of cross-cultural management function in organizations, while some approaches have concluded to realize the influence of cultural diversity on the ways of knowing, doing and learning. The paper has examined Hofstede’s Cultural Dimensions and investigated some problems related to MNEs’ cross-cultural management in Chinese cases. Based on some key discussions in academic fields, the paper would highlight more implications from Hofstede’s Cultural Dimensions to argue Chinese cases in current global background.

        Effects of Ring Pack Friction Heat on Temperature Fields of Piston Set-Liner

        To investigate the temperature fields of piston set-liner under the friction heat of ring pack, it is established a friction heat allocation model of ring pack based on the heat transfer theory of semi-infinite body. In this model, the piston ring, lubrication film and liner are treated as a three-layer medium. Furthermore, the distribution model of ring pack friction heat, the unsteady heat-mixed lubrication and friction model of piston ring-liner, and the heat transfer model of piston assembly-liner are also built and coupled with the allocation model to develop a coupled heat transfer model of piston set-liner. Employing the model for 6110 diesel engine, the effects of ring pack friction heat on the temperature fields of piston set-liner are simulated. Numerical results demonstrate that the friction heat allotted to the piston ring, lubrication film and liner accounts for nearly 47 %, 21 % and 32 % of ring pack friction heat, respectively. Due to the effects of friction heat, the maximum temperature rises on the piston ring, piston and liner surfaces run approximately up to 8.5 ℃, 5 ℃ and 5 ℃, separately. The established allocation and distribution models related to friction heat in this paper can be applied to any type of piston set-liner and provide the theoretical foundation for the optimal design of piston ring-liner.

      • Apoptotic Effects of psiRNA-STAT3 on 4T1 Breast Cancer Cells in Vitro

        Background: The aim of this study was to investigate the effect of a Lipofectamine2000 (Life2000) Transfection Reagent transfected psiRNA-STAT3 plasmid on 4T1 breast cancer cells. Materials and Methods: MTT was used to detect the cell proliferation of breast cancer 4T1 cells at different periods (0h, 6h, 8h, 10h); the cell cycle was assessed by flow cytometry; variation of apoptosis and mitochondrial membrane potential was observed under a fluorescence microscope; immunohistochemical staining was used to determine the expression of caspase-3 and cyclin-D1 protein. Results: An obvious effect of inhibition to 4T1 cancer cells could be observed at 8h after the psiRNA-STAT3 was transfected. Typical alterations of apoptotic morphological features were visible in the psiRNA-STAT3 treatment group. Mitochondrial membrane potential decreased significantly, the number of cells was increased in G0/G1 phase, and the number of cells was decreased in S phase, and the data were statistically significant (p<0.05), compared with the Scramble and Mock groups. Expression of caspase-3 protein was increased significantly, while that of cyclin D1 was significantly decreased. Conclusions: Life2000 transfected psiRNA-STAT3 plasmid can inhibit 4T1 tumor cell proliferation and promote apoptosis of 4T1 tumor cells, which process depends on the regulation of expression of cyclin D1 and caspase-3 protein.

        Genome Sequencing Highlights the Plant Cell Wall Degrading Capacity of Edible Mushroom Stropharia rugosoannulata

        The basidiomycetous edible mushroom Stropharia rugosoannulata has excellent nutrition, medicine, bioremediation, and biocontrol properties. S. rugosoannulata has been widely and easily cultivated using agricultural by-products showing strong lignocellulose degradation capacity. However, the unavailable high-quality genome information has hindered the research on gene function and molecular breeding of S. rugosoannulata. This study provided a high-quality genome assembly and annotation from S. rugosoannulata monokaryotic strain QGU27 based on combined Illumina-Nanopore data. The genome size was about 47.97 Mb and consisted of 20 scaffolds, with an N50 of 3.73 Mb and a GC content of 47.9%. The repetitive sequences accounted for 17.41% of the genome, mostly long terminal repeats (LTRs). A total of 15,726 coding gene sequences were putatively identified with the BUSCO score of 98.7%. There are 142 genes encoding plant cell wall degrading enzymes (PCWDEs) in the genome, and 52, 39, 30, 11, 8, and 2 genes related to lignin, cellulose, hemicellulose, pectin, chitin, and cutin degradation, respectively. Comparative genomic analysis revealed that S. rugosoannulata is superior in utilizing aldehyde-containing lignins and is possible to utilize algae during the cultivation.

        TIP60 contributes to porcine embryonic development by regulating DNA damage response

        <P><B>Abstract</B></P> <P>The acetyltransferase TIP60 (also known as Kat5) is a member of the MYST family of histone acetyltransferases and was initially identified as a cellular protein. TIP60 acetylates histone and non-histone proteins and is involved in diverse biological processes, including apoptosis, cell cycle, and DNA damage responses. In this study, a specific inhibitor of TIP60 was used to detect the function of TIP60 in porcine parthenogenetic embryos. The results showed that TIP60 inhibition impaired porcine parthenogenetic embryonic development. The mechanism of TIP60 was also determined. We found that the TIP60 inhibition impaired embryonic development by ROS induced DNA damage, as demonstrated by the number of γH2A in the nuclei. TIP60 inhibition triggered DNA damage through the regulation of p53-p21 pathway and TIP60 played a role in DNA repair. TIP60 inhibition decreased the efficiency of DNA repair by regulating 53BP1-dependent repair after DNA damage. Inhibition of TIP60 also increased the adaptive response, autophagy, by modulating LC3. Therefore, TIP60 plays a role in early porcine parthenogenetic embryonic development by regulating DNA damage and repair.</P> <P><B>Highlights</B></P> <P> <UL> <LI> TIP60 inhibition disrupted the embryonic development. </LI> <LI> TIP60 inhibition triggered the DNA damage and incomplete DNA repair due to the excessive generation of ROS. </LI> <LI> TIP60 inhibition induced the apoptosis and autophagy. </LI> </UL> </P>
