        Arc Envelope Grinding of Sapphire Steep Aspheric Surface with SiC-Reinforced Resin-Bonded Diamond Wheel

        In order to improve the grinding wheel wear during the sapphire steep aspheric surface grinding process, a SiC-reinforced resin-bonded hemispherical diamond wheel was used and the arc envelope grinding performance was investigated. Firstly, the mapping relationship between the contours of the grinding wheel and the aspheric surface was established based on the grinding conditions. The wear of the hemispherical diamond wheel was modelled, and the result indicates that the maximum wear occurred at the edge of the hemisphere, decreases along the generatrix and increases near the center. Then, the formtrued diamond wheel was used for grinding the sapphire steep aspheric surface. The concave and convex surface form error obtained at the central part of Φ 50 mm are 2.5 μm and 1.3 μm, respectively. The surface roughness R a is 230–450 nm, which is affected by the material removal rate and the sapphire crystal anisotropy. The SiC-reinforced resin-bonded diamond wheel possesses favorable self-sharpening ability and sufficient diamond grain retention capacity for sapphire grinding. The wear distribution shows that the most severe wear parts of the grinding wheel are at the edge and the center of the grinding zone, which is consistent with the model-predicted results.

        Mechanical Properties and Damage Characteristics of Coal-Rock Combination with Different Dip Angles

        The effect of the dip angle of an interlayer coal seam on rock mass mechanical behavior and damage characteristics is seldom discussed. To address the lack of the existing research, a numerical simulation was conducted using the Particle Flow Code (PFC2D), and its damage to mechanical behavior, acoustic emission (AE) characteristics, and damage characteristics were analyzed. The outcomes exhibit the peak strength and elastic modulus of coal-rock combinations are between that of rock and coal. As the coal inclination angle rises, the elastic modulus and peak strength gradually decrease. AE model experiences the initial growth period, stable growth period, and rapid decline period. The value of AE hits and the generation of the AE signal is affected by the size of the inclination angle. The degree of damage to the coal-rock combination gradually rises as the angle rises. The damage process of the coal and rock composite sample encompasses an initial damage phase, damage stable evolution phase, accelerated damage phase, and a failure phase. The damage values of the stable evolution stage for varying dip angle models are also different. As the inclination angle increases, the damage value gradually increases, while the strain at the same damage value gradually decreases.

      • Isolation and Characterization of Mouse Testis Specific Serine/Threonine Kinase 5 Possessing Four Alternatively Spliced Variants

        Phosphorylation on serine/threonine or tyrosine residues of target proteins is an essential and significant regulatory mechanism in signal transduction during many cellular and life processes, including spermatogenesis, oogenesis and fertilization. In the present work, we reported the isolation and characterization of mouse testis-specific serine/threonine kinase 5 (Tssk5), which contains four alternatively spliced variants including, Tssk5$\alpha$, Tssk5$\beta$, Tssk5$\gamma$ and Tssk5$\delta$. Moreover, the locus of Tssk5 is on chromosome 14qC3 and the four variants had a similar high expression in the testis and the heart; however, had a low expression in other tissues, except for Tssk5$\alpha$ which also had comparably high expression in the spleen. Each variant of Tssk5 expression began in the testis 16 days after birth. Aside from TSSK5$\alpha$, the other isoforms have an insertion of ten amino acid residues (RLTPSLSAAG) in region VIb (HRD domain) (His-Arg-Asp). Moreover, only TSSK5$\alpha$ exhibited kinase activity and consistently, a further Luciferase Reporter Assay demonstrated that TSSK5$\beta$, TSSK5$\gamma$ and TSSK5$\delta$ cannot be stimulated at the CREB/CRE responsive pathway in comparison to TSSK5$\alpha$. These findings suggest that TSSK5$\beta$, TSSK5$\gamma$, TSSK5$\delta$ may be pseudokinases due to the insertion, which may damage the structure responsible for active kinase activity. Pull-down assay experiments indicated that TSSK5$\beta$, TSSK5 $\gamma$ and TSSK5$\delta$ can directly interact with TSSK5$\alpha$. In summary, these four isoforms with similar expression patterns may be involved in spermatogenesis through a coordinative way in testis.

        Alpha-Hemolysin from Staphylococcus aureus Obstructs Yeast-Hyphae Switching and Diminishes Pathogenicity in Candida albicans

        The use of antibiotics can disrupt the body’s natural balance and increase the susteptibility of patients towards fungal infections. Candida albicans is a dimorphic opportunistic fungal pathogen with niches similar to those of bacteria. Our aim was to study the interaction between this pathogen and bacteria to facilitate the control of C. albicans infection. Alpha-hemolysin (Hla), a protein secreted from Staphylococcus aureus, causes cell wall damage and impedes the yeast–hyphae transition in C. albicans. Mechanistically, Hla stimulation triggered the formation of reactive oxygen species that damaged the cell wall and mitochondria of C. albicans. The cell cycle was arrested in the G0/G1 phase, CDC42 was downregulated, and Ywp1 was upregulated, disrupting yeast hyphae switching. Subsequently, hyphae development was inhibited. In mouse models, C. albicans pretreated with Hla reduced the C. albicans burden in skin and vaginal mucosal infections, suggesting that S. aureus Hla can inhibit hyphal development and reduce the pathogenicity of candidiasis in vivo.
