Bacillus subtilis의 세포내 아눌라아제가 부분정제되고 그의 작용 모드와 일반적 특성이 조사되었다. 이 효소는 gel filtration에 의하여 분자량을 추정하였을 때 49,000이었고, 등전점은 5.2이었다. 기질에 대한 친화성의 지표인 K_m값은 설탕에 대해서는 10mM, 라피노오스에 대해서는 18mM이었다. 이 효소는 산성쪽에서는 질로 사용, inulinase 활성측정 경우와 동일한 방법에 따라 측정하였다. 단, invertase 활성은 1분간에 2umole의 환원당을 생산하는 효소량을 1단위로 표시하였다. An intracellular inulase(β-fructofuranoside fructohydrolase, EC 3.2.1.26) from Bacillus subtilis has been partially purified and its mode of action and general properties were studied. The enzyme had an apparent molecular weight of 49,000 as estimated by gel filtration and its pI point was 5.2. Substrate concentration studies showed an apparent K_m of 10 mM for sucrose and of 18 mM for raffinose. The enzyme was an acid-labile protein with a pH optimum of 6.6. The optimum temperature was 50℃. The enzyme acts on straight chain oligo-and poly-fructosides of the inulin series via a exo-wise cleavage mechanism, as well as on sucrose.
Studies on Fungal Lipids Containing γ - Linolenic Acid - 1. Fatty Acid Composition of Mucor sp.

감마-리놀렌산 생산능이 있는 새로운 곰팡이 균주를 탐색하기 위하여 토양으로부터 감마-리놀렌산 생산능이 있는 3개의 곰팡이 균주를 분리하였으며, 이들 균주를 진탕 배양을 행하여 균체성장, 지질생성량 및 지방산조성을 비교 조사하였다. 3가지 균주중에서 FA-007 균주성장도 양호하였고, 지질함량이 건조균체중 21.1%, 감마-리놀렌산 함량이 총지방산중 15.6%로 높은 특성을 나타내었다. 이 균주는 형태학적 특성을 조사한 결과 Mucor sp.로 동정되었다. 이 균주가 생산하는 지질은 중성지질이 75.2%, 당지질이 5.3% 및 인지질이 19.5%로 구성되어 있었고, 중성지질과 인지질에서의 감마-리놀렌산 함량은 각각 15.5% 및 18.3%를 차지하였다. For a study on the production of y-linolenic acid(GLA) by fungi, 3 fungal strains were isolated from soil. Their cell growth, lipid content and fatty acid composition were compared in shake flask culture. Among these fungi, the fungus, designated as FA-007, has high lipid content(21.1%) and GLA content(15.6% of total fatty acids). The fungal strain FA-007 was tentatively identified as Mucor sp. on the basis of morphological characteristics. Fungal oil produced by this fungus was composed of 75.2% neutral lipid, 5.3% glycolipid and 19.5% phospholipid. Although the GLA content in phospholipid was higher than it in neutral lipid, the GLA content in neutral lipid was high as 15.5%.
미나리(Oenanthe stolonifera DC.)의 향기성분

미나리(Oenanthe stolonifera DC)의 향기성분을 상압 수증기 증류법으로 추출하여 GC 및 GCMS로 분석한 결과 63가지 성분이 분리되었고, 그중에서 탄화수소류 20종(61.94%), alcohol류 9종(8.76%), ester류 1종(1.34%), aldehyde류 1종(2.29%), ketone류 3종(11.52%) 및 miscellaneous 5종 (2.35%) 등 39가지 성분을 동정하였다. 미나리 향기성분은 주로 탄화수소류(61.94%)로 구성되어 있었으며, 주성분은 limonene(12.12%), pulegone(9.48%), germacrene D(8.34%) 및 β-pinene(7.68%)등이었다. The volatile fraction of Oenanthe stolonifera DC. was extracted by a steam distillation under the atmospheric pressure and analyzed by gas chromatography(GC) and gas chromatography/mass spectrometry(GC-MS). The experimental results revealed the presence of 63 volatile components. Among them 39 components identified were composed of 20 hydrocarbons (61.94%), 9 alcohols(8.76%), 3 ketones(11.52%), 1 ester(1.34%), 1 aldehyde(2.29%) and 5 miscellaneous(2.35%). The major volatile components of Oenanthe stolonifera DC. were limonene(12.12%), pulegone(9.48%), germacrene D(8.34%) and α-pinene(7.68%).

Lipids of pumpkin seed were extracted by the mixture of chloroform - methanol (2:1, V/V) and fractionated into neutra lipids, glycolipids and Phospholipids by silicic acid column chromatogaphy. Components and fatty acid composition of individual lipid class were determined by thin layer and gas-liquid chromate-graphies. The results are summarized as follows: 1. Lipid content of pumpkin seed was 40.1275 and the lipid consisted of neutral lipids(93.96%), glycolipids(1.92%) and phospholipids(4.12%). 2. The major components of the neutral lipids were triglyceride and free fatty acid, and their contents were 79.50% and 7.74%, respectively. 3. The major fatty acids of the neutral lipids were linoleic acid(53.19%), oleic acid(19.41%) and palmitic acid(18.08%). 4. The major components of the glycolipids were cerebroside and steryl glycoside, and their contents were 52.39%. 5. The major fatty acids of the glycolipids were palmitic acid(48.20%) and linoleic acid(22.95%). 6. The major component of the phospholipids was phosphatidyl choline and it's content was 53.33%. 7. The major fatty acids of the phospholipids were linoleic acid(36.45%), palmitic acid(24.83%) and oleic acid(20.95%). 8. The major fatty acids of the triglyceride in the pumpkin seed were linoleic acid(51.67%), oleic acid(19.40%) and palmitic acid(19.18%). 9. The major fatty acids of the total lipid were linoleic acid(52.13%), oleic acid(19.45%) and palmitic acid(18.65%). An odd-number fatty acid, heptadecanoic acid was also identified and it's content was 0.14%. 10. In view of the fatty acid composition, the pumpkin seed oil was classified as the group of oleic-linoleic oil.
Aspergillus ficuum 조효소액으로부터 Exoinulinase의 정제 및 특성

Aspergillus ficuum이 생산하는 exoinulinase가 CM-Sephadex, DEAE-Sepharose 6B 및 HPLC gel filtration을 통해 단백질 ㎎당 약 2,800U의 specific activity로 정제되었다. 이 효소는 native 상태에서 약 83,000±1,000의 분자량을 나타냈으며 당을 함유하고 있었다. 이 효소는 최적 pH가 4.4∼4.7이었고 55℃에서 8시간 노출 후에도 그 활성을 95% 유지하였다. 이 효소의 I/S ratio는 약 0.35이고 전형적인 non-speific β-fructofuranosidase의 특성을 나타내었으며 raffinose와 stachyose를 분해할 수 있었다. An exoinulinase (EC 3.2.1.80) was purified from a commercial inulinase preparation from Aspergillus ficuum using ion exchange chromatography on CM-Sephadex C-50 and DEAE-Sepharose 6B and HPLC gel filtration on a Protein Pak 125 column. Native exoinulinase had a molecular weight of 83,000±1,000 and was glycoprotein. Optimal pHs of the enzyme were ranged from 4.4 to 4.7. About ninety five percent of the whole activity was maintained even after incubation of 8 hours at 55℃. The enzyme was a typical non-specific β-fructofuranosidase, of which I/S ratio appears to be 0.35.
