        Monte Carlo Simulation on Kinetics of Batch and Semi-Batch Free Radical Polymerization

        Based on Monte Carlo simulation technology, we proposed a hybrid routine which combines reaction mechanism together with coarse-grained molecular simulation to study the kinetics of free radical polymerization. By comparing with previous experimental and simulation studies, we showed the capability of our Monte Carlo scheme on representing polymerization kinetics in batch and semi-batch processes. Various kinetics information, such as instant monomer conversion, molecular weight, and polydispersity etc. are readily calculated from Monte Carlo simulation. The kinetic constants such as polymerization rate kp is determined in the simulation without of “steady-state” hypothesis. We explored the mechanism for the variation of polymerization kinetics those observed in previous studies, as well as polymerization-induced phase separation. Our Monte Carlo simulation scheme is versatile on studying polymerization kinetics in batch and semi-batch processes.

        A hemoglobin-based oxygen-carrying biomimetic nanosystem for enhanced chemo-phototherapy and hypoxia alleviation of hepatocellular carcinoma

        Tumor microenvironment is characterized by low pH, high reactive oxygen species and hypoxia, whichprovides a suitable environment for cancer growth. The hypoxia not only elevates tumor angiogenesisand metastasis, but also is responsible for the development of treatment resistance, which graduallybecomes a significant impediment for cancer therapy. Therefore, we developed a biomimetic nanosystemcontaining hemoglobin extracted from red blood cells, chemotherapy drug sorafenib, sensitizer ursolicacid and photosensitizer indocyanine green for enhanced chemo-photo combination therapy of hepatocellularcarcinoma, which could not only enhance the chemotherapy effect of sorafenib bowing to thesensitizing effect of ursolic acid, but also achieved synergetic phototherapy in virtue of indocyaninegreen. Besides, the nanoparticles could effectively delivery exogenous oxygen to tumor site and amelioratethe tumor hypoxic environment with the assistance of hemoglobin. The dual-sensitization drugdelivery system was expected to effectively reduce the resistance of traditional treatment methodsagainst tumor hypoxia, providing a novel prospect for the synergistic hepatocellular carcinomatreatment.

      • Knockdown of GCF2/LRRFIP1 by RNAi Causes Cell Growth Inhibition and Increased Apoptosis in Human Hepatoma HepG2 Cells

        Background: GC-binding factor 2 (GCF2) is a transcriptional regulator that represses transcriptional activity of the epidermal growth factor receptor (EGFR) by binding to a specific GC-rich sequence in the EGFR gene promoter. In addition to this function, GCF2 has also been identified as a tumor-associated antigen and regarded as a potentially valuable serum biomarker for early human hepatocellular carcinoma (HCC) diagnosis. GCF2 is high expressed in most HCC tissues and cell lines including HepG2. This study focused on the influence of GCF2 on cell proliferation and apoptosis in HepG2 cells. Materials and Methods: GCF2 expression at both mRNA and protein levels in HepG2 cells was detected with reverse transcription (RT) PCR and Western blotting, respectively. RNA interference (RNAi) technology was used to knock down GCF2 mRNA and protein expression. Afterwards, cell viability was analyzed with a Cell Counting Kit-8 (CCK-8), and cell apoptosis and caspase 3 activity by flow cytometry and with a Caspase 3 Activity Kit, respectively. Results: Specific down-regulation of GCF2 expression caused cell growth inhibition, and increased apoptosis and caspase 3 activity in HepG2 cells. Conclusions: These primary results suggest that GCF2 may influence cell proliferation and apoptosis in HepG2 cells, and also provides a molecular basis for further investigation into the possible mechanism at proliferation and apoptosis in HCC.

        Multi-Index Seismic Capacity Evaluation of Buckling-Restrained Braced Frames

        The seismic capacity of a structure is determined by the performance index that reaches its ultimate bearing or deformation capacity fi rst. This paper presents a multi-index seismic capacity evaluation method for accurately evaluating the seismic capacity of a structure. The normalized response curves of several indices are concurrently plotted to form a multi-index seismic capacity evaluation fi gure, in which the seismic capacity and demand values that correspond to various indices can be determined by vertical and horizontal threshold lines. Based on an incremental dynamic analysis (IDA), a 6-story buckling-restrained braced frame (BRBF) and a series of comparable 6-story steel frames are analyzed using the proposed method to verify the method and investigate the seismic behavior of BRBFs. The results show that the seismic performance of buckling-restrained braces is not the only factor that determines the seismic capacity of BRBFs and indicate that the multiindex seismic capacity evaluation method can eff ectively identify the critical index of a structure and the weakest links that restrict the structural seismic capacity.

        A Modified PiggyBac Transposon System Mediated by Exogenous mRNA to Perform Gene Delivery in Bovine Mammary Epithelial Cells

        Transposons are widely used for geneticengineering in various model organisms. Recently, piggyBac(PB) has been developed as a transposable and efficientgene transfer tool in mammalian cells. In the present study,we developed three types of PB transposon systemscontaining a dual plasmid system (DPS), a single plasmidsystem (SPS), and a DNA-mRNA combined system (DRPS)and characterized their basic properties in HEK293 cells. The basic elements of the donor plasmid included aselectable-reporter gene expression cassette, two loxP sitesin the same orientation, a multiple cloning site, and twochicken β-globin insulator core elements. We further identifiedthe function of the selectable-reporter and examined PBintegration sites in the human genome. Moreover, wecompared the transposition efficacy and found that SPStransposed more efficiently, as compared to DPS; integrationinto the host genome was determined by measuring PBaseactivity. Results discovered the loss of PBase activity in theDRPS, indicating that this system is much more biologicallysafe, as compared to DPS and SPS. Finally, we employedthe DRPS to successfully perform a gene delivery intobovine mammary epithelial cells (BMECs). Taken together,the information from this study will improve the flexibilityof PB transposon systems and reduce the genotoxicity ofPBase in genetic engineering.

        Deployment Dynamics of Large-Scale Flexible Solar Arrays with Deployable Mast

        In this paper, deployment dynamics of large-scale flexible solar arrays with deployable mast is investigated. The adopted solar array system is introduced firstly, then kinematic description and kinematic constraint equations are deduced, and finally, dynamics equation of the system is established by the Jourdain velocity variation principle and a new method to deal with topology changes of the deployable mast is introduced. The dynamic behavior of the system is studied in detail. Simulation results indicate that the proposed model is effective to describe the deployment dynamics of the solar arrays and that the introduced method is applicable for topology changes.

        A Novel Form Analysis Method Considering Pretension Process for Suspen-dome Structures

        Suspen-dome is a kind of new prestressed space grid structure which is formed by combing a single-layer reticulated shell andtensegrity system. For the existence of lower flexible cable-strut system, form analysis is crucial for the mechanical performance ofsuspen-domes. This paper develops a novel form analysis method considering the influence of pretension process for suspen-domestructures. Some definitions of form analysis for suspen-dome structures are first expounded. Then an iterative method is presentedfor form analysis, and a sequential analysis method is proposed for pretension process simulation. By combining these two methods,a form analysis method considering pretension process is constructed for suspen-dome structures. Two examples are employed toverify the proposed method. Numerical example results show that both the error of nodal coordinates and cable forces can stablyconverge to set tolerances. According to a scheduled pretension scheme, form analysis results can accurately achieve the expectedinitial state. Engineering example results show that without considering the influence of pretension process, the form analysis wouldbe error and the final state after all cables tensioned deviates greatly from the expected initial state. However, this can be achievedaccurately through the proposed method in this paper.

      • Refining and Validating a Two-stage and Web-based Cancer Risk Assessment Tool for Village Doctors in China

        The big gap between efficacy of population level prevention and expectations due to heterogeneity and complexity of cancer etiologic factors calls for selective yet personalized interventions based on effective risk assessment. This paper documents our research protocol aimed at refining and validating a two-stage and web-based cancer risk assessment tool, from a tentative one in use by an ongoing project, capable of identifying individuals at elevated risk for one or more types of the 80% leading cancers in rural China with adequate sensitivity and specificity and featuring low cost, easy application and cultural and technical sensitivity for farmers and village doctors. The protocol adopted a modified population-based case control design using 72, 000 non-patients as controls, 2, 200 cancer patients as cases, and another 600 patients as cases for external validation. Factors taken into account comprised 8 domains including diet and nutrition, risk behaviors, family history, precancerous diseases, related medical procedures, exposure to environment hazards, mood and feelings, physical activities and anthropologic and biologic factors. Modeling stresses explored various methodologies like empirical analysis, logistic regression, neuro-network analysis, decision theory and both internal and external validation using concordance statistics, predictive values, etc..

      • Dynamics and Liver Disease Specific Aspects of Quality of Life Among Patients with Chronic Liver Disease in Yunnan, China

        Background: Patients with chronic liver diseases (CLD) may have compromised health related quality of life (HRQoL). Hepatitis B virus (HBV) infection has long been the leading cause of CLD including liver cancer and cirrhosis. Knowledge on different symptom profiles of CLD should help in development of comprehensive treatment and patient care plans. Objective: To access the facets of HRQoL in chronic liver diseases throughout their spectrum of severity. Materials and Methods: A cross-sectional study was conducted in the First Affiliated Hospital of Kunming Medical University in Yunnan Province of China. Both out- and inpatients undergoing treatment protocols for different HBV related liver disease states were consecutively collected from December 2012 to June 2013. ANOVA was used to compare the mean scores of EQ-5D and chronic liver disease questionnaire (CLDQ) among 5 disease groups. The relationship between demographic variables predicting global CLDQ scores and the domains of CLDQ was analysed. Results: A total of 1040 patients including 520 without complications, 91 with compensated cirrhosis, 198 with decompensated cirrhosis, 131 with HCC and 100 with liver failure were recruited. All domains of CLDQ, the means of EQ-5D value and EQ VAS exhibited significant decline with worsening of disease severity from uncomplicated HBV to liver failure. The multivariate regression demonstrated the reduction of mean scores of CLDQ domain at advanced stage. Patients with liver failure and HCC had more HRQoL impairment than other disease states. No effect of patient gender was found. Patient age was associated with 'fatigue' and 'worry' domains (p=0.006; p=0.004) but not with other domains and global scores of CLDQ and ED-5D. Conclusions: The HRQoL in chronic hepatitis B patients is greatly affected by disease states. Care for HBV-related diseases should consider not only the outcomes of treatment strategies but also improvement in patient wellbeing.
