        The Molecular Mechanism of Long Non-Coding RNA MALAT1-Mediated Regulation of Chondrocyte Pyroptosis in Ankylosing Spondylitis

        Long non-coding RNAs (lncRNAs) may be important regulators in the progression of ankylosing spondylitis (AS). The competing endogenous RNA (ceRNA) activity of lncRNAs plays crucial roles in osteogenesis. We identified the mechanism of the differentially expressed lncRNA MALAT1 in AS using bioinformatic analysis and its ceRNA mechanism. The interaction of MALAT1, microRNA-558, and GSDMD was identified using integrated bioinformatics analysis and validated. Loss- and gain-of-function assays evaluated their effects on the viability, apoptosis, pyroptosis and inflammation of chondrocytes in AS. We found elevated MALAT1 and GSDMD but reduced miR-558 in AS cartilage tissues and chondrocytes. MALAT1 contributed to the suppression of cell viability and facilitated apoptosis and pyroptosis in AS chondrocytes. GSDMD was a potential target gene of miR-558. Depletion of MALAT1 expression elevated miR-558 by inhibiting GSDMD to enhance cell viability and inhibit inflammation, apoptosis and pyroptosis of chondrocytes in AS. In summary, our key findings demonstrated that knockdown of MALAT1 served as a potential suppressor of AS by upregulating miR-558 via the downregulation of GSDMD expression.

      • Empirical likelihood confidence regions for autoregressive models with explanatory variables

        The empirical likelihood method is very useful for establishing confidence region of the parameters of interest. In this paper, an empirical likelihood confidence region for the parameters of a univariate AR(p) model with a single explanatory variable which enters in the model through a nonlinear function is studied. Since the analytical expression of the nonlinear function is unknown, it is replaced by a nonparametric estimator, which is subsequently plugged in the estimating equations for the parameters of interest (the autoregressive parameters) to obtain the empirical likelihood confidence region. Our approach is to establish an empirical likelihood ratio statistic which is asymptotically chi-squared distributed. Some simulation results are also presented to illustrate the performance of the empirical likelihood method. An application to a real data set is provided.

        Marginal bone level change during sequential loading periods of partial edentulous rehabilitation using immediately loaded self-tapping implants: a 6.5-year retrospective study

        PURPOSE. A large number of studies have suggested the practicability and predictability of immediate implant function, but few studies have reported marginal bone level changes during sequential loading periods. The purpose of this study was to evaluate the marginal bone remodeling of immediately loaded self-tapping implants both at each time point and during each loading period between two time points. MATERIALS AND METHODS. The patients included in this retrospective study were treated with immediately loaded NobelSpeedy Replace implants between August 2008 and July 2009. Differences in the marginal bone level (MBL) at each time point and the marginal bone level change (ΔMBL) between two time points were analyzed with Bonferroni correction (P < .05). RESULTS. Overall, 24 patients (mean age, 47.3 ± 12.8 years) with 42 immediately loaded implants and a median follow-up of 6.5 years (IQR, 67.8 months) were included. The cumulative survival rate after 10 - 12 years was 95.2%. Continuous but slow marginal bone loss was observed during long-term follow-up. MBL at both 7.5 years and 11 years was significantly lower than that at loading, 6 months, 2 years and 4 years (P < .05). No bone loss difference was found in any period before 4 years of follow up (P > .05). The loading period of 4 years to 7.5 years showed the largest ΔMBL compared to those of other time periods (P < .05). CONCLUSION. Slight bone loss occurred continuously, and more radical changes of marginal bone can be observed during the period of 4-7.5 years. Thus, long-term effective follow-up of immediately loaded implants is needed.

      • Flexible INAR(1) models for equidispersed, underdispersed or overdispersed counts

        Equidispersed, underdispersed and overdispersed count data are commonly encountered in practice. To better describe these data characteristics, this paper develops two classes of INAR(1) processes, which not only can model a wide range of overdispersion and underdispersion, but also have ability to describe the zero-inflated and zero-deflated characteristics of the count data. The probabilistic and statistical properties of the two processes are studied. Estimators of the model parameters are derived by using conditional maximum likelihood (CML) and modified conditional least squares (MCLS) methods. Some asymptotic properties and numerical results of the estimators are investigated. Three real examples are given to show the flexibility and usefulness of the proposed models.

        Fault Diagnosis for Conventional Circuit Breaker Based on One-Dimensional Convolution Neural Network

        The vibration signal generated by the operating mechanism of conventional circuit breaker contains abundant mechanical state information. Aiming at traditional fault diagnosis methods that need to realize signal feature extraction based on feature selection, a fault diagnosis model based on one-dimensional convolutional neural network is proposed. In the diagnosis model, multiple convolutional neural networks are designed according to the type and degree of faults, and the network is set as a large convolutional kernel to enlarge the receptive field region; the raw vibration signal is used as the model input for training, and the corresponding fault type and degree are output after hierarchical diagnosis. The experimental results show that the model can automatically extract the fault signal features, effectively complete the fault diagnosis of the contact system for the conventional circuit breaker, and has good generalization ability. The model in this paper has a higher comprehensive diagnosis recognition rate compared with other methods, reaching 98.84%.

        Trastuzumab combined with irinotecan in patients with HER2-positive metastatic colorectal cancer: A phase II single-arm study and exploratory biomarker analysis

        Purpose The human epidermal growth factor receptor 2 (HER2) is an established therapeutic target for various kinds of solid tumors. HER2 amplification occurs in approximately 1% to 6% of colorectal cancer. In this study, we aimed to assess the efficacy and safety of trastuzumab in combination with chemotherapy in HER2-positive metastatic colorectal cancer (mCRC). Materials and Methods An open-label, phase II trial (Clinicaltrials.gov: NCT03185988) was designed to evaluate the antitumor activity of trastuzumab and chemotherapy in HER2-positive digestive cancers excluding gastric cancer in 2017. Patients from this trial with HER2-positive, KRAS/BRAF wild-type, unresectable mCRC were analyzed in this manuscript. Eligible patients were treated with trastuzumab (8 mg/kg loading dose and then 6 mg/kg every 3 weeks) and irinotecan (120 mg/m2 days 1 and 8 every 3 weeks). The primary endpoint was the objective response rate. Results Twenty-one HER2-positive mCRC patients were enrolled in this study. Seven patients (33.3%) achieved an objective res-ponse, and 11 patients (52.4%) had stable disease as their best response. The median progression-free survival (PFS) was 4.3 months (95% confidence interval, 2.7 to 5.9). Four of the 21 patients (19.0%) had grade 3 adverse events, including leukopenia, neutropenia, urinary tract infection, and diarrhea. No treatment-related death was reported. Exploratory analyses revealed that high tumor tissue HER2 copy number was associated with better therapeutic response and PFS. Alterations in the mitogen-activated protein kinase pathway, HER2 gene, phosphoinositide 3-kinase/AKT pathway, and cell cycle control genes were potential drivers of trastuzumab resistance in mCRC. Conclusion Trastuzumab combined with chemotherapy is a potentially effective and well-tolerated therapeutic regimen in mCRC with a high HER2 copy number.

        The differentiation of rat-induced pluripotent stem cells into alveolar type II epithelial cells with a three-step induction protocol

        Induced pluripotent stem (iPS) cells derive from autologous somatic cells, the application prospect of iPS cells forregenerative medicine and tissue engineering is better than embryonic stem cells (ESCs) to some extent. Alveolar type II(AT II) epithelial cells play key role in the injured lung tissue regeneration and function recovery. The differentiation of iPScells into AT II cells could provide available source for injured lung treatment. In this study, rat iPS (riPS) cells wereresuscitated and proliferated for 14 days before differentiation. A modified three-step induction protocol similar to thereported ESCs inducing procedure was used in this study for the differentiation groups. Routine cell culture was done to theriPS cell control group (riPS-con). At stage 3, cells of day 7 (Diff. 7) and day 14 (Diff. 14) were collected for the real-timepolymerase chain reaction tests for gene expressions of Oct4, Nanog, SPA, SPB, SPC, SPD, and CC10. Immunofluorescencestaining of SPC and SSEA-1 was conducted. At the end of the differentiation, cell morphology becameoutstretched and epithelium-like. Cells of the Diff. 14 group positively expressed SPC and negatively expressed SSEA-1,which is contrary to the riPS-con group. In the Diff. 7 and the Diff. 14 groups, the expression of Oct4, Nanog, and SPBdecreased (P < 0.05), whereas the expression of SPA, SPC, SPD (P < 0.05), and CC10 (P > 0.05) increased. This studyindicated that riPS cells can successfully differentiate into AT II epithelial cells with the three-step induction protocol andmay be further applied to implanting in decellularized rat lung scaffolds and building a bio-artificial lung.
