        Cross-currents of Change and Continuity:Korean National Literature and Korean National Cinema

        The decade of the 1960s in Korea is normally regarded as a transitional period, but this transition has been consistently viewed in either economic or political terms. In this paper I examine the 1960s and the 1970s as a period of cultural transition―from a neo-Confucian emphasis on “high” art to a broader, more inclusive acceptance of “popular” forms of cultural expression. Although these two realms of cultural production are often viewed as fundamentally antithetical, I argue that there is actually significant continuity in terms of both artistic expression and cultural engagement. In particular, I look at the trajectories of Korean national literature and cinema and their areas of confluence. I examine the career of Kim Sŭng-ok, who in the 1960s was the preeminent literary voice of a new generation and who in the 1970s was the screenwriter for some of the most popular films of that era, and I show how Kim’s career changes track parallel changes in both literature and cinema. Artistically, Kim continued his literary expressions of a new sensibility in his screenplays, bringing to cinema a new infusion of seriousness and respectability. Culturally, Kim continued his explorations of the ways in which rapid changes in Korean politics and the booming economy led to miscommunication and chaos in society. By consistently exploring processes of national identity formation and re-formation, Kim’s 1960s literature can be seen as crucial instances of Korean national literature (minjok munhak). In the same manner, Kim’s 1970s screenplays can be seen as foundational moments in a Korean national cinema.

        Literature of the Bittersweet: Kim Sung-ok and 1960s Korea

        This paper centers on the erstwhile novelist Kim Seung-ok with a focus not only on his watershed works of the 1960s, but also on his lesser-known works of the 1970s as well as on the circumstances and possible reasons for his decision to quit writing in the 1980s. His works from the 60s address certain basic human contradictions and are in this respect timeless, and these same works are also firmly grounded in their larger socio-cultural contexts of 1960s Korea. This article attempts to place the word firmly here sous rature. In new critical terms, the Kim's settings can not be understood as anything but Korea, in the then and now. This characteristic is shared, however, with highly ideological literature that at times seems to want to beat the reader over the head with the problems and author-sponsored solutions of then and now. In order to understand Kim's paradoxical position in Korean literary history, one must view his works from within the context of the debate between pure and engagement literature.

      • FACT complex is required for DNA demethylation at heterochromatin during reproduction in <i>Arabidopsis</i>

        <▼1><P><B>Significance</B></P><P>The chromatin remodeling activities of the FACT (facilitates chromatin transactions) complex are required for many cellular functions, including transcription, DNA replication, and repair. Here, we demonstrate that the two FACT subunits, SSRP1 and SPT16, are also required for genome-wide DNA demethylation and regulation of gene imprinting during <I>Arabidopsis</I> reproduction. Without FACT, <I>Arabidopsis</I> seeds undergo abnormal development and exhibit aberrant DNA hypermethylation, including at imprinting control region loci. We show that FACT associates with the DEMETER (DME) DNA demethylase, facilitating DNA demethylation at over half of DME’s targets, specifically those which reside in heterochromatin. These results provide insight into upstream events in the DNA demethylation pathway and reveal the importance of chromatin remodeling for DNA demethylation during <I>Arabidopsis</I> reproduction.</P></▼1><▼2><P>The DEMETER (DME) DNA glycosylase catalyzes genome-wide DNA demethylation and is required for endosperm genomic imprinting and embryo viability. Targets of DME-mediated DNA demethylation reside in small, euchromatic, AT-rich transposons and at the boundaries of large transposons, but how DME interacts with these diverse chromatin states is unknown. The STRUCTURE SPECIFIC RECOGNITION PROTEIN 1 (SSRP1) subunit of the chromatin remodeler FACT (facilitates chromatin transactions), was previously shown to be involved in the DME-dependent regulation of genomic imprinting in <I>Arabidopsis</I> endosperm. Therefore, to investigate the interaction between DME and chromatin, we focused on the activity of the two FACT subunits, SSRP1 and SUPPRESSOR of TY16 (SPT16), during reproduction in <I>Arabidopsis</I>. We found that FACT colocalizes with nuclear DME in vivo, and that DME has two classes of target sites, the first being euchromatic and accessible to DME, but the second, representing over half of DME targets, requiring the action of FACT for DME-mediated DNA demethylation genome-wide. Our results show that the FACT-dependent DME targets are GC-rich heterochromatin domains with high nucleosome occupancy enriched with H3K9me2 and H3K27me1. Further, we demonstrate that heterochromatin-associated linker histone H1 specifically mediates the requirement for FACT at a subset of DME-target loci. Overall, our results demonstrate that FACT is required for DME targeting by facilitating its access to heterochromatin.</P></▼2>

        Tissue integration of zirconia and titanium implants with and without buccal dehiscence defects

        Purpose: The purpose of the present study was to validate an experimental model for assessing tissue integration of titanium and zirconia implants with and without buccal dehiscence defects. Methods: In 3 dogs, 5 implants were randomly placed on both sides of the mandibles: 1) Z1: a zirconia implant (modified surface) within the bony housing, 2) Z2: a zirconia implant (standard surface) within the bony housing, 3) T: a titanium implant within the bony housing, 4) Z1_D: a Z1 implant placed with a buccal bone dehiscence defect (3 mm), and 5) T_D: a titanium implant placed with a buccal bone dehiscence defect (3 mm). The healing times were 2 weeks (one side of the mandible) and 6 weeks (the opposite side). Results: The dimensions of the peri-implant soft tissue varied depending on the implant and the healing time. The level of the mucosal margin was located more apically at 6 weeks than at 2 weeks in all groups, except group T. The presence of a buccal dehiscence defect did not result in a decrease in the overall soft tissue dimensions between 2 and 6 weeks (4.80±1.31 and 4.3 mm in group Z1_D, and 4.47±1.06 and 4.5±1.37 mm in group T_D, respectively). The bone-to-implant contact (BIC) values were highest in group Z1 at both time points (34.15%±21.23% at 2 weeks, 84.08%±1.33% at 6 weeks). The buccal dehiscence defects in groups Z1_D and T_D showed no further bone loss at 6 weeks compared to 2 weeks. Conclusions: The modified surface of Z1 demonstrated higher BIC values than the surface of Z2. There were minimal differences in the mucosal margin between 2 and 6 weeks in the presence of a dehiscence defect. The present model can serve as a useful tool for studying peri-implant dehiscence defects at the hard and soft tissue levels.

        Selective COX (cyclooxygenase)-2 inhibitors including celecoxib have been shown to induce apoptosis and cell cycle changes in various tumor cells. New inhibitors are recently being developed as chemomodulating agents. We evaluated celecoxib and screened 150 synthetic compounds for anti-proliferative activities in vitro. Effects of celecoxib on COX activity, cell growth, cell cycle distribution, and apoptosis induction were determined in A549 COX-2 overexpressing human non-small cell lung cancer (NSCLC) cells. The COX inhibition of celecoxib increased with concentration up to 82% at 1μM after 24 hr exposure. Forty μM and 50μM of celecoxib induce G_1 arrest, and TUNEL-positive apoptotic cells, respectively. Among 150 compounds, several compounds were selected for having greater COX-2 inhibitory activity and higher selectivity than celecoxib with growth inhibitory activity. Celecoxib showed concentration-dependent COX inhibitory activity, and ability to induce cell cycle arrest and apoptosis in human NSCLC cells in vitro. Among synthetic analogues screened, several compounds showed promising in vitro activity as COX-2 inhibitory anticancer agents, which warrant further evaluation in vitro and in vivo.

      • Highly stable trypsin-aggregate coatings on polymer nanofibers for repeated protein digestion

        <P>A stable and robust trypsin-based biocatalytic system was developed and demonstrated for proteomic applications. The system utilizes polymer nanofibers coated with trypsin aggregates for immobilized protease digestions. After covalently attaching an initial layer of trypsin to the polymer nanofibers, highly concentrated trypsin molecules are crosslinked to the layered trypsin by way of a glutaraldehyde treatment. This process produced a 300-fold increase in trypsin activity compared with a conventional method for covalent trypsin immobilization, and proved to be robust in that it still maintained a high level of activity after a year of repeated recycling. This highly stable form of immobilized trypsin was resistant to autolysis, enabling repeated digestions of BSA over 40 days and successful peptide identification by LC-MS/MS. This active and stable form of immobilized trypsin was successfully employed in the digestion of yeast proteome extract with high reproducibility and within shorter time than conventional protein digestion using solution phase trypsin. Finally, the immobilized trypsin was resistant to proteolysis when exposed to other enzymes (i.e., chymotrypsin), which makes it suitable for use in “real-world” proteomic applications. Overall, the biocatalytic nanofibers with trypsin aggregate coatings proved to be an effective approach for repeated and automated protein digestion in proteomic analyses.</P>

      • Signal Integrity Design and Analysis of a Multilayer Test Interposer for LPDDR4 Memory Test With Silicone Rubber-Based Sheet Contact

        <P>As the data rate of Low Power Double Data Rate 4 (LPDDR4) memory now exceeds 3.2 Gb/s, it is becoming more difficult to meet the target specifications. While testing has become of utmost importance, it is not viable to have a direct access to the signal pins in a package on package configuration due to the densely located array of solder balls; instead, a test interposer with an excellent electrical performance needs to be adopted to provide test access. In this paper, we first propose a novel test interposer scheme for testing LPDDR4 memory packages. For accurate testing without significant influence on the intrinsic signal path, the proposed test interposer is designed considering a number of signal integrity issues such as intersymbol interference, jitter, impedance matching, and crosstalk. Furthermore, by adopting silicone rubber sheet in place of soldering, the proposed test interposer enhances reusability of the packages with a fast setup time. Moreover, a reconstruction method is proposed that can reconstruct the voltage at application processor using the waveform captured on the test interposer, instead of probing at the ball gray array directly. Through a series of simulations and measurements, we experimentally verified the proposed test interposer. The proposed test interposer scheme can be widely adopted for testing of high-performance packages with its high accuracy and practicality.</P>

      • Chip-Level Simultaneous Switching Current Measurement in Power Distribution Network Using Magnetically Coupled Embedded Current Probing Structure

        <P>A simultaneous switching current (SSC) drawn by an integrated circuit (IC) creates simultaneous switching noise on power nets, which in turn causes jitters in the I/O signals and reduces the maximum clock frequency. For a thorough analysis of high-speed ICs, there is a dire need to measure currents at specific power pins of the ICs. In this paper, a novel magnetically coupled embedded current probing structure is proposed for measuring the SSC on the chip level resulting from the logical activity of the I/O buffers. SSCs are found by capturing the magnetic flux induced by the SSC of interest, with the proposed embedded current probing structure using magnetic coupling, and then reconstructing the original current waveform using the transfer impedance profile. Through a series of measurements with test vehicles fabricated on the chip level, we experimentally verified the proposed probing structures in the time and frequency domains and proved that they can effectively measure the SSC. Finally, future directions for improvements are discussed at the end of this paper.</P>
