Biological Potential of Novel Specific Casein-Derived Peptides

This study aimed to evaluate the biological potential of functional food, i.e., specific peptides obtained from the hydrolysis of milk protein, by assessing their antioxidant and antibacterial properties. For the preparation of casein hydrolysates, commercial enzymes were added to 10% casein solution in a 1:200 (w/v) ratio, and samples were collected each hour. Based on the assessment of the degree of hydrolysis (DH) of casein hydrolysates, it was observed that the concentration of all enzymatic hydrolysates increased rapidly from 30 to 40 minutes. However, no change was observed in their concentrations after 150 minutes. Protamex<sup>®</sup> and Neutrase<sup>®</sup> exhibited the highest DH when compared to other enzymes. Furthermore, SDS-PAGE was performed for analyzing the proteolytic pattern of each enzyme, except for Flavourzyme<sup>®</sup>, and peptides in the size range of 20-25 kDa were identified. Subsequently, peptides produced by two enzymes were isolated using a preparative liquid chromatography system. Overall, NF3, NF4, PF5, and PF6 showed higher antioxidant potential than other peptide fractions. Moreover, NF7 and PF3 exhibited the highest antibacterial activity. In this study, we evaluated the biological potential of novel caseinderived peptides that may find application in the food and healthcare industry.

The objective of this study was to develop the performance applying conscientiousness among big 5 personality model. Now, young people pursue the conscientiousness following the trend with universalized conscientiousness and want its new value. However, Korea where Confucianism idea is prevailed requires universal conscientiousness. The authors faced this reality and elicited the emotions describing the reality of our society. As a result, they found the ego that required conscientiousness but became mad finally. This study was to investigate what the conscientiousness in the present times requested, and new elements of the conscientiousness are anticipated.
S-256 NT- proBNP complements the ACEF Score in Predicting 3-Year Mortality following AMI

Background: The ACEF score and NT-proBNP are useful in predicting mortality in ACS. But few data exists regarding acute MI. We aimed to use ACEF score and NT-proBNP level to predict risk of long term mortality after AMI. Methods: We enrolled AMI patients who had PCI between January 2004 and December 2009 from 9 university hospitals in South Korea. Patients were ACEF scored and serum NT-proBNP was checked. The primary endpoint was all-cause mortality during a median follow-up of 3.38 years. Results: Of 3217 patients,the all-cause mortality was 18.59% and was significantly related to higher ACEF scores (p<0.001). Higher NT-proBNP level was associated with increased mortality (p<0.001). In a Cox proportional hazards model, the independent predictors of all-cause death included the ACEF score[adjusted HR 1.829, 95% CI 1.603-2.087; p<0.001] and NT-proBNP[adjusted HR 1.657, 95% CI 1.461-1.879; p<0.001]. When NT-proBNP was added to the individual components of the ACEF score,it displaced many factors in the score. The Kaplan-Meier survival curve related to whether the patients had a below or above-median ACEF score and a below or above-median NT-proBNP level (pooled over strata,log rank 301.8, p<0.001). The combination of ACEF score and NT-proBNP improved risk prediction for mortality[AUC, 0.75 (95% CI, 0.734-0.765); p<0.001], which significantly exceeded ACEF score and NT-proBNP (p<0.001). Conclusions: NT-proBNP complements ACEF score for predicting long term mortality.Inclusion of NT-proBNP is useful in risk-stratifying patients following AMI.
Basic, Research : Effects of Flow on Primary Hepatocyte Culture in a Microfluidic Chip

Background: Previous in vitro studies have reported that hepatocyte has unique characteristics. Its resistance to grow in vitro, and it loses its proper functions rapidly. Since portal flow is important in liver regeneration and maintenance of liver function in vivo, maintenance of flow effects on culture system is important. In this study, application of microfluidic technology has enabled development of a chip that can mimic portal flow. The aim of the present study was to compare the effects of flow on primary hepatocyte culture. Methods: Two different types microchips were fabricated, in which the culture medium moves with steady flow. Using optimal flow microchips, 2D monolayer and 3D spheroid of hepatocytes were cultured for 2 weeks. Hepatocyte functional activities were evaluated by albumin secretion and urea synthesis. Results: Flat flow chip showed increased albumin secretion along with improved urea synthesis compared with static culture (Figure 1). Above all, 3D culture on concave chip with flow showed best performance in hepatocyte culture in vitro. Conclusions: These data suggest that maintenance of flow mimicking portal flow has a significant impact on primary hepatocyte culture . This chip may also be a useful tool in basic and translational research using hepatocyte, moreover can be a significant contribution in drug industry by enabled drug toxicity test.
Calcium Solubilization Ability and Anti-Inflammatory Effects of Hydrolyzed Casein

This study performed to evaluate the applicability of functional dairy food materials by comparing the calcium solubilization ability and anti-inflammatory effects of hydrolyzed casein protein. Commercial enzyme (Alcalase??; Neutrase??; Protamex??; Flavourzyme??) was added to the 10% casein solution to prepare the casein hydrolysates. Samples obtained every hour [1:200 (w/v)]. According to results of measuring the degree of hydrolysis (DH), all of four enzymatic hydrolysates increased rapidly from 30 to 40 min, and after 150 min, there were no change. Protamex?? and Neutrase?? had the highest DH compared to others enzymatic hydrolysates. After that, peptides obtained throughout a preparative liquid chromatography system. In the calcium solubility experiments, neutrase fraction (NF) 4 and NF7 showed similar activities with casein phosphopeptide (CPP). In vitro cell experiments showed that no cytotoxicity except for NF6. Also, the production of nitric oxide (NO) inhibited as the concentration of fraction samples increased. The cytokine (IL-1α, IL-6, and TNF-α) production was lower than lipopolysaccharide (+) group significantly. Therefore, the possibility of anti-inflammatory activity found in the hydrolyzed samples. According to the above experiments, NF3 and Protamex Fraction (PF) 3 selected. Amino acids selected throughout an AccQ-Tag system. As a result, 17 species of amino acids and several species of unknown amino acids identified. Both fractions had the highest content of phenylalanine. This study identified the potential of biologically active and functional peptides derived from casein that affect the food and dairy industry.

Background: The number of patients with chronic liver disease or cirrhosis is increasing. For these patients, liver transplantation has been the only solution, but, because of the shortage of donors and immune rejection, human-liver-cell-based therapies using autologous liver resection might be an alternative approach. However, continuous supply of human primary hepatocytes (hHeps) is challenging because of the poor viability of isolated hHeps from partial hepatectomy. Here, we overcame these obstacles by co-culturing hHeps with human adipose- derived stem cells (hADSCs), which can be obtained in large quantities, repeatedly, under local anesthesia. Methods: hHeps were isolated from resection margins after partial hepatectomy using a two-step collagenase technique. hADSCs were from subcutaneous adipose tissue. Spheroids were generated using polydimethylsiloxane-based concavemicromolds. For spheroid formation, a 1:1 mixture of hHeps and hADSCs was inoculated and cultured on microwells. Results: Co-cultured spheroids aggregated more rapidly, and the viability was significantly higher than mono-cultured spheroids. Co-cultured spheroids exhibited higher liver-specific functions compared to mono-cultured, though they had only half the number of hHeps. The albumin and urea level of co-cultured spheroids was 15% and 20% higher on day 7, respectively. A quantitative cytochrome P450 assay showed that enzymatic activity of co-cultured spheroids was 20% higher than mono-cultured spheroids. Conclusions: hADSCs co-cultured with hHeps assist the compact formation of hepatic spheroids and behave like hHeps. Co-cultured spheroids might enable autologous hHeps as a safer method to whole organ transplantation to cure patients, overcoming donor shortages. Additionally, they could be used in the bioartificial liver maintaining cell viability and function.
