      • MicroRNA-214 Regulates the Acquired Resistance to Gefitinib via the PTEN/AKT Pathway in EGFR-mutant Cell Lines

        Patients with non-small cell lung cancer (NSCLC) who have activating epidermal growth factor receptor (EGFR) mutations derive clinical benefit from treatment with EGFR-tyrosine kinase inhibitors ((EGFR-TKIs)-namely gefitinib and erlotinib. However, these patients eventually develop resistance to EGFR-TKIs. Despite the fact that this acquired resistance may be the result of a secondary mutation in the EGFR gene, such as T790M or amplification of the MET proto-oncogene, there are other mechanisms which need to be explored. MicroRNAs (miRs) are a class of small non-coding RNAs that play pivotal roles in tumorigenesis, tumor progression and chemo-resistance. In this study, we firstly successfully established a gefitinib resistant cell line-HCC827/GR, by exposing normal HCC827 cells (an NSCLC cell line with a 746E-750A in-frame deletion of EGFR gene) to increasing concentrations of gefitinib. Then, we found that miR-214 was significantly up-regulated in HCC827/GR. We also showed that miR-214 and PTEN were inversely expressed in HCC827/GR. Knockdown of miR-214 altered the expression of PTEN and p-AKT and re-sensitized HCC827/GR to gefitinib. Taken together, miR-214 may regulate the acquired resistance to gefitinib in HCC827 via PTEN/AKT signaling pathway. Suppression of miR-214 may thus reverse the acquired resistance to EGFR-TKIs therapy.

        Design optimization and development of SMC composite tray

        In the engineering application, trays are easy to break down to result in anchorage failure in the composite anchoring systems. Therefore, the research carried out the force analysis with mechanics of materials to observe the main stress concentration anddeformation of the tray. From the findings of the force analysis, the structure and key parameters of the tray were optimizedwith reference of the existing tray design. Besides, the study turns to the finite element software to simulate and analyze thetray. The results manifest that tray failure during the support mainly results from the expansion and deformation of the taperhole squeezed by the nut, which causes the tray taper hole to rupture and crackle extend, thus leading to its crack. What’smore, the tray breaks for the compression of the tray edge by the surrounding rock. The maximum deformation at the largeend of the optimized tray tapered hole was reduced from 33.8 mm to 4.7 mm, approximately 86% with the shear stress reducedfrom 781.67 Mpa to 258.83 Mpa, about 66.8%. Using Sheet Molding Compound (SMC) to mould trays with new structure andconducting the test of tray bearing capacity, it can be found that its bearing capacity is up to 250 KN. After the taper hole ofthe tray is locally strengthened, its bearing capacity is increased to 304 KN.

        ATM Signaling Pathway Is Implicated in the SMYD3-mediated Proliferation and Migration of Gastric Cancer Cells

        Purpose: We previously found that the histone methyltransferase suppressor of variegation, enhancer of zeste, trithorax and myeloid-nervy-deformed epidermal autoregulatory factor-1 domain-containing protein 3 (SMYD3) is a potential independent predictive factor or prognostic factor for overall survival in gastric cancer patients, but its roles seem to differ from those in other cancers. Therefore, in this study, the detailed functions of SMYD3 in cell proliferation and migration in gastric cancer were examined. Materials and Methods: SMYD3 was overexpressed or suppressed by transfection with an expression plasmid or siRNA, and a wound healing migration assay and Transwell assay were performed to detect the migration and invasion ability of gastric cancer cells. Additionally, an MTT assay and clonogenic assay were performed to evaluate cell proliferation, and a cell cycle analysis was performed by propidium iodide staining. Furthermore, the expression of genes implicated in the ataxia telangiectasia mutated (ATM) pathway and proteins involved in cell cycle regulation were detected by polymerase chain reaction and western blot analyses. Results: Compared with control cells, gastric cancer cells transfected with si-SMYD3 showed lower migration and invasion abilities (P<0.05), and the absence of SMYD3 halted cells in G2/M phase and activated the ATM pathway. Furthermore, the opposite patterns were observed when SMYD3 was elevated in normal gastric cells. Conclusions: To the best of our knowledge, this study provides the first evidence that the absence of SMYD3 could inhibit the migration, invasion, and proliferation of gastric cancer cells and halt cells in G2/M phase via the ATM-CHK2/p53-Cdc25C pathway. These findings indicated that SMYD3 plays crucial roles in the proliferation, migration, and invasion of gastric cancer cells and may be a useful therapeutic target in human gastric carcinomas.

        China and the RMA

        This article explores the possibility of a revolution in military affairs (RMA) in the Chinese People`s Liberation Army. After evaluating the social, economic, and technological foundation for an RMA in China, the authors then outline the theorctical underpinning of the RMA and its relation to military innovation. Using this analysis as a template, the article then assesses the current status of the RMA in the PLA, paying particular attention to C4 I modernization, as well as preparations for the implementation of the RMA at the unit level, including network-based war-gaming, IW training, IW exercises, and acquisition of informationized equipment. The authors conclude that the PLA has already initiated an RMA, whether the Chinese leadership realizes it or not. Further, the rapid informationization of Chinese society and the dual-use nature of the global information economy have permitted the Chinese military to make important strides. At the same time, however, China`s RMA has only begun to change its largely semi-mechanized force, while the RMA in countries such as the United States is much farther along. As n result, Chinese military thinkers have advocated "walking on two legs," pursuing mechanization and informationalization of the PLA simultaneously to carry out both symmetric and asymmetric strategies.

        Modification of C-doped a-SiO2 after Swift Heavy-Ion Irradiation

        Amorphous SiO2 (a-SiO2) thin films were thermally grown on single-crystalline silicon. These a-SiO2/Si samples were first implanted (C-doped) with 100-keV carbon ion at room temperature (RT) at a dose of 5.0 × 1017 C-ions/cm2 and were then irradiated at RT by using 853 MeV Pb ions at doses of 5.0 × 1011, 1.0 × 1012, 2.0 × 1012, and 5.0 × 1012 Pb-ions/cm2, respectively. The microstructures and the photoluminescence (PL) properties of these samples induced by Pb ions were investigated using fluorescence spectroscopy and transmission electron microscopy. We found that high-energy Pb-ion irradiation could induce the formation of a new phase and a change in the PL property of C-doped a-SiO2/Si samples.         NATURE AND CATALYTIC PERFORMANCE OF PtSAPO-5 MOLECULAR SIEVE CATALYST SYNTHESIZED BY ONE STEP

        Pt catalysts supported on silicoaluminophosphate molecular sieves were synthesized by adding platinum salts directly to the gel mixture of SAPO. The synthesized products were marked as PtSAPO-5, in order to distinguish with the SAPO-5 supported platinum catalyst prepared by impregnation, which was marked as Pt/SAPO-5. The as-synthesized and calcined PtSAPO-5 samples were characterized by XRD, ^(29)Si MAS NMR, ^(129)Xe NMR and NH₃-TPD, and their catalytic activities were evaluated by the, hydroisomerization of n-hexane. The results revealed that in PtSAPO-5, the platinum could catalyze the removal of the templates entrapped in the as-synthesized samples. The presence of Pt in the gel mixture can increase silicon content in the framework of SAPO-5, and the acid sites. In the activated PtSAPO-5 catalyst, Pt was highly dispersed in the channel of SAPO-5 molecular sieve. Pt exists in the form of big particles outside of the channel of Pt/SAPO-5 prepared by impregnating method, and impregnation with H₂PtCl_6 improved the strength of acid sites. PtSAPO-5 has a higher activity and selectivity than Pk/SAPO-5 in the hydroisomerization of n-hexane. This is because the former has medium acid sites and high dispersion of Pt, while the channel of Pt/SAPO-5 is somewhat retarded by the big platinum particles, and the acid sites are stronger than the former.

        Seismic experiment and analysis of rectangular bottom strengthened steel-concrete composite columns

        In order to study the working mechanism of rectangular steel-concrete composite columns subjected to compression-bending load and further determine the seismic performance index, a bottom strengthened rectangular steel reinforced concrete (SRC) column with concealed steel plates and a bottom strengthened rectangular concrete filled steel tube (CFST) columns were proposed. Six column models with different configurations were tested under horizontal low cyclic loading. Based on the experiments, the load-bearing capacity, stiffness and degradation process, ductility, hysteretic energy dissipation capacity, and failure characteristics of the models were analyzed. The loadbearing capacity calculation formulas for a normal section and an oblique section of bottom strengthened rectangular steel-concrete composite columns were pesented and a finite element (FE) numerical simulation of the classical specimens was performed. The study shows that the load-bearing capacity, ductility, and seismic energy dissipation capacity of the bottom strengthened rectangular steel-concrete composite columns are significantly improved compared to the conventional rectangular steel-concrete composite columns and the results obtained from the calculation and the FE numerical simulation are in good agreement with those from the experiments. The rectangular steel-concrete composite column with bottom strengthened shows better seismic behavior and higher energy dissipation capacity under suitable constructional requirements and it can be applied to the structure design of highrise buildings.

        Adaptive Learning Path Recommendation based on Graph Theory and an Improved Immune Algorithm

        Adaptive learning in e-learning has garnered researchers’ interest. In it, learning resources could be recommended automatically to achieve a personalized learning experience. There are various ways to realize it. One of the realistic ways is adaptive learning path recommendation, in which learning resources are provided according to learners’ requirements. This paper summarizes existing works and proposes an innovative approach. Firstly, a learner-centred concept map is created using graph theory based on the features of the learners and concepts. Then, the approach generates a linear concept sequence from the concept map using the proposed traversal algorithm. Finally, Learning Objects (LOs), which are the smallest concrete units that make up a learning path, are organized based on the concept sequences. In order to realize this step, we model it as a multi-objective combinatorial optimization problem, and an improved immune algorithm (IIA) is proposed to solve it. In the experimental stage, a series of simulated experiments are conducted on nine datasets with different levels of complexity. The results show that the proposed algorithm increases the computational efficiency and effectiveness. Moreover, an empirical study is carried out to validate the proposed approach from a pedagogical view. Compared with a self-selection based approach and the other evolutionary algorithm based approaches, the proposed approach produces better outcomes in terms of learners’ homework, final exam grades and satisfaction.
