Objective: This study was designed to investigate the effects of an Aspergillus sulphureus xylanase expressed in Pichia pastoris on the growth performance, nutrient digestibility and gut microbes in weanling pigs. Methods: A total of 180 weanling pigs (initial body weights were $8.47{\pm}1.40kg$) were assigned randomly to 5 dietary treatments. Each treatment had 6 replicates with 6 pigs per replicate. The experimental diets were wheat based with supplementation of 0, 500, 1,000, 2,000, and 4,000 U xylanase/kg. The experiment lasted 28 days (early phase, d 0 to 14; late phase, d 15 to 28). Results: In the early phase, compared to the control, average daily gain (ADG) was higher for pigs fed diets supplemented with xylanase and there was a quadratic response in ADG (p<0.05). In the entire phase, ADG was higher for the pigs fed 1,000 or 2,000 U/kg xylanase compared to the control (p<0.05). The gain to feed ratio was higher for pigs fed diets supplemented with 1,000 or 2,000 U/kg xylanase compared to the control (p<0.05). Increasing the amount of xylanase improved the apparent total tract digestibility of dry matter, crude protein, neutral detergent fiber, calcium, and phosphorus during both periods (p<0.05). Xylanase supplementation (2,000 U/kg) decreased the proportion of Lachnospiraceae (by 50%) in Firmicutes, but increased Prevotellaceae (by 175%) in Bacteroidetes and almost diminished Enterobacteriaceae (Escherichia-Shigella) in Proteobacteria. Conclusion: Xylanase supplementation increased growth performance and nutrient digestibility up to 2,000 U/kg. Supplementation of xylanase (2,000 U/kg) decreased the richness of gut bacteria but diminished the growth of harmful pathogenic bacteria, such as Escherichia-Shigella, in the colon.

Objective: This study aimed to investigate the effects of dietary supplementation of a fatty acid-balanced oil, instead of soybean oil, on reproductive performance, nutrient digestibility, blood indexes, milk composition in lactating sows, and fecal microbial composition in piglets. Methods: Twenty-four sows (Landrace×Yorkshire, mean parity 4.96) were randomly allotted to two treatments with twelve pens per treatment and one sow per pen based on their backfat thickness and parity. The experiment began on day 107 of gestation and continued until weaning on day 21 of lactation, lasting for 28 days. The control group (CG) was fed a basal diet supplemented with 2% soybean oil and the experimental group (EG) was fed the basal diet supplemented with 2% fatty acid-balanced oil. Results: The fatty acid-balanced oil supplementation increased (p<0.05) the apparent total tract digestibility of dry matter, crude protein, and gross energy in sows. The lower (p<0.05) serum high-density lipoprotein cholesterol and albumin levels of sows were observed in the EG on day 21 of lactation. Dietary supplementation with the fatty acid-balanced oil decreased the fat content, increased the immunoglobulin G level, and changed (p<0.05) some fatty acid content in milk. Moreover, the fatty acid-balanced oil supplementation changed (p<0.05) the fecal microbial composition of piglets, where the average relative abundance of Spirochaetota was decreased (p<0.05) by 0.55% at the phylum level, and the average relative abundance of some potentially pathogenic fecal microorganism was decreased (p<0.05) at the species level. Conclusion: The fatty acid-balanced oil improved nutrient digestibility, changed the serum biochemical indices and milk composition of sows, and ameliorated the fecal microbial composition of piglets.
Cytotoxic lignans from Viburnum foetidum

Two new lignans, 3,4,40-trihydroxy-30,9-dimethoxy-9,90-epoxylignan (1), 3,40-dihydroxy-30,4, 9-trimethoxy-9,90-epoxylignan (2), together with one known compound,4,40-dihydroxy-3,30,9-trimethoxy-9,90-epoxylignan(3), were isolated from the 95 % EtOH extract of Viburnumfoetidum. The structures of the two new compounds wereelucidated on the basis of 1D, 2D-NMR, and mass spectralanalysis. All the lignans were in vitro evaluated for theircytotoxic activities against four tumor cell lines (A549, SKOV-3, SKMEL-2 and HCT15).
Inhomogeneous Fatigue Characteristics of Welded Ti–6Al–4V Thick Plate by the Electron Beam Method

To recognize the specific safety problem brought by an increasing thickness, the layered microstructures, fatigue propertiesand the fractographies of a 100 mm-thick Ti–6Al–4V joint via electron beam welding were investigated. The results showthat the seam zone has textural and mechanical heterogeneities in the layered thickness regions. The mid layer has the lowestfatigue life and different fracture-position tendency (more broken in seam zone) due to the hybrid influences of the grain sizeand the phase fraction of α′ and β phases. The fatigue cracks initiate on the surfaces of fatigue specimens and grow in thetransgranular fracture mode. The fatigue deformation mode is adapted to the planar-slip mechanism with different degreesof dislocation blocking in distinct layers, which results in a “mid-layer favorite” crack initiation trend. Moreover, the midlayer has the highest crack propagation rate either.
