        Investigation on shaping machining method for deep hole keyway based on on-line symmetry detection and compensation

        Machining of deep hole keyway often suffers from low processing accuracy, low processing symmetry degree between keyway and the reference plane and is difficult to control. In order to enhance the processing quality, a mathematical model of space symmetry error was established based on space projection, orthogonal combination and linear fitting methods so as to study rotatable and shift-able space objects. The space symmetry error was separated into included angle error and displacement error between the fitting straight line and the benchmark line. Through included angle rotation and location measurement along with quantitative translation, the space symmetry error compensation was realized. On the basis of the proposed methods, a Double combined manual self-centering fixture (DCMSF) was designed to realize the radial self-centering and axial location for effective clamping of deep hole keyway, and the reference plane of builtin straight slot was derived out to make it detectable and controllable. Meanwhile, an Online multi-degree of freedom symmetry detection device (OMFSDD) and corresponding Numerical control (NC) programs were also designed for automatic detection and control of symmetry error. Subsequently, mass production of high symmetric deep hole keyway were realized by means of NC gear shaper servo control of rotating shaft angle compensation and bi-directional cutter back-off amount translation compensation, and combination of NC control of fixed angle processing. Deep hole keyway processing experiments indicate that the proposed method can steadily keep the symmetry within 0.03 mm. Thus, the rationality and accuracy of the proposed method was validated which provides a novel approach to high precision deep hole keyway processing.

        Identity-based Deniable Authenticated Encryption for E-voting Systems

        Deniable authentication (DA) is a protocol in which a receiver can generate an authenticator that is probabilistically indistinguishable from a sender. DA can be applied in many scenarios that require user privacy protection. To enhance the security of DA, in this paper, we construct a new deniable authenticated encryption (DAE) scheme that realizes deniable authentication and confidentiality in a logical single step. Compared with existing approaches, our approach provides proof of security and is efficient in terms of performance analysis. Our scheme is in an identity-based environment; thus, it avoids the public key certificate-based public key infrastructure (PKI). Moreover, we provide an example that shows that our protocol is applicable for e-voting systems.

        Transcriptome-based identification of the optimal reference genes as internal controls for quantitative RT-PCR in razor clam (Sinonovacula constricta)

        Quantitative real-time PCR (qRT-PCR) is a standard method to measure gene expression in function exploring. Accurate and reproducible data of qRT-PCR requires appropriate reference genes, which are stably expressed under different experimental conditions. However, no housekeeping genes were validated as internal controls for qRT-PCR in Sinonovacula constricta. In this study, we classified the transcriptome data of two tissues for Vibrio infection and Cd2+ stress into ten clusters based on the gene expression patterns. Among them, cluster 5 had the most stable gene expression patterns regardless of tissues and treatments as the database for candidate reference genes. A total of 55 orthologs of classical housekeeping genes in the clam transcriptome were annotated. Combined the expression profiles and housekeeping genes in S. constricta, we chose eight candidate reference genes and validated their expression in Vibrio-infected samples and different tissues by qRT-PCR. Their expression stability was analyzed by three different algorithms geNorm, NormFinder and BestKeeper. Although the rank of the eight candidate reference genes is different in different treatments using different software, RS9 could be the best reference genes for normalization of qRT-PCR expression data in S. constricta under various treatments considering the above analysis. Meanwhile, the ranking of genes based on the CV values of transcriptomic data was similar to the validation results. This study provides for the first time a list of suitable reference genes for S. constricta and a valuable resource for further studies of clam immune defense systems.

        A Pilot Study of Clinical Evaluation and Formation Mechanism of Irritable Bowel Syndrome-like Symptoms in Inflammatory Bowel Disease Patients in Remission

        Background/Aims Some inflammatory bowel disease (IBD) patients in remission suffer from irritable bowel syndrome (IBS)-like symptoms (IBD-IBS). The pathogenesis has not yet been elucidated. The study aim is to evaluate relationships among quality of life (QOL), psychological status, and visceral sensitivity, and explore the formation mechanism of IBD-IBS. Methods Forty-seven patients with Crohn’s disease in remission, 24 ulcerative colitis in remission, 26 IBS, and 20 healthy controls were included in the study. The abdominal pain, QOL, anxiety, and depression were evaluated through questionnaires. Visceral sensitivity was measured by rectal balloon distension. The serum levels of 5-hydroxytryptamine (5-HT) and nerve growth factor (NGF) were measured by enzyme-linked immunosorbent assay. The expressions of tryptase, 5-HT, NGF, and related receptors in colonic tissues were detected by immunohistochemistry and western blot. Results Prevalence of IBS-like symptoms in Crohn’s disease and ulcerative colitis patients in clinical remission was 29.8% and 50.0%, respectively. The QOL was lower, the anxiety/depression scores were higher in IBD-IBS patients than those without IBS-like symptoms. Additionally, patients with IBD-IBS existed visceral hypersensitivity. Besides, abdominal pain was associated with poor QOL, visceral hypersensitivity, anxiety, and depression in IBD-IBS patients. The number of mast cells (MCs) and expressions of 5-HT, NGF, and related receptors were higher in IBD-IBS patients than those with no such symptoms. The serum levels of 5-HT and NGF positively correlated with abdominal pain and visceral hypersensitivity. Conclusion IBD-IBS patients may have low QOL and psychological abnormalities, as wells as visceral hypersensitivity which may be related to increased 5-HT and NGF levels released from activated mast cells.         New Analysis of Reduced-Version of Piccolo in the Single-Key Scenario

        The lightweight block cipher Piccolo adopts Generalized Feistel Network structure with 64 bits of block size. Its key supports 80 bits or 128 bits, expressed by Piccolo-80 or Piccolo-128, respectively. In this paper, we exploit the security of reduced version of Piccolo from the first round with the pre-whitening layer, which shows the vulnerability of original Piccolo. As a matter of fact, we first study some linear relations among the round subkeys and the properties of linear layer. Based on them, we evaluate the security of Piccolo-80/128 against the meet-in-the-middle attack. Finally, we attack 13 rounds of Piccolo-80 by applying a 5-round distinguisher, which requires 2<sup>44</sup> chosen plaintexts, 2<sup>67.39</sup> encryptions and 2<sup>64.91</sup> blocks, respectively. Moreover, we also attack 17 rounds of Piccolo-128 by using a 7-round distinguisher, which requires 2<sup>44</sup> chosen plaintexts, 2<sup>126</sup> encryptions and 2<sup>125.49</sup> blocks, respectively. Compared with the previous cryptanalytic results, our results are the currently best ones if considering Piccolo from the first round with the pre-whitening layer.

        miR-195/miR-497 Regulate CD274 Expression of Immune Regulatory Ligands in Triple-Negative Breast Cancer

        Purpose: Immune suppression is common in patients with advanced breast cancer but the mechanisms underlying this phenomenon have not been sufficiently studied. In this study, we aimed to identify B7 family members that were able to predict the immune status of patients, and which may serve as potential targets for the treatment of breast cancer. We also aimed to identify microRNAs that may regulate the expression of B7 family members. Methods: The Cancer Genome Atlas data from 1,092 patients with breast cancer, including gene expression, microRNA expression and survival data, were used for statistical and survival analyses. Polymerase chain reaction and Western blot were used to measure messenger RNA and protein expression, respectively. Luciferase assay was used to investigate direct microRNA target. Results: Bioinformatic analysis predicted that microRNA (miR)-93, miR-195, miR-497, and miR-340 are potential regulators of the immune evasion of breast cancer cells, and that they exert this function by targeting CD274, PDCD1LG2, and NCR3LG1. We chose CD274 for further investigations. We found that miR-195, miR-497, and CD274 expression levels were inversely correlated in MDA-MB-231 cells, and miR-195 and miR-497 expressions mimic inhibited CD274 expression in vitro. Mechanistic investigations demonstrated that miR-195 and miR-497 directly target CD274 3´ untranslated region. Conclusion: Our data indicated that the level of B7 family members can predict the prognosis of breast cancer patients, and miR-195/miR-497 regulate CD274 expression in triple negative breast cancer. This regulation may further influence tumor progression and the immune tolerance mechanism in breast cancer and may be able to predict the effect of immunotherapy on patients.
