        곰팡이, 효모 및 유산균에 의한 보리의 혼합발효

        곰팡이로써는 Rhizopus delemar IFO 4746, 효모로써는 Hansenula anomala IFO 0568, 유산균으로써는 Lactobacillus sp.를 선발하여 이들을 고체상태의 찐 보리기질에 동시에 접종한 후 30℃로 2일, 37℃로 3일 혼합배양하여 풍미가 우수한 보리발효산물을 얻었다. 발효시 시간경과에 따라 pH는 24시간만에 급격히 떨어져 배양 5일후에는 3.4였고, 산도는 계속 증가하여 5일후에는 1.9%였으며, 색도는 계속 감소하였다. Fermented barley food was produced by the mixed culture fermentation with a tri-culture system of a mold, a yeast and a lactobacillus. When Rhizopus delemar IFO 4746, Hansenular anomala IFO 0568 and Lactobacillus sp. L-5 were selected and cultivated on steamed barley at 30℃ for 2days and 37℃ for 3days, the fermented product of good quality was obtained. During fermentation, changes in acidity, pH, water content and color of fermented barley were examined.

        전분분해효소와 유산균에 의한 보리의 유산발효

        효소로써는 태평양화학주식회사에 액화아밀라아제를, 유산균으로써는 Lactobacillus sp. L-5를 선발하여, 이들을 고체상태의 찐 보리기질에 동시에 접종한 후 37℃로 48시간 배양한 결과 풍미가 우수한 보리의 유산발효산물이 얻어졌다. 또한, 보리기질의 수분함량을 70%내외로 할 경우 발효산물의 물성이 우수하였다. 회전드럼형발효조를 사용하여 37℃, 15rpm의 조건하에서 발효할 경우, 유산균은 대수증식기에서 0.6hr^-1의 비증식속도로 증식하여 배양 12시간만에 정체기로 들어갔으며, 산도는 계속 증가하여 배양 48시간후에 1.17%였다. 발효경과에 따라 보리맛은 계속 감소하여 48시간 배양후에는 요구르트적 풍미를 갖는 호상(糊狀)의 발효산물로 되었다. Fermented barley food was produced by the combining action of an enzyme and a lactobacillus. When Lactobacillus sp. L-5 and commercial liquefying amylase from Tae Pyeong Yang Chemical Co. were selected, inoculated on steamed barley and cultivated at 37℃ for 48hrs, the fermented product of good quality was obtained. In batch cultivation using rotary drum fermentor, viable cell count reached 1.1×10 exp (9)CFU/g after 12hrs' cultivation, and specific growth rate in logarithmic phase was 0.6hr^-1. Viable cell count, acidity, pH, concentration of reducing sugar and viscosity of the 48hrs' fermentation product from rotary drum fermentor was 4.3×10 exp (8)CFU/g. 1.17%, 3.1, 10.7% and 1430cp.

        五拗湯이 家兎와 白鼠의 呼吸 및 循環器系에 미치는 影響

        Cheongyeuldodamtang(CDT) have been used in oriental medicine for many centuries as a therapeutic agent for heat syndrome of apoplexy. The effects of CDT on the vascular system is not known. The purpose of this Study was to investigate effects of CDT on the dial arterial diameter, action mechanism of CDT and effects of CDT on the blood flow improvement in pathologic model injected lipopolysaccharide(LPS) 0.2㎖/hr/kg. The changes of Cerebral Hemodynamics [regional cerebral blood flow(rCBF), mean arterial blood pressure(BP), cardiac muscle contractile force(CMF), pial arterial diameter] were determinated by Laser-Doppler Flowmetry(LDF) and video microscopy methods and video analyzer. The results were as follows , Pial arterial diameter was significantly increased by CDT in a dose-dependent manner, A blood vessle movement induced LPS was recovered by CDT. Preteatment with L-NNA significantly inhibited CDT induced increased rCBF CDT inhibited CMF by pretreatment with L-NNA Pretreatment with methylene blue significantly inhibited CDT induced increased rCBF. Pretreatment with methylene blue accelerated CDT induced non-affected BP. CDT accelerated CMF by pretreatment with methylene blue. CDT induced non-affected BP. CDT decreased CMF by pretreatment with indomethacin. These results suggest that CDT causes a diverse response of cerebral hemodynamics (rCBF, BP, CMF and pial arterial diameter). The cerebral hemodynamics(rCBF, BP, CMF) is also mediated by nitric oxide synthase, cyclic GMP(guanylyl cyclase) and prostaglandin (cyclooxygenase).         고빈도 120 Hz 전침이 Cold-Restraint 스트레스 유발 흰쥐의 위점막에 미치는 영향(影響)

        Objective: The present studies investigated the effects of 120 Hz high frequency electroacupuncture (EA) on the stress-induced stomach dysfunction in relation to its effect on the level of stress hormones and gastric mucosal damages. The gastric mucosal injury was induced by cold-restraint stress and two acupoints corresponding to Zusanli and Sanyinjiao in man were used. Methods: Cold-restraint stress produced typical gastric lesions in all rats of the stressed groups, but the number of ulcers as well as the mean ulcer diameter were reduced by 120 Hz EA pre-treatment. Results: The degranulation value of gastric mast cell was significantly higher in cold-restrained rats than in control ones. However, with the significant reduction of degranulation values of gastric mast cells in EA pre-treated rats compared with cold-restrained rats Cold-restarint stress induced an elevated mRNA expression of pro-inflammatory gone such as cyclooxygenases-2 and tumor necrosis factor (TNF)-α, but these expression were down-regulated in EA pre-treated rats. Immunohistochemical analysis showed that while the inhibitory-κBα and TNF-α immunorection in the surface epithelium of the stomach tended to increase, both reactions in the EA pre-treated rats showed similar pattern as observed in controls. Conclusion: These results suggest that 120 Hz EA may act as a therapeutical means for gastric mucosal damages through an activation of pituitary adrenal system. It could be concluded that 120 Hz high frequency electroaucupuncture affords a good protective potential against stress-induced gastrointestinal dysfunction.

      • Modified Conductance Method for Extraction of Subgap Density of States in a-IGZO Thin-Film Transistors

        <P>We propose a modified conductance method for extraction of the subgap density of states (DOS) in amorphous indium-gallium-zinc oxide thin-film transistors by using the measured capacitance and conductance through the capacitance-voltage (C-V) measurement. In the proposed method, the subgap DOS [g<SUB>A</SUB>(E)] is extracted from the frequency-dispersive C-V characteristics by localized traps in the active channel region. The extracted g<SUB>A</SUB>(E) shows a superposition of the exponential tail states and the exponential deep states over the bandgap (N<SUB>TA</SUB> = 3 × 10<SUP>18</SUP> cm<SUP>-3</SUP> · eV<SUB>-1</SUB>, N<SUB>DA</SUB> = 2.8 × 10<SUP>17</SUP> cm<SUP>-3</SUP> · eV-1, kT<SUB>TA</SUB> = 0.04 eV, and kT<SUB>DA</SUB> = 0.77 eV). We note that the gate-bias-dependent Cfree by free electron charges can be separated from C<SUB>loc</SUB> by localized trap charges through the proposed method.</P>

        자궁경부암 세포의 증식에 관여하는 annexin-I의 역할

        목적: 인간 자궁경부암에서 annexin-I의 역할을 규명하기 위해 annexin -I의 발현과 암세포증식과의 상관관계를 조사하였다. 방법: 면역조직화학 (immunohistochemical) 염색법 과 Western blotting으로 자궁경부암 조직에서 annexin-I 발현의 정도와 분포를 조사하였다. 악성세포주인 SiHa와 HeLa 세포주를 tamoxifen, β-estradiol과 all-trans retinoic acid로 3일 또는 5일간 처리한 후 각 세포의 증식을 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT ) colorimetric assay로 측정하였고 동시에 유세포측정기(flow cytometry)로 annexin-I의 발현을 조사하였다. 결과: Annexin-1에 대한 면역조직화학 분석 결과 자궁경부암세포가 진한 과립상으로 염색되어 관찰되었으며, Western blotting 분석에서는 annexin-I의 발현이 정상이나 양성조직보다 암조직에서 증가하였다. 한편, tamoxifen, β-estradiol 또는 all-trans retinoic acid와 같은 세포증식 조절물질을 이용한 세포주 실험에서는, tamoxifen이나 β-estradiol로 SiHa와 HeLa 세포주를 증식시킨 뒤 annexin-I의 발현이 증폭되었고 all-trans retinoic acid에 의한 항증식 후 annexin-I의 발현이 감소되었다. 결론: Annexin-I의 발현은 정상조직보다 자궁경부암에서 증가되어 있었고 자궁경부암 세포주의 증식과 비례해서 annexin-I의 발현이 증가되었다 Objectives: To investigate the role of annexin-I in human cervical cancer, we evaluated the expression of annexin-I and the relation with the proliferation of cancer cells. Methods: By immunohistochemical analysis and the western blotting of annexin-I , we investigated the extent and distribution of the expression of annexin-I in cervical cancer tissues. After treating the human cancer cell lines ( SiHa and HeLa cell lines ) with tamoxifen, estradiol, and retinoic acid for 5 days to make the cells proliferate and antiproliferate, we measured the proliferation simultaneously with 3-(4,5- dimethyl thiazol- 2-yl)-2,5-diphenyl-tetr -azolium bromide (MTT) colorimetric assay and the expression level of annexin-I with flowcytometry. Results: In the immunohistochemical stains, a granular staining pattern involving the entire cytoplasm was more heavily observed in malignant lesions than in normals. In the western blotting, the antibodies against 35-kDa annexin-I appeared to react more strongly with the lysates of cancer tissues than normal and benign tissues. In SiHa and HeLa cell lines with tamoxifen and β- estradiol treatment, increased expressions of annexin-I were noted with correlated increased proliferation of cells, and with the treatments of all trans retinoic acid, decreased expressions of annexin-I were noted with correlated decreased proliferation of cells. Conclusions: The results suggest that the expression of annexin-I might correlate with cervical cancer than normal and the proliferation of cancer cells.
