        발효 황기 추출물의 항산화, 항염, 콜라게나아제 및 엘라스타아제 활성 저해 효과

        본 연구는 발효 황기 추출물이 발효 전 황기 추출물 보다 항산화, 항염, 콜라게나아제 및 엘라스테아제 억제 효과가 뛰어난지 평가하고자 하였다. FAE의 총 폴리페놀 및 플라보노이드 함량이 AE보다 높은 것으로 나타났으며, DPPH, ABTS 및 환원력 측정에서 FAE에서 AE보다 높은 항산화 활성을 보였다. HaCaT 세포에 SIN-1을 처리하여 효소 활성을 측정하였을 때 FAE 추출물의 처리는 감소된 SOD, catalase, GPx 활성을 AE보다 증가시키는 것을 확인하였다. 또한 LPS로 자극된 RAW264.7 세포에서 증가된NO, PGE2 및 IL-6의 발현을 AE보다 FAE에서 더 억제하는 것을 확인하였다. 또한, collagenase와 elastase의 억제활성이 AE보다 FAE에서 우수하였다. 종합하자면, FAE는 AE보다 항산화, 항염, 콜라게나아제 및 엘라스타아제 억제 효능이 있는 천연기능성 소재 및 건강기능식품 등의 소재로 활용될 수 있을 것이라 사료된다. We aimed to evaluate the antioxidant, anti-inflammatory, anti-collagenase, and anti-elastase effects of fermented and non-fermented Astragalus membranaceus extracts (FAE and AE). The total polyphenol and flavonoid contents of FAE were higher than those of AE. DPPH, ABTS, and reducing power assays confirmed that the radical scavenging efficacy and reducing power of FAE were higher than those of AE. In SIN-1-stimulated HaCaT cells, reduced superoxide dismutase, catalase, and glutathione peroxidase activity levels were rescued in the FAE group but not in the AE group. FAE also suppressed increases in NO, PGE2, and IL-6 levels more potently than AE in LPS-stimulated RAW264.7 cells. FAE also exhibited more potent collagenase and elastase inhibitory activities than AE. In summary, this study suggests that FAE can be used in functional foods and cosmetics with antioxidant, anti-inflammatory, anti-collagenase, and anti-elastase properties.

        배추의 형질전환용 선발항생제로서 Paromomycin의 이용

        정상' 배추와 '서울' 배추의 배축절편을 선발마커로서 hygromycin 저항성유전자를 갖고 있는 pCAMBIA1301와 paromomycin저항성유전자를 갖고 있는 pPTN290으로 각각 형질 전환된 LBA4404 또는 EHA101균주와 공동배양한 후 선발배지에서 배양하면서 형질전환체를 선발하였다. 형질전환빈도는 사용된 항생제와 품종에 따라서 현저하게 차이가 있었으며, 특히 paromomycin은 hygromycin보다 효과적이었고 정상 배추는 서울배추보다 양호하였다. 가장 높은 형질전환빈도는 (0.70%) 100mg/L paromomycin이 첨가된 선발배지에서 정상배추의배축을 배양할 경우 얻어졌다. GUS양성반응으로 확인 한 결과 정상배추에서 9개체와 서울배추에서 3개체를 각각 얻었으며, 온실에서 생장한 후 $T_1$종자를 수확하였다. $T_1$ 종자를 다시 발아시켜 유식물체를 얻은 후 GUS양성반응을 확인함으로서 외래유전자가 안정적으로 발현하고 있음을 확인하였다. Hypocotyl explants of Chinese cabbage (us. 'Jeong Sang' and 'Seoul') produced adventitious shoots on Murashige and Skoog (MS) basal medium supplemented with 4mg/L $AgNO_3$, 5 mg/L acetosyringone, 4 mg/L 6-benzyladenine and 3mg/L alpha-naphthaleneacetic acid (SI) after cocoultivation with strains of Agrobacterium tumefaciens (LBA4404) harboring the pCAMBIA1301 and the $_PPTN290$ containing hygromycin-resistance gene and paromomycin-resistance gene as a selectable marker genes, respectively. There was a significant difference in the frequency of transgenic plants depending on antibiotics and cultivars used. Paromomycin was better than hygromycin, and cultivar 'Jeong-sang' was higher than 'c.v. Seoul' in the frequency of transgenic plants. In particular, the highest frequency (0.70%) of transgenic plants was obtained from selection medium (SI) containing 100mg/L paromomycin in c.v., 'Jeong-sang' GUS positive response were obtained 9 plants and 3 plants from the cultivars, 'Jeong-sang' and 'Seoul', respectively. They were grown to maturity in a greenhouse and normally produced $T_1$ seeds. GUS histochemical assay for progeny $(T_1)$ revealed that the transgenes were expressed in the plant genome.

        배추의 배축절편으로부터 캘러스와 뿌리 발생을 통한 안정적 형질전환

        '정상' 배추의 배축절편을 선발마커로서 paromomycin 저항 성유전자를 갖고 있는 pPTN290으로 각각 형질전환된 EHA101, LBA4404, GV3101균주와 공동배양한 후 갤러스유도배지에서 형질전환캘러스를 얻은 후, 뿌리유도배지에서 부정근을 그리고 신초유도배지에서 신초를 각각 순차적으로 유도하였다. 형질전환캘러스를 얻은 후, 뿌리유도배지에서 부정근을 그리고 신초유도배지에서 신초를 가각 순차적으로 유도하였다. 형질전환캘러스 형성은 Agrobacterium균주에 따라 차이가 있었으며, 특히 EHA101균주에 공동배양된 배축절편으로부터 최대 6.1%까지 얻어졌다. 또한 각각의 형질전환캘러스 클론으로부터 형질전환 부정근과 신초 발생은 EHA101균주에서 60.7%와 38.2%, LBA4404에서 8.3%와 0%, GV3101에서 20.5%와 85.7%까지 각각 얻을 수 있었다. 형질전환식물체는 특별한 형태적 이상 없이 온실에서 정상적으로 자라 $T_{2}$종자를 얻을 수 있었다. GUS방법으로 7개의 후대 유식물체를 분석한 결과 gus유전자가 안정적으로 발현하고 있음을 확인하였고, 배추 genome에 single 또는 multiple copy로 전달되고 있음을 추측할 수 있었다. Hypocotyl explants of Chinese cabbage (cvs. "Jeong Sang") produced transgenic calli on callus induction medium (MS salt, B5 vitamin, 5 mg/L acetosyringone, 1 mg/L 2,4-D, 3% sucrose, 400 mg/L cefotaxime, 100 mg/L paromomycin, pH 5.8) after cocultivation with strains of Agrobacterium tumefaciens (EHA101, LBA4404, GV3101) harboring the pPTN290 containing paromomycin-resistance gene as a selectable marker, and then they transferred to root induction medium (1/2MS salt, MS vitamins, 2% sucrose, 100 mg/L paromomycin, 100 mg/L cefotaxime, pH 5.8) and shoot induction medium (MS salt, B5 vitamin, 4 mg/L $AgNO_3$, 4 mg/L 6-benzyladenine, 3 mg/L alpha-naphthaleneacetic acid, 100 mg/L paromomycin, 100 mg/L cefotaxime, 3% sucrose, pH 5.8) in order. There was a significant difference in the frequency of transgenic calli depending on Agrobacterium strains. In particular, the highest frequency (6.1%) of transgenic calli was obtained from the hypocotyls cocultivated with EHA101 strains. Also, the frequency (%) of transgenic root and plants from each transgenic callus clone were obtained with 60.7% and 38.2% in EHA101, with 8.3% and 0% in LBA4404, with 20.5% and 85.7% in GV3101 strains, respectively. They were grown to maturity in a greenhouse and normally produced $T_2$ seeds. GUS histochemical assay for progeny ($T_2$) revealed that the transgenes was expressed in the plant genome, and progeny analysis from 7 independent transgenic events demonstrated that the transformants transmitted the transgene as a single or multiple functional locus.

        Genome-wide Screening to Identify Responsive Regulators Involved in the Virulence of Xanthomonas oryzae pv. oryzae

        Two-component systems (TCSs) are critical to the pathogenesis of Xanthomonas oryzae pv. oryzae (Xoo). We mutated 55 of 62 genes annotated as responsive regulators (RRs) of TCSs in the genome of Xoo strain PXO99A and identified 9 genes involved in Xoo virulence. Four (rpfG, hrpG, stoS, and detR) of the 9 genes were previously reported as key regulators of Xoo virulence and the other 5 have not been characterized. Lesion lengths on rice leaves inoculated with the mutants were shorter than those of the wild type and were significantly restored with gene complementation. The population density of the 5 mutants in planta was smaller than that of PXO99A at 14 days after inoculation, but the growth curves of the mutants in rich medium were similar to those of the wild type. These newly reported RR genes will facilitate studies on the function of TCSs and of the integrated regulation of TCSs for Xoo pathogenesis.

        Analysis of a Survey on Public Perceptions and Preferences of the “ Smart Green Office ” to Help Encourage Creation of the Program

        본 연구는 설문을 통해 ‘스마트 그린오피스(Smart Green Office)’에 대해 대중의 인식과 선호도를 알아보고, 활성화를 위한 기반을 마련하고자 수행하였다. 총 195명을 대상으로 설문조사를 진행하였으며, 실내식물에 대한 인식과 ‘스마트 그린오피스’의 인식 및 선호도에 대하여 조사하였다. 설문조사 결과, 설문에 참여한 대부분의 응답자들은 실내식물을 선호하며, 접근성과 필요성에 대해 긍정적으로 답변한 것으로 조사되었다. 응답자들이 실내식물을 통해 ‘공기 질 향상(52.0%)’ 및 ‘스트레스 해소(34.0%)’등 긍정적인 효과를 경험한 적이 있기 때문에 실내식물의 기능을 인지하고 필요성을 크게 인식하는 것으로 판단된다. ‘스마트 그린오피스’의 인지도에 대해 조사한 결과, 알거나 들어본 경험이 있는 사람은 15.4%로 적었으나 개념을 설명한 후, 필요성을 느끼고 설치를 원하는 응답자들 은 90% 이상으로 조사되었다. 응답자들은 ‘공기정화(83.6%)’ 기능 때문에 ‘스마트 그린오피스’ 도입 이 필요하다고 하였으며, 오피스 내 도입 시‘공기정화 식물(70.8%)’로 이루어진 벽면형태(41.0%)’를 선호하는 것으로 조사되었다. 또한, ‘비용 (45.6%)’과 ‘유지 관리(28.2%)’등의 문제가 ‘스마트 그린오피스’ 도입 시 고려하는 점으로 조사되었으며, 설치 시 비용은 ‘200만원 이하(56.4%)’와 ‘200~450 만원(38.5%)’을 선호하는 것으로 조사되었다. ‘스마트 그린오피스’의 인식 및 선호도와 더불어 비활성화에 대한 원인을 조사한 결과, ‘낮은 인지도’ 및 ‘스 마트 그린오피스’의 개념을 인지하지 못한 ‘불필요’가 각각 25.1%와 42.6%로 조사되었으며, ‘적극적인 마케팅 및 홍보 (46.2%)와 ‘실내식물에 대한 관심도 향상(38.0%)’이 이를 해결하기 위한 방법이라고 응답하였다. 따라서, ‘스마트 그린오피스’의 올바른 의미를 알리고, 공기정화 식물로 이루어진 벽면형태의 디자인 개발 및 모델 제시와 같이 ‘스마트 그린오피스’가 활성화될 수 있도록 홍보를 통한 다양한 정보 제공 등 여러 방안 모색이 필요할 것으로 판단된다. This aim of this study was to investigate public perceptions of and preferences for a “smart green office” to help establish the program. Our survey comprised 195 respondents who answered questions about their perception of indoor plants and their awareness of and preference for a smart green office. Survey results indicated that most people preferred indoor plants and had positive attitudes about the accessibility and need for a smart green office. Because the respondents had experienced the positive effects of indoor plants, such as improved air quality (52.0%) and stress relief (34.0%), they recognized the function of indoor plants and the need for a smart green office program. According to a public survey on the awareness of this type of program, 15.4% of the respondents either knew about or had heard of the concept; however, after explaining the program in more detail, the >90% of the respondents recognized the need for a smart green office and wanted to implement the program. The main reason given to implement the program was to help purify the air (83.6%), and the respondents preferred the wall garden style (41.0%) and air-purification plants (70.8%). In addition, when asked about the cost (45.6%) and maintenance (28.2%) of implementing the program, and costs were expected to be < 2 million KRW (56.4%) and 200 - 450 million KRW (38.5%), respectively. Additionally, when asked about the reasons for deactivation of the program, respondents answered “low awareness” (25.1%) and “low needs” (42.6%). They also suggested that those promoting the concept must “strengthen marketing” (46.2%) and “increase indoor plant interest” (38.0%) to enhance the program; therefore, various methods, such as better publicizing a smart green office program and providing a wall garden design using air-purification plants are needed to better inform the public and to encourage acceptance of the program.

      • Generation of retinal pigment epithelial cells from human embryonic stem cell-derived spherical neural masses

        <P>Dysfunction and loss of retinal pigment epithelium (RPE) are major pathologic changes observed in various retinal degenerative diseases such as aged-related macular degeneration. RPE generated from human pluripotent stem cells can be a good candidate for RPE replacement therapy. Here, we show the differentiation of human embryonic stem cells (hESCs) toward RPE with the generation of spherical neural masses (SNMs), which are pure masses of hESCs-derived neural precursors. During the early passaging of SNMs, cystic structures arising from opened neural tube-like structures showed pigmented epithelial morphology. These pigmented cells were differentiated into functional RPE by neuroectodermal induction and mechanical purification. Most of the differentiated cells showed typical RPE morphologies, such as a polygonal-shaped epithelial monolayer, and transmission electron microscopy revealed apical microvilli, pigment granules, and tight junctions. These cells also expressed molecular markers of RPE, including Mitf, ZO-1, RPE65, CRALBP, and bestrophin. The generated RPE also showed phagocytosis of isolated bovine photoreceptor outer segment and secreting pigment epithelium-derived factor and vascular endothelial growth factor. Functional RPE could be generated from SNM in our method. Because SNMs have several advantages, including the capability of expansion for long periods without loss of differentiation capability, easy storage and thawing, and no need for feeder cells, our method for RPE differentiation may be used as an efficient strategy for generating functional RPE cells for retinal regeneration therapy.</P>

      • FCP 3 : Platelet rich plasma induced hair growth in alopecia patients partially through dermal papilla cell proliferation, BCl-2 and KGF up-regulation

        Alopecia is a common disorder with significant negative impact on their social and psychological well-being. There are many modalities to treat various alopecia, such as oral finasteride, topical minoxidil, triamcinolone intralesional injection, excimer laser and so on. However, the treatment results of these treatment are not always satisfactory. In vitro study, PRP treated dermal papilla cells showed the increased cell proliferation rates, BCl-2 and KGF up-regulation. We tried to evaluate efficacy and safety of sequential treatment, Platelet-rich plasma(PRP) injection, followed by mesotherapy in various alopecia. Included in this study were males and females between the ages of 20 and 60 years, in good health. Two independent investigators evaluated clinical improvement using a quartile grading scale by comparing digital photographs taken prior to treatment and at 3 months after the last treatment. The result was a relatively clinical improvement in hair count and hair thickness demonstrated in all the patients. There were no significant side effects except mild pain, transient bleeding at injection or microneedling site. Taken together, PRP induced hair growth in alopecia patients partially through dermal papilla cell proliferation, BCl-2 and KGF up-regulation.

        Comparison of Adults with Attention-Deficit Hyperactivity Disorder Depending on the Age of Being Diagnosed in Childhood and Adulthood: Based on Retrospective Review in One University Hospital

        Objectives: The study aimed to identify the characteristics of attention-deficit hyperactivity disorder (ADHD) that was not diagnosed in childhood or adolescence, but only in adulthood. Methods: The characteristics of patients diagnosed with ADHD in adulthood were compared with those of patients diagnosed in childhood were assessed via a retrospective review of the medical records at one university hospital from 2005 to 2013. If the age at which they were confirmed as having ADHD was less than 19 years old, they were grouped as childhood-diagnosed group (CD); if they were 19 years old or more, they were grouped as adulthood-diagnosed group (AD). Results: The CD and AD included 50 (46.3%) and 58 (53.7%) patients, respectively. Inattention was the most common symptom in both groups. Behavioral and emotional problems were the second most frequent symptoms in the CD and AD, respectively. The intelligent quotient was significantly higher in the AD than in the CD. The most common comorbidity was depression in the CD and personality disorder in the AD. The most common reason for visiting the hospital was referral by acquaintances in the CD and media coverage in the AD. Conclusion: Clinicians should put ADHD on the index of suspicion when they examine adults with various psychiatric symptoms, because the diagnosis of ADHD might have been missed in childhood and the symptoms of ADHD might have changed as they grew up.
