The antigenic potential of CFA-001, cefazolin, a cephalosporin derivative produced by an enzymatic semisynthesis, was determined in Hartley guinea pigs. A battery of tests employed consisted of active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA), and indirect hemagglutination test (IHA). The results were as follows: 1) In ASA, no signs attributable to anaphylaxis was observed in guinea pigs sensitized with CFA-001, whereas OVA-sensitized animals induced severe anaphylactic symptoms; 2) guinea pigs did not produce antibodies against CFA-001 when sensitized with or without Freund's complete adjuvant (FCA) in homologous PCA tests. Meanwhile, antibodies against ovalbumin (OVA) were clearly detected; 3) No CFA-001-specific hemagglutination was observed in the IHA using sera obtained from CFA-001-sensitized guinea pigs. These results suggest that CFA-001 has no antigenicity potential in guinea pigs.
Analysis of high-fat diet-induced inflammatory responses in Rhbdf2 knockout mice

This study investigated the characteristics of obesity induced by a high-fat diet (HD) over 13 weeks in Rhbdf2 gene knockout (KO) mice. Forty 7-week-old Rhbdf2 wild and KO mice were used and the mice were divided into 4 groups: Wild-ND (n=10, Rhbdf2 wild mice, normal diet (ND)), Wild-HD (n=10, Rhbdf2 wild mice, HD), KO-ND (n=10, Rhbdf2 KO mice, ND) and KO-HD (n=10, Rhbdf2 KO mice, HD). The relative epididymal fat weight in KO-HD was significantly increased compared with that in KO-ND (P<0.01). The relative liver and spleen weights in KO-HD were decreased compared with those in Wild-HD (p < 0.05) and KO-ND (p < 0.01). The mRNA expression of SOD1 in KO-ND was significantly reduced compared with that in Wild-ND (p < 0.05). In Wild-ND and HD, the mRNA expressions of TNF-α and IL-6 in epididymal fat were significantly increased compared with those in KO-ND and HD (p < 0.01). A significant increase of TNF- α and IL-6 mRNA expression was observed in KO-HD compared with KO-ND (p < 0.01). These results indicated that Rhbdf2 genes may regulate high fat diet-induced obesity damage by anti-inflammatory and anti-oxidative roles in fat tissue of mice.

Objective The purpose of this study was to observe the natural course of remained untreated spondylolysis adjacent to previous fusion segments in patients with multi-level lysis and its clinical outcome. Methods Nineteen patients who underwent selective fusion of multi-level spondylolysis (MLS) at a single institute were enrolled. As a matched cohort for comparison, 19 patients who had single-level spondylolysis (SLS) and undergone single-level fusion with similar demographics and preoperative radiologic measurements as the MLS group were included. We evaluated the preoperative, postoperative, and last follow-up angular displacement and sagittal translation on dynamogram radiographs, and axial and radial pain using the visual analogue scale in both groups. We then compared the increment in radiological instability and clinical outcome between the MLS and SLS groups. Results There were no significant differences in patient demographics and preoperative radiological measurements, including disc degeneration, facet degeneration, lumbar lordosis, pelvic incidence, and sacral table angle between both groups. Both groups showed an increase in the average angular displacement and slippage during the final follow-up as compared to preoperative findings, but no significant difference was noticed between them. Both the MLS and SLS groups showed improvement in lower back pain and leg pain from before surgery, but with no statistical significance. Conclusion Selective fusion in patients with multiple spondylolysis can be an alternative surgical option without increasing the risk of adjacent segment degeneration under strict narrow indications. However, a thorough preoperative evaluation is needed to prevent early surgical failure.

Background: Many chronic obstructive pulmonary disease (COPD) patients receiving monotherapy continue to experience symptoms, exacerbations and poor quality of life. This study aimed to assess the efficacy and safety of direct switch from once-daily tiotropium (TIO) 18 μg to indacaterol/glycopyrronium (IND/GLY) 110/50 μg once daily in COPD patients in Korea. Methods: This was a randomized, open-label, parallel group, 12-week trial in mild-to-moderate COPD patients who received TIO 18 μg once daily for ≥12 weeks prior to study initiation. Patients aged ≥40 years, with predicted post-bronchodilator forced expiratory volume in 1 second (FEV<sub>1</sub>) ≥50%, post-bronchodilator FEV<sub>1</sub>/forced vital capacity <0.7 and smoking history of ≥10 pack-years were included. Eligible patients were randomized in a 1:1 ratio to either IND/GLY or TIO. The primary objective was to demonstrate superiority of IND/GLY over TIO in pre-dose trough FEV1 at week 12. Secondary endpoints included transition dyspnea index (TDI) focal score, COPD assessment test (CAT) total score, and rescue medication use following the 12-week treatment, and safety assessment. Results: Of the 442 patients screened, 379 were randomized and 347 completed the study. IND/GLY demonstrated superiority in pre-dose trough FEV1 versus TIO at week 12 (least squares mean treatment difference [Δ], 50 mL; p=0.013). Also, numerical improvements were observed with IND/GLY in the TDI focal score (Δ, 0.31), CAT total score (Δ, -0.81), and rescue medication use (Δ, -0.09 puffs/day). Both treatments were well tolerated by patients. Conclusion: A direct switch from TIO to IND/GLY provided improvements in lung function and other patient-reported outcomes with an acceptable safety profile in patients with mild-to-moderate airflow limitation.

The objective of this study was to evaluate the pre-heating treatment effects on the antioxidant properties of ethanolic garlic and onion extracts. Garlic and onion with or without heating (100oC, 30 min) were extracted with ethanol, and the total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, iron chelating ability, reducing power, and antioxidant activity in a linoleic acid emulsion system were evaluated. Garlic (41%) had a higher drying yield than onion (11%). Regardless of pre-heating, ethanol extracts of onion resulted in an approximately 25-fold higher yield than those of garlic. Thermal treatment before extraction decreased the levels of ethanol-soluble phenolics for both garlic and onion. Regardless of pre-heating, the radical scavenging abilities of ethanol extracts from garlic were greater than the ethanol extracts from onion. The iron chelating abilities of ethanol extracts from fresh and heated garlic were 85 and 81% at 10 mg/mL, respectively, whereas those of onion extracts were 10 and 9% at the same concentration, respectively. However, no differences in reducing power between garlic and onion extracts were observed. Both garlic and onion inhibited the formation of hydroperoxide in linoleic acid emulsion systems when ethanol was used as a solvent. Overall, garlic extracts had greater antioxidant activity than onion extracts, and the antioxidant activity of garlic and onion extracts were not significantly affected by thermal treatment.
Microbiological survey of Korean animal facilities from 2014 to 2019

To survey the mouse microbiological contamination rate in Korean animal facilities, we tested for common contaminants in these facilities using serological, culture, and parasitological methods. A total of 21,292 experimentally housed mice from 206 animal facilities, including hospitals, universities, companies and research institutes, were tested over a 6-year period from 2014 to 2019. The most commonly found contaminants were various species of non-pathogenic protozoa (47.2%). The most common pathogenic bacteria were Staphylococcus aureus (21.2%), Pasteurella pneumotropica (12.5%) and Pseudomonas aeruginosa (5.8%). Mouse hepatitis virus (6.1%) was detected, but no other viral or bacterial pathogens were found. These results establish that the main pathogens that currently contaminate animal facilities in Korea are opportunistic pathogens, whereas important such as those in categories B or C, were decreased.
