      • SIGN-R1, a C-type lectin, enhances apoptotic cell clearance through the complement deposition pathway by interacting with C1q in the spleen

        Complements, such as C1q and C3, and macrophages in the splenic marginal zone (MZMs) play pivotal roles in the efficient uptake and processing of circulating apoptotic cells. SIGN-R1, a C-type lectin that is highly expressed in a subpopulation of MZMs, regulates the complement fixation pathway by interacting with C1q, to fight blood-borne Streptococcus pneumoniae. Therefore, we examined whether the SIGN-R1-mediated classical complement pathway plays a role in apoptotic cell clearance and immune tolerance. SIGN-R1 first-bound apoptotic cells and this binding was significantly enhanced in the presence of C1q. SIGN-R1–C1q complex then immediately mediated C3 deposition on circulating apoptotic cells in the MZ, leading to the efficient clearance of them. SIGN-R1-mediated C3 deposition was completely abolished in the spleen of SIGN-R1 knockout (KO) mice. Given that SIGN-R1 is not expressed in the liver, we were struck by the finding that C3-deposited apoptotic cells were still found in the liver of wild-type mice, and dramatically reduced in the SIGN-R1 KO liver. In particular, SIGN-R1 deficiency caused delayed clearance of apoptotic cells and aberrant secretion of cytokines, such as TNF-α, IL-6, and TGF-β in the spleen as well as in the liver. In addition, anti-double- and single-stranded DNA antibody level was significantly increased in SIGN-R1-depleted mice compared with control mice. These findings suggest a novel mechanism of apoptotic cell clearance which is initiated by SIGN-R1 in the MZ and identify an integrated role of SIGN-R1 in the systemic clearance of apoptotic cells, linking the recognition of apoptotic cells, the opsonization of complements, and the induction of immune tolerance.

        Comparative Studies on Metabolic Rate and Calpain/Calpastatin Activity between Hanwoo and Holstein Beef

        The objectives of this study were to examine the effect of early short-term temperature conditioning on metabolic rate and calpain/calpastatin system and to compare variations in metabolic rate and calpain/calpastatin system between Hanwoo and Holstein beef. Longissimus thoracis et lumborum of the right carcass from 3 Hanwoo and 3 Holstein bulls were removed within 30 min of exsanguinations, cut into three pieces, and then temperature conditioned until 3 h postmortem (PM) at 2, 16, and $30^{\circ}C$, respectively. Rigor values (R-values; $R_248$, $R_250$, and $R_258$), pH, muscle temperature, glycogen content, $\mu$- and m-calpain activities, and calpastatin acitivity were measured at 1, 3, 9, and 24 h PM, respectively. Hanwoo beef had higher muscle temperature, faster metabolic rate at early PM stage in R-values, and lower $\mu$-calpain activity than Holstein beef (p<0.05). The $30^{\circ}C$ treatment maintained muscle temperature of $30^{\circ}C$ until 3 h PM and resulted in faster pH decline at 3 and 9 h PM (p<0.05) than other treatments. The $16^{\circ}C$ had higher (p<0.05) muscle temperature at 3 h PM than the $2^{\circ}C$, but no difference in all other traits was observed between the $2^{\circ}C$ and the $16^{\circ}C$. Early shortterm temperature treatment used in this study was not sufficient to effectively activate calpain/calpastatin system. Correlations among all traits except m-calpain and muscle temperature were generally high (r>0.60; p<0.001). Among R-values, $R_258$ had higher correlations with other metabolic traits than those of $R_248$ and $R_250$. These data suggest that early PM metabolic rate, $\mu$-calpain activity, and calpastatin activity may be closely related to each other. Variations in metabolic rate and $\mu$-calpain activity at early PM stage between Hanwoo and Holstein beef may imply variations in meat quality between both breeds.

      • Interleukin-18, transforming growth factor-β, and vascular endothelial growth factor gene polymorphisms and susceptibility to primary glomerulonephritis

        <P>Several studies have showed an association of gene polymorphisms with the development of glomerulonephritis (GN). We investigated the effects of gene polymorphisms on the development of GN by analyzing polymorphisms in the interleukin (IL)-18, transforming growth factor (TGF)-β, and vascular endothelial growth factor (VEGF) genes in Korean patients with primary GN. The study included 146 normal subjects (controls) and 100 patients diagnosed with primary GN by kidney biopsy. The gene polymorphisms A-607C and G-137C in <I>IL-18</I>, C-509T and T869C in <I>TGF-</I>β<I>1</I>, and C-2578A and C405G in <I>VEGF</I> were investigated in DNA extracted from peripheral blood. Significant differences were observed between the GN and control groups in the genotype and allele frequencies of A-607C <I>IL-18</I> and C405G <I>VEGF</I>. The frequencies of the <I>IL-18</I>−607CC genotype [<I>P</I> = 0.001, odds ratio (OR) = 2.473] and the <I>VEGF</I> 405GG genotype (<I>P</I> = 0.001, OR = 2.473) were significantly increased in the GN group. The combination of <I>IL-18</I>−607CC+ and <I>VEGF</I> 405GG+ genotypes had a higher risk for developing GN in comparison with the combination of <I>IL-18</I>−607CC− and <I>VEGF</I> 405GG− genotypes (<I>P</I> < 0.001, OR = 8.642). In the haplotype analysis of the <I>IL-18</I> gene, the CG haplotype was significantly more frequent in the GN group than the control group (61.5% <I>vs</I> 46.9%, <I>P</I> = 0.002). These results show that the −607CC genotype of the <I>IL-18</I> gene and the 405GG genotype of the <I>VEGF</I> gene are associated with susceptibility to and the development of primary GN.</P>

        OGLE-2016-BLG-1190Lb: The First<i>Spitzer</i>Bulge Planet Lies Near the Planet/Brown-dwarf Boundary

        <P>We report the discovery of OGLE-2016-BLG-1190Lb, which is likely to be the first Spitzer microlensing planet in the Galactic bulge/ bar, an assignation that can be confirmed by two epochs of high-resolution imaging of the combined source-lens baseline object. The planet's mass, M-p = 13.4 +/- 0.9 M-J, places it right at the deuteriumburning limit, i. e., the conventional boundary between 'planets' and 'brown dwarfs.' Its existence raises the question of whether such objects are really ' planets' (formed within the disks of their hosts) or 'failed stars' (lowmass objects formed by gas fragmentation). This question may ultimately be addressed by comparing disk and bulge/bar planets, which is a goal of the Spitzer microlens program. The host is a G dwarf, M-host = 0.89. +/- 0.07 M-circle dot, and the planet has a semimajor axis a similar to 2.0 au. We use Kepler K2 Campaign 9 microlensing data to break the lens-mass degeneracy that generically impacts parallax solutions from Earth-Spitzer observations alone, which is the first successful application of this approach. The microlensing data, derived primarily from near-continuous, ultradense survey observations from OGLE, MOA, and three KMTNet telescopes, contain more orbital information than for any previous microlensing planet, but not quite enough to accurately specify the full orbit. However, these data do permit the first rigorous test of microlensing orbital-motion measurements, which are typically derived from data taken over < 1% of an orbital period.</P>

        OGLE-2016-BLG-0613LABb: A Microlensing Planet in a Binary System

        <P>We present the analysis of OGLE-2016-BLG-0613, for which the lensing light curve appears to be that of a typical binary-lens event with two caustic spikes but with a discontinuous feature on the trough between the spikes. We find that the discontinuous feature was produced by a planetary companion to the binary lens. We find four degenerate triplelens solution classes, each composed of a pair of solutions according to the well-known wide/close planetary degeneracy. One of these solution classes is excluded due to its relatively poor fit. For the remaining three pairs of solutions, the most-likely primary mass is about M-1 similar to 0.7M(circle dot) , while the planet is a super Jupiter. In all cases, the system lies in the Galactic disk, about halfway toward the Galactic bulge. However, in one of these three solution classes, the secondary of the binary system is a low- mass brown dwarf, with relative mass ratios (1: 0.03: 0.003), while in the two others the masses of the binary components are comparable. These two possibilities can be distinguished in about 2024 when the measured lens-source relative proper motion will permit separate resolution of the lens and source.</P>

        OGLE-2016-BLG-0263Lb: Microlensing Detection of a Very Low-mass Binary Companion through a Repeating Event Channel

        <P>We report the discovery of a planet-mass companion to the microlens OGLE-2016-BLG-0263L. Unlike most low-mass companions that were detected through perturbations to the smooth and symmetric light curves produced by the primary, the companion was discovered through the channel of a repeating event, in which the companion itself produced its own single-mass light curve after the event produced by the primary had ended. Thanks to the continuous coverage of the second peak by high-cadence surveys, the possibility of the repeating nature due to source binarity is excluded with a 96% confidence level. The mass of the companion estimated by a Bayesian analysis is Mp = 4.1(-2.5)(+6.5) M-j. The projected primary-companion separation is a(perpendicular to) = 6.5(-1.9)(+1.3) au. The ratio of the separation to the snow-line distance of a(perpendicular to/)a(s1) similar to 15.4 corresponds to the region beyond Neptune, the outermost planet of the solar system. We discuss the importance of high-cadence surveys in expanding the range of microlensing detections of low-mass companions and future space-based microlensing surveys.</P>

      • OGLE-2016-BLG-1469L: Microlensing Binary Composed of Brown Dwarfs

        <P>We report the discovery of a binary composed of two brown dwarfs, based on the analysis of the microlensing event OGLE-2016-BLG-1469. Thanks to the detection of both finite-source and microlens-parallax effects, we are able to measure both the masses M-1 similar to 0.05M(circle dot) and. M-2 similar to 0.01M(circle dot), and the. distance D-L similar to 4.5 kpc, as well as the projected separation a(perpendicular to) similar to 0.33 au. This is the third brown-dwarf binary detected using the microlensing method, demonstrating the usefulness of microlensing in detecting field brown-dwarf binaries with separations of less than 1 au.</P>

      • In vitro and in vivo antifungal activities of decursin and decursinol angelate isolated from Angelica gigas against Magnaporthe oryzae, the causal agent of rice blast

        Blast is considered the most important fungal disease of rice due to its wide distribution and destructiveness under favorable conditions. Development of new effective and environmentally benign agents against the causal pathogen, Magnaporthe oryzae, is of great interest. In the course of a search for natural antifungal compounds in medicinal plants, we found that the methanol extract of Angelica gigas roots showed a potent control efficacy against rice blast caused by M. oryzae. We isolated antifungal coumarins from the extract, and they were identified as decursin and decursinol angelate. Antifungal activities of these compounds, along with kasugamycin, were tested on M. oryzae in vivo and in vitro. In an in vivo assay, the three compounds effectively suppressed the development of rice blast at concentrations more than 100μg/mL. Coumarins showed relatively weak inhibitory effect on fungal mycelial growth when compared to kasugamycin. However, they strongly inhibited M. oryzae spore germination, which was not observed in kasugamycin treatments. This is the first report demonstrating that decursinol angelate can provide control against rice blast and that the two coumarins inhibit M. oryzae spore germination. In addition, the wettable powder formulation of the crude extract of A. gigas prohibited the development of blast symptoms on rice plants more effectively than liquid concentrate formulation of kasugamin, a commercial fungicide. Based on our study, we propose that coumarin compounds as well as the A. gigas root crude extract can be used as natural, benign fungicides for controlling rice blast.

        HgTe/Cdte superlattices grown on CdZnTe(211)B by MBE

        Hg-Te-CdTe superlattices have received much interests over the last several years as a potential material for its applications for detecting devices and optoelectronics. We have grown the HgTe-CdTe superlattice using MBE. in our lab. We have carried out DCRC spectroscopy after growth of HgTe-CdTe superlattice with varying the superlattice periods and controlling the barrier thickness and we have that the presence of the main peak and the satellite peaks. We obtained 20 arcsec of FWHM over 100 periods of superlattice. We also note that high peak intensity shows the high quality of the sample and each layer of superlattice has abrupt interfaces. The angular separation between the main peak(m=0) and the first satellite peak(m=$\pm$1) is increased when the barrier layer thickness in superlatice layers are decreased. The separation between the first setellite peak(m=$\pm$1) and the second satellite peak(m$\pm$2) is increased similarly. The number of the satellite peak is a qualitative measure of the interfacial abruptness of the superlattice.
