<P><B>Abstract</B></P> <P>Peptide nucleic acids (PNAs), artificially synthesized DNA analogues, hybridize strongly with DNA and are useful for fluorescence melting curve analyses (FMCA) based on the thermal denaturation of the probe-target duplex. In this study, we developed a PNA-based one-step real-time RT-PCR assay for the differential and qualitative detection of the porcine reproductive and respiratory syndrome virus genotypes PRRSV1 and PRRSV2. The specificity of the assay was analyzed in silico using previously reported primers and probes and was subsequently verified using Korean PRRSV panels and clinical samples. Seven clinical samples showing low curves with high Ct values were confirmed as negative by FMCA. The sensitivities of one-step real-time PCR for PRRSV1 and PRRSV2 were 15 and 11 copies, respectively, and the results were in 100% agreement with those of conventional RT-PCR combined with nested PCR using clinical samples. Therefore, the assay is highly specific for the detection of current PRRSV1 and PRRSV2 without non-specific amplification by FMCA.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We developed a peptide nucleic acid-based FMCA for PRRSV detection. </LI> <LI> The assay was applied to PRRSV, which has high sequence diversity and divergence. </LI> <LI> The assay was highly specific for the detection of currently circulating genotypes. </LI> <LI> The one-step real-time RT-PCR method for PRRSV1 and PRRSV2 exhibited high sensitivity. </LI> </UL> </P>
Trajectory Generation for a Car-Like Mobile Robot using Closed-Loop Prediction

This paper proposes a trajectory generation method using the closed-loop prediction to follow reference path, which are provided by a global path planner. Using the proposed method, smooth trajectories can be generated by considering the position of a mobile robot and its kinematic properties. The feasibility of the proposed method was verified by simulations in various situations.
Polynomial Curve-based Longitudinal Motion Planning for Safe Deceleration

This paper proposes a longitudinal motion planning method to reduce the velocity of an unmanned ground vehicle safely when it encounters stopped vehicles or obstacles on the road. The proposed method can generate the smooth velocity and acceleration profiles using the quintic polynomial curve. The optimal quintic polynomial curve can be determined by a numerical optimization approach. The performance of the proposed method is verified by simulations.
Sampling-Based Retraction Method for Improving the Quality of Mobile Robot Path Planning

This paper presents a method for improving the quality of the initial path produced by the probabilistic roadmap (PRM)-based mobile robot path planner. The sampling-based retraction method modifies the initial path to achieve approximate maximum safety by removing unsafe and redundant sections. The updated directions and distances of the waypoints on the initial path are determined by approximately modeling clearances around the initial paths using random samples. The proposed me-thod can control the update speed to induce smooth convergence. The performance of the proposed method was verified by simulation.
Robust range-only beacon mapping in multipath environments

This study proposes a robust range-only beacon mapping method for registering the locations of range-only beacons automatically. The proposed method deals with the multipath propagation of signals from range-only beacons using the range-only measurement association (RoMA) and an unscented Kalman filter (UKF). The RoMA initially predicts the candidate positions of a range-only beacon. The location of the range-only beacon is then updated using the UKF. With the proposed method, the locations of range-only beacons are accurately estimated in a multipath environment. The proposed method also provides the location uncertainty of each range-only beacon. Simulation results using the model for multipath propagation and experimental results in a real indoor environment verify the performance of the proposed method.
Broadband optical filter based on high purity isotope gas for the carbon-13 breath test

In this study, we developed a novel isotope gas flter for a mid-infrared (IR) LED-based NDIR spectroscopy system. Our novel isotope gas flter contains high-purity CO2 gas (99.99% content), and the intrinsic absorption characteristic of contained gas works as an optical stopband flter. Our flter achieves a 25 dB attenuation performance in the region of 4190–4350 nm, and is a core component in distinguishing the signal from diferent carbon isotopes such as 12CO2 and 13CO2 for an NDIR carbon-13 (C-13) based breath test. Compared with dielectric flters normally used for C-13 breath tests, our flter can provide more useful and consistent performance than dielectric flters in the mid-IR region.
