<B>ABSTRACT</B><P>The adaptive response to hydrogen peroxide (H2O2) in <I>Pseudomonas aeruginosa</I> involves the major catalase, KatA, and OxyR. However, neither the molecular basis nor the relationship between the aforementioned proteins has been established. Here, we demonstrate that the transcriptional activation of the <I>katA</I> promoter (<I>katAp</I>) in response to H2O2 was abrogated in the <I>P. aeruginosa</I> PA14 <I>oxyR</I> null mutant. Promoter deletion analyses revealed that H2O2-mediated induction was dependent on a region of DNA −76 to −36 upstream of the H2O2-responsive transcriptional start site. This region harbored the potential operator sites (OxyR-responsive element [ORE]) of the <I>Escherichia coli</I> OxyR binding consensus. Deletion of the entire ORE not only abolished H2O2-mediated induction but also elevated the basal transcription, suggesting the involvement of OxyR and the ORE in both transcriptional activation and repression. OxyR bound to the ORE both <I>in vivo</I> and <I>in vitro</I>, demonstrating that OxyR directly regulates the <I>katAp</I>. Three distinct mobility species of oxidized OxyR were observed in response to 1 mM H2O2, as assessed by free thiol trapping using 4-acetamido-4′-maleimidylstilbene-2,2′-disulfonic acid. These oxidized species were not observed for the double mutants with mutations in the conserved cysteine (Cys) residues (C199 and C208). The uninduced transcription of <I>katAp</I> was elevated in an <I>oxyR</I> mutant with a mutation of Cys to serine at 199 (C199S) and even higher in the <I>oxyR</I> mutant with a mutation of Cys to alanine at 199 (C199A) but not in <I>oxyR</I> mutants with mutations in C208 (C208S and C208A). In both the C199S and the C208S mutant, however, <I>katAp</I> transcription was still induced by H2O2 treatment, unlike in the <I>oxyR</I> null mutant and the C199A mutant. The double mutants with mutations in both Cys residues (C199S C208S and C199A C208S) did not differ from the C199A mutant. Taken together, our results suggest that <I>P. aeruginosa</I> OxyR is a bona fide transcriptional regulator of the <I>katA</I> gene, sensing H2O2 based on the conserved Cys residues, involving more than one oxidation as well as activation state <I>in vivo</I>.</P>

Objective: To compare short-term treatment outcomes at hospital discharge and hospital length of stay (LOS) in patients with spontaneous intracerebral hemorrhage (sICH) before and after introduction of resident physician work time limit (WTL). Methods: We retrospectively reviewed consecutive patients treated for sICH at our institution between 2016 and 2019. Then we dichotomized these patients into two groups, pre-WTL and post-WTL. We analyzed demographic elements and clinical features, and hospital length of stay (LOS). We evaluated short-term outcome using modified Rankin scale score at hospital discharge and then divided it into “good” and “poor” outcome groups. We subsequently, compared short-term treatment outcome and hospital LOS between the pre-WTL and post-WTL groups. Results: Out of 779 patients, 420 patients (53.9%) were included in the pre-WTL group, and 359 (46.1%) in post-WTL. The mortality rate in sICH patients was higher in the post-WTL group (pre-WTL; 13.6% vs. post-WTL; 17.3%), but there was no statistically significant difference in short-term outcome including mortality (p=0.332) between the groups. The LOS also, was not significantly different between the two groups (pre-WTL; 19.0 days vs. post-WTL; 20.2 days) (p=0.341). The initial Glasgow Coma Scale score, personal stroke history, and mean age were the only independent outcome predicting factors for patients with sICH. Conclusions: Some neurosurgeons may expect poorer outcome for sICH after implementation of the WTL of the K-MHW for resident physician however, enforcement of the WTL did not significantly influence the short-term outcome and hospital LOS for sICH in our hospital. Further well-designed multi-institutional prospective studies on the effects of WTL in sICH patient outcome, are anticipated.

Single layer Sn doped In2O3(ITO) films and ITO 50 nm/Au 10 nm/ITO 40 nm (IAI) multilayer films were prepared with electron beam assisted magnetron sputtering on glass substrates. The effects of the Au interlayer, post-deposition atmosphere annealing and intense electron irradiation on the methanol gas sensitivity were investigated at room temperature. As deposited ITO films did not show any diffraction peaks in the XRD pattern, while the IAI films showed the diffraction peak for In2O3 (400). In this study, the gas sensitivity of ITO and IAI films increased proportionally with the methanol vapor concentration and an intense electron beam irradiated IAI film shows the higher sensitivity than the others film. From the XRD pattern, it is supposed that increased crystallization promotes the gas sensitivity. This approach is promising in gaining improvement in the performance of IAI gas sensors used for the detection of methanol vapor at room temperature.

이 연구에서는 영재들의 심리적인 문제점을 해소할 수 있는 방안의 하나로 반려견을 활용하여 인간의 성격과 반려견종을 매칭 할 수 있는 방법을 고안하고자 하였다. NEO 성격검사의 5가지 요소들을 반려견 양육과 특성에 맞추어 적용하였으며, 각각의 요소별로 반려견의 품종을 분류하여 반려견 양육 희망자의 성격에 부합하는 반려견품종 매칭 가이드라인을 작성하였다. 반려견 양육 희망자의 성격과 부합되는 반려견 품종의 매칭을 위해 인간의 성격 요인을 분석하는 기존의 방법들을 검토하였으며, 반려견 양육요소에 적용이 가장 적절한 5요인(신경증, 외향성, 개방성, 친화성, 성실성) 성격검사를 반려견 양육에 부합되도록 적용하였다. (사)한국애견협회의 등록 견종 상위 50위를 선별하여 각 견종의 고유 성격과 양육 특징을 인간의 5가지 성격 요소별로 분류하였다. 이러한 연구는 선행되어지지 않은 최초의 시도이기에 신뢰도의 확보를 위해서 지속적인 보완작업과 수렴타당도, 판별타당도 등의 타당화 작업이 수행될 필요가 있으며, 본 연구에서 개발된 결과의 타당성 검증을 위한 후속 연구를 수행하여, 신뢰성과 편리성이 있는 인간 성격에 부합하는 반려견 선택 도구를 제시하고자 한다. In this study, five each elements of the NEO personality test were applied according to the companion dog rearing and characteristics, and guideline to choice breed of companion dog was presented based on the personality of the dog rearing applicant. The literature review presented the issue of abandonment and disruption of companion animal, and investigated policies and systems for suppression of abandonment and disruption rates of companion animals in abroad. Methods of analyzing human personality were reviewed and five factors (Neuroticism, Extraversion, Openness to Experience, Agreeableness, Conscientiousness) were applied for matching prospective adopter and specific dog breed which following their characters. The top 50 registered dogs of the Korea Pet Association were selected and the unique personality and parenting characteristics of each dog were classified based on five characteristic elements of humans. Since these studies are the first attempt not to be preceded, continuous complementary and convergent feasibility and validation work need to be carried out to ensure reliability, and follow-up studies to validate reliable dog breed secection system to character of prospective adoptors.
Quality assessment by using Rhod-2 staining in porcine matured and fertilized oocytes

Mitochondrion is an organelle for regulating calcium (Ca2+) homeostasis. Mitochondrial Ca2+ plays important roles on oocyte maturation, fertilization and embryonic development for ATP production. Low quality oocytes have mitochondrial dysfunction, which lead to overloaded Ca2+ in mitochondria. Recently, Rhod-2 is well known as a mitochondrial derived Ca2+ indicator. However, the changes of Rhod-2 in matured or fertilized porcine oocytes have not been reported. Therefore, the aim of study was to identify the effects of mitochondrial Ca2+ using Rhod-2 on quality assessment of matured oocyte and zygotes in pigs. Thus, we classified two groups (group 1: G1, compact COCs and group 2: G2, uncompact COCs) according to differences of cumulus cells amount and cytoplasm morphology in germinal vesicle (GV) stage of porcine COCs. Therefore, we investigated number of Rhod-2 spots in matured and fertilized oocytes from G1 and G2 groups. The Rhod-2 spot numbers were separated into four parts; n<10, 10≤ n < 20, 20 ≤ n < 30, and 30 < n. The Rhod-2 spots number of G2 group had greater than G1 group in part of 20 ≤ n. Additionally, we investigate mean number of Rhod-2 spots from G1 and G2 groups in matured and fertilized oocytes. As a result, we confirmed that average number of Rhod-2 spots in G2 group increased than that of G2 group. Finally, we also measured the Rhod-2 intensity in matured and fertilized oocytes of G1 and G2 groups. Interestingly, the Rhod-2 intensity in G2 group was higher than that of G1 group. (oocyte: p < 0.001 and fertilized oocyte: p < 0.05). These results demonstrated that changes in Rhod-2 spots and intensity were increased in low quality of matured and fertilized oocytes. Therefore, our results suggest that the differences in mitochondrial calcium level are associated with morphological quality of porcine COCs.
