<P>A new self-immolative linker motif, Ortho Hydroxy-Protected Aryl Sulfate (OHPAS), was devised, and OHPAS-containing antibody drug conjugates (ADC) were tested in vitro and in vivo. Conveniently synthesized using Sulfur Fluorine Exchange (SuFEx) chemistry, it is based structurally on diaryl sulfate, with one aryl acting as a payload and the other as a self-immolative sulfate unit having a latent phenol function at the ortho position. The chemically stable OHPAS linker was stable in plasma samples from 5 different species, yet it can release the payload molecule smoothly upon chemical or biological triggering. The payload release proceeds via intramolecular cyclization, producing a cyclic sulfate coproduct that eventually hydrolyzes to a catechol monosulfate. A set of OHPAS-containing ADCs based on Trastuzumab were prepared with a drug to antibody ratio of ∼2, and were shown to be cytotoxic in 5 different cancer cell lines in vitro and dose-dependently inhibited tumor growth in a NCI-N87 mouse xenograft model. We conclude that OHPAS conjugates will be of considerable use for delivering phenol-containing payloads to tissues targeted for medical intervention.</P> [FIG OMISSION]</BR>

In this paper, the travel control of the spherical wheeled robot with a mecanum wheel is impelemented. Four typical wheels or three omni wheels are used to consist of the ball-bot. the slip is occured when the typical wheels is used to the ball-bot. In order to reduce these slip, the spherical wheeled robot with macanum wheels is proposed. Through some experiments, we find that the proposed spherical wheeled robot with a mecanum wheel is superior to the conventional spherical wheeled robot with typical wheels.

이 논문에서는 투자비용 감소를 위해 라즈베리파이와 아두이노로 구현한 IoT 플러그를 제안한다. 기존 연구에서는 노후화된 시설에 대해 큰 비용이 발생하며, 전력소모량을 측정하는데 오차가 있다. 노후 시설에 대한 투자비용을 줄이기 위해 가격이 저렴한 라즈베리파이와 아두이노를 이용하여 IoT 플러그를 구현한다. 라즈베리파이 상에 서버를 구현하고 사용자와 nodeMCU 사이의 통신을 담당한다. nodeMCU는 릴레이모듈과 스위치에 연결되어 자동/수동으로 전원을 제어할 수 있다.

Background/Aims: Pretreatment nutritional status is an important prognostic factor in patients treated with conventional cytotoxic chemotherapy. In the era of target therapies, its value is overlooked and has not been investigated. The aim of our study is to evaluate the value of nutritional status in targeted therapy. Methods: A total of 2012 patients with non-small cell lung cancer (NSCLC) were reviewed and 630 patients with activating epidermal growth factor receptor (EGFR) mutation treated with EGFR tyrosine kinase inhibitor (TKI) were enrolled for the final analysis. Anemia, body mass index (BMI), and prognostic nutritional index (PNI) were considered as nutritional factors. Hazard ratio (HR), progression-free survival (PFS) and overall survival (OS) for each group were calculated by Cox proportional analysis. In addition, scores were applied for each category and the sum of scores was used for survival analysis. Results: In univariable analysis, anemia (HR, 1.29; p = 0.015), BMI lower than 18.5 (HR, 1.98; p = 0.002), and PNI lower than 45 (HR, 1.57; p < 0.001) were poor prognostic factors for PFS. Among them, BMI and PNI were independent in multi-variable analysis. All of these were also significant prognostic values for OS. The higher the sum of scores, the poorer PFS and OS were observed. Conclusions: Pretreatment nutritional status is a prognostic marker in NSCLC patients treated with EGFR TKI. Hence, baseline nutritional status should be more carefully evaluated and adequate nutrition should be supplied to these patients.
Acoustofluidic Microbioreactor Using Surface Acoustic Wave-induced Acoustic Streaming Flow

Microbioreactors have been widely utilized as an alternative to conventional benchtop reactors, since the miniaturized platforms off er advantages including reduced sample volume and homogeneous microenvironments. Here, we proposed an acoustofl uidic microbioreactor based on surface acoustic wave (SAW)-induced acoustic streaming fl ow (ASF). The SAW- induced ASF, which originates from the wave attenuation in a fl uid, allows rapid mixing and heat transfer for enhanced mass and heat transfer within the sample fl uid. We conducted thorough numerical and experimental investigations on the acousto-hydrodynamics and heat transfer phenomena to fi nd an optimal frequency in the prescribed cylindrical microwell. We found that the homogenous chemical concentration and temperature distributions within the fl uid were rapidly achieved by the SAW-induced ASF in the proposed device. For proof-of-concept demonstration of practical applicability, we cultured Escherichia coli as a model cell using the proposed acoustofl uidic microbioreactor. From comparative evaluation with con- ventional platforms including a shaker incubator and a microplate shaker, we confi rmed that the bacteria growth rate was enhanced in the proposed acoustofl uidic microbioreactor due to the high homogeneity in the chemical concentration and temperature by the acoustic agitation, without any moving mechanical components. We expect that the proposed ASF-based microbioreactor can be broadly utilized for various biological applications that require homogeneous mixing and temperature gradient within a reaction medium.
