東國大學校 演習內容 劑四團地에서 三年前부터 밤나무 胴枯病이 發生하였다. 이로 인하여 밤나무에 치명적인 枯死現象이 나타나기 시작하였다. 本 硏究는 一次的으로 防除對策에 觀한 것으로 本 病原菌의 生理·生態的인 性質究明에 역점을 두었다. 究明結果를 要約하면 다음과 같다. 1) 分生胞子의 增殖은 6種의 培地中 Potato decoction이 가장 優秀한 것으로 밝혀졌다. 2) 本 菌은 2℃∼25℃ 範圍에서 增殖하고 最適溫度는 18℃∼25℃였다. 3) Potato decoction 培地 增殖曲線은 培養 2日째부터 上昇하여 10日 째에는 最高度에 달하였으며 그 以後 50日까지 增殖曲線은 거의 變化가 없었다. 4) 培地의 pH는 3.0∼8.0까지 增殖되었으나 最適 pH는 5.0∼6.0으로 나타났다. 5) chestnut leaves decoction에서의 Sucrose와 Glucose와의 關係에는 0∼10% 濃度에서 兩糖이 모두 增殖에 기여하는 傾向이 나타났으나 그중 Glucose가 增殖에 더 良好하였다. 6) 供試한 無機, 有機窒素化合物 모두가 分生胞子增殖에 도움을 주었고 特히 KNO_3와 CO(NH)_2가 良好하였다. 有機窒素源 中에서는 L-arginine, L-asparagin 順이었다. 7) 炭素源과 分生胞子의 增殖關係에는 公試한 7種의 炭素源中 Mannitol이 가장 良好한 增殖을 보였으며 그 다음으로 Glycerin, 가장 不良한 것은 Maltose 였다. To elucidate the physiological natures of Endothia parasitica conidia multiplication some cultural experiments were conducted. The fungus did not form hyphae in solution media, but multiplied conidia directly in a budding way. Amount of conidial multiplication was counted by using Thomas' haema" tocytometer. In other cases, dry weight of conidial messes was measured after centrifugation of theliquid culture. The results obtained are as follows: 1) Endothia parasitica conidia was well multiplied on potato decoction and soy medium, on the contrary, poor in Richaed's and Czapek's solution. None of multiplication was observed in chestnut-leaves decoction. 2) The potential temperature range of Endothia parasitica conidia multiplication was 2°to 30℃, and the optimum tempereture was 18°-25℃. 3) The multiplication of Endothia parasitica conidia was rapidly proceeded from the second day after incubation and reached reached maximum on 10th day. Hereafter, it showed little increase 50 days. 4) The range of pH value in which the multiplication was possible was 3.0 to 8.0 and the optimum pH was 5.0 to 6.0. 5) Addition of glucose to the chestun-leaves decoction up to 10T promoted well the multiplication of Endothia parasitica, but an inffective result was obtained from the addition of sucrose. 6) Among the elements of nitrogen sources added to the culture media, CO (NH)_2 and KNO_3 were best and followed by L-arginine, L-glutamine, and L-asparagine in order to decreasing effect. 7) Good multiplication of Endothia parasitica was resulted in Richard's solution containing mannitor or glycerin and poor multiplication with maltose as its carbon sources.
