        출생 후 제대혈관 폐쇄에서의 세포고사

        Background: The mechanism of the closure of umbilical vessels is known to be multifactorial. In order to verify that apoptosis is one of the possible closure mechanisms, we studied to identify apoptosis in umbilical vessels and evaluate its mechanism by studying apoptosis-related gene and the relationship between the pattern of apoptosis and gestational age (GA). Methods: Twelve umbilical cords of GA of 37-42 weeks were obtained immediatly (less than 10 min. for minimal ongoing external influences) after birth. The presence of apoptotic cells was demonstrated by electron microscopy (EM) and terminal deoxynucleotidyl transferasemediated dUTP nick end labeling (TUNEL). Immnohistochemical staining and Western blotting were used for the analysis of the proteins of apoptosis-related gene. Results: Apoptosis of the smooth muscle cells of media and intima of umbilical vessels was identified at EM, regardless of GA from 37 to 42 weeks. The proportion of TUNEL(+) cells was 80% in intima, 40% in media, 80% in connective tissue of umbilical cord. The expressions of bax-a, bcl-Xs/L were strong in intima, in media and connective tissue, but those of bcl-2 were detected in only in connective tissue, regardless of GA in immunohistochemistry. The ratios of expressions of bax- a to bc1-2, bcl-Xs to bcl-XL, did not change with increasing GA from 37 to 42 weeks in Western blot- ting. Conclusion: Apoptosis was identified in umbilical vessels. The closure of umbilical vessels can be explained by apoptosis where the ratios of bax-a/bcl-2, bcl-Xs/ bcl-XL play an important role. The fact that there were no differences in the extent of apoptosis and the expressions of bax-a/bcl-2, bcl-Xs/bcl-XL according to GA, suggests that apoptosis of umbilical vessels is more dependent on the external stimuli during delivery than GA.

        키워드 네트워크 분석을 통한 항공우주산업 분야 과학기술의 연구 동향 분석

        연구 동향 분석은 데이터를 이용하여 특정 연구 분야의 동향을 분석하는 방법이다. 본 연구에서는 논문 키워드 네트워크를 통하여 연구 분야의 동향을 살펴본다. 항공우주산업은 주목 받고 있지만 항공우주 산업의 근간이 되는 과학기술에 대한 특징을 데이터에 근거해 분석한 연구는 많이 진행되지 않았다. 본 연구에서는 분석을 위해 2009년~2020년 발행된 항공우주산업 관련 3편의 SCI 저널 (Progress in Aerospace Science, Aerospace Science and Technology, IEEE Transaction On Aerospace and Electronic System) 논문을 선정하였으며, PAS에서 168편, AST에서 844편, TAES에서 2627편 총 3639편의 논문에서 5026개의 키워드를 수집하였다. 수집된 키워드는 정제하여 하나의 논문에서 생성된 키워드들을 링크로 서로 연결하며, 이렇게 구성된 키워드 링크 데이터를 UCINET 툴을 이용해 네트워크를 구성하고 네트워크 분석을 통해 연구 동향 분석을 수행한다. 본 연구를 통해 항공우주 기술 분야의 키워드 네트워크는 평균적으로 하나의 키워드가 8.788개의 키워드와 연결되어있고, 밀도는 0.002 값을 가지며, 클러스터링 계수는 0.451로 매우 낮다. 이는 항공우주산업 기술에서는 항공기 기체, 비행체 제어, 공력, 환경규격, 항공 전자 및 소프트웨어, 인공위성, 추진, 항공기 엔진, 항공기 인증, 발사체, 무인기, 탐지 추적, 광학 등의 다양한 분야가 연구 및 통합되기 때문이다. 또한, 키워드 간 평균 거리가 4.05 값을 가지는데 이는 항공우주기술 분야가 좁은 세상 네트워크(small world network)를 이루고 있으며, 멱함수의 법칙의 특징을 가지는 것을 말한다. 네트워크에서 중심성이 높은 노드는 중요한 위치를 가지며 다른 노드들에 많은 영향력을 끼친다. 연결 중심성(Degree centrality), 매개 중심성(betweenness centrality) 지표를 활용하여 네트워크 내의 중요 키워드들을 분석하고 키워드들의 관계를 살펴봄으로서 항공우주산업의 연구동향을 분석한다. 본 연구를 통해 중요한 키워드로 ‘수신기(Receiver)’, ‘평가(Estimation)’, ‘탐지 및 추적(target track)’, ‘공기역학(aerodynamic)’, ‘궤적(Trajectory)’, ‘전산유체역학(computational fluid dynamic)’, ‘무인항공기(unmanned aerial vehicle)’등의 키워드가 선정되었으며, 이들 키워드는 항공우주산업 과학기술의 근간에서 중요한 키워드로 나타남을 확인할 수 있었다. 본 연구에서는 이 키워드를 중심으로 중요하게 선정된 키워드간의 관계를 살펴본다. Research trend analysis is a method of analyzing trends by using data in a specific research field. In this study, trends in the research field are examined and evaluated with thesis keyword network based on characterize the subject and keyword semantic analyze. Although the aerospace industry has received increasing attention in field of studies, science and technology characteristic analyzing in the aspect of data has not been frequently researched. For this study, 3 SCI journals papers (Progress in Aerospace Science, Aerospace Science and Technology, IEEE Transaction On Aerospace and Electronic System) related to the aerospace industry were selected, which were published from 2009 to 2020. And we collected a total 3639 papers that contains 5026 keywords which are 168 papers form PAS, 844 papers from AST and 2627 papers from TAES. The collected keywords from each paper were connected as one link, and performed network configuration with a linked keyword data set by using UCINET tool. Constructed keyword link data set was analyzed for aerospace trend research. Through this study, we found that a single keyword from aerospace technology field was connected with 8.788 keywords in average, the network keyword density had a value of 0.002 and the clustering coefficient was very low as 0.451. This is because various fields such as aircraft airframe, vehicle control, aerodynamics, environmental standards, avionics and software, satellite, propulsion, aircraft engine, aircraft certification, launch vehicle, unmanned aerial vehicle, detection and tracking and optics are researched and integrated in the aerospace industry technology. In addition, the average distance among keywords has a value of 4.05, which means that the aerospace technology field forms a small world network and has the characteristic of the power law. A node with high centrality in the network link has an important position and has a lot of influence on other nodes. And the research trend of the aerospace industry is analyzed per important keywords in the network using the degree centrality and between centrality indicators and examining the relationship between keywords. From this study, important keywords such as ‘Receiver’, ‘Estimation’, ‘target track’, ‘aerodynamic’, ‘Trajectory’, ‘Computational fluid dynamics’, ‘computational fluid dynamic’ and ‘unmanned aerial vehicle’ were selected. And it confirms that these keywords appeared as important foundation of science and technology in the aerospace industry. For this study also examines the relationship between keywords selected as centering keyword.

        LP9M80-H Isolated from Liriope platyphylla Could Help Alleviate Diabetic Symptoms via the Regulation of Glucose and Lipid Concentration

        LP9M80-H는 맥문동(Liriope platyphylla)으로부터 메탄올과 헥산을 이용하여 추출한 새로운 추출물로서 ICR 마우스에서 인슐린분비를 촉진하며, 간과 뇌 조직에서 인슐린 신호경로를 활성화시키는 것으로 알려져 있다. 본 연구에 서는 LP9M80-H가 당뇨와 비만의 치료에 미치는 효과를 조사하기 위하여, OLETF 모델동물에 LP9M80-H를 2주간 투여한 후 당뇨와 비만과 관련된 주요인자의 변화를 관찰하였다. 비록 체중은 두 집단간에 차이가 없었으나 복부 지방량은 vehicle 투여군보다 LP9M80-H 투여군에서 적었다. 또한, 혈중 포도당농도는 LP9M80-H를 투여한 OLETF랫드가 대조군에 비하여 약간 낮았으나 인슐린의 농도는 유의적으로 크게 증가하였다. 혈청 내 3가지 주요 지질의 농도는 LP9M80-H를 투여한 OLETF 랫드에서 유의적으로 감소하였고, 지방의 산화를 촉진하는 아디포넥틴의 농도도 LP9M80-H를 투여한 OLETF 랫드에서 감소하였다. 더불어, 체내에 분비된 인슐린이 표적장기에 미치는 영향을 관찰하기 위하여 간조직에서 인슐린 수용체와 인슐린 수용체기질(IRS)의 발현을 관찰하였으며, 이러한 2가지 단백질은 LP9M80-H를 투여한 OLETF 랫드에서 vehicle 투여군에 비해 유의미하게 감소하였다. 또한, 인슐린 신호경로의 다운스트림에 위치하는 포도당 수송체 중에서 Glut-2와 Glut-3 발현은 LP9M80-H를 투여한 OLETF 랫드에서 유의미하게 감소하는 반면에, Glut-4 발현은 일정하게 유지되었다. 따라서 이러한 결과는 LP9M80-H는 포도당항상성과 지질농도의 조절을 통하여 당뇨와 비만의 증상을 완화시키는데 기여할 것으로 사료된다. It was reported that the novel compounds (LP9M80-H) of Liriope platyphylla regulate glucose transporter (Glut) biosynthesis by activating the insulin-signaling pathway in the liver and brain of ICR mice. To investigate the therapeutic effects of LP9M80-H on the pathology of diabetes and obesity, alterations of key factors related to symptoms were analyzed in the Otsuka Long Evans Tokushima Fatty (OLETF) rats treated with LP9M80-H for 2 weeks. The abdominal fat masses in the LP9M80-H-treated group were lower than the vehicle-treated group, although there was no difference in body weight between the two groups. Additionally, when compared to the vehicle-treated group, LP9M80-H treatment induced a significant decrease in glucose levels and an increase in the insulin concentration in the blood of OLETF rats. A high level of insulin protein was also detected in pancreatic β cells of LP9M80-H-treated OLETF rats. A significant reduction in the concentration of lipids and adiponectin was detected only in LP9M80-H-treated OLETF rats. Furthermore, the expression of insulin receptor β and the insulin receptor substrate (IRS) was dramatically decreased in LP9M80-H-treated OLETF rats compared to the vehicle-treated group. Of the glucose transporters located downstream of the insulin-signaling pathway, glucose transporters (Glut) -2 and -3 were significantly decreased in LP9M80-H-treated OLETF rats, while the level of Glut-4 was maintained under all conditions. Therefore, these results suggest that LP9M80-H may contribute to relieving symptoms of diabetes and obesity through glucose homeostasis and regulation of lipid concentration.

        LP-M, a Novel Butanol-Extracts Isolated from Liriope platyphylla, could Induce the Neuronal Cell Survival and Neuritic Outgrowth in Hippocampus of Mice through Akt/ERK Activation on NGF Signal Pathway

        맥문동(Liriope platyphylla)은 동양의학에서 오래 동안 항염증제, 당뇨 혹은 비만 치료제, 그리고 신경세포활성화 약제로 사용되어 왔다. 본 연구에서는 맥문동으로부터 NGF의 발현을 촉진하는 새로운 물질을 개발하고 이들의 작용기전을 밝히기 위하여 맥문동으로부터 13가지의 새로운 추출물을 확보하고 이들의 기능을 세포주와 마우스 실험을 통해 분석하였다. 그 결과, 상대적으로 세포독성이 낮고 NGF의 분비량이 많은 LP-E, LP-M, LP-M50, LP2E17PJ 등 4가지 추출물을 확보하였다. 또한 이들 중에서 LP2E17PJ를 제외한 3가지 추출물에 의해 분비된 NGF는 PC12세포의 neuritic outgrowth를 촉진하였다. 더불어 추출방법과 추출량의 측면에서 효과적인 LP-M을 C57BL/6 마우스에 2주간 투여하여 뇌조직에서 NGF mRNA의 발현을 확인하였다. LP-M은 오직 피질에서만 두 종류의 receptor 중에서 low affinity neceptor를 통한 억제신호를 전달하였으며, 해마에서는 유의적인 변화를 유도하지 못했다. 그러나 high affinity receptor를 통한 신호전달을 해마에서만 활성화 신호를 전달하였고 피질에서는 변화를 유도하지 못했다. 이러한 결과는 맥문동 추출물인 LP-M은 마우스 대뇌의 해마에서 high affinity receptor를 통한 신호전달을 통해 neuritic outgrowth를 촉진함을 확인하였다. Liriope platyphylla has been used in oriental medicine as an effective medical plant to improve symptoms of cough, sputum production, neurodegenerative disorders, obesity and diabetes for long time. In order to investigate the effects of novel extracts on nerve growth factors (NGF)-stimulated neuritic outgrowth, the alteration of NGF expression and NGF receptor signaling pathway were detected in neuroblastoma cells and C57BL/6 mice. Of a total of 13 novel extracts, 4 extracts (LP-E, LP-M, LP-M50, LP2E17PJ) showed high viability on MTT assay. Also, all of these extracts induced NGF secretion and NGF mRNA expression in neuroblastoma cells. However, the NGF-induced neuritic outgrowth from PC12 cells was only stimulated by LP-E, LP-M and LP-M50. Furthermore, we selected LP-M as a best candidate, based on method and amounts of extraction, in order to verify its effect in mice. C57BL/6 mice were treated with 50 mg/kg of LP-M for 2 weeks and the effects on NGF regulation were analyzed with various methods. The expression of NGF mRNA was significantly increased in LP-M treated mice compared to vehicle treated mice. Also, the signaling pathway of p75NTR was inhibited in the cortex by LP-M treatment, with no change in the hippocampus of brain. However, the signaling pathway of TrkA was dramatically activated in only hippocampus via LP-M treatment. Therefore, these results suggest that the novel four extracts of L. platyphylla may contribute to the regulation of NGF expression and secretion in neuronal cells. LP-M was especially considered to be an excellent candidate for a neurodegenerative disease-therapeutic drug.

        γ-Secretase 활성억제단백질인 TMP21의 과발현이 신경세포주에서 NGF 수용체 신호전달과 정에 미치는 영향

        TMP21은 AD의 원인으로 작용하는 Aβ-42 펩타이드 생성에 중요한 γ-secretase 활성을 억제하는 p24 family에 속하는 type I 막 단백질이다. 본 연구에서는 TMP21이 세포의 성장과 분화에 중요한 NGF 수용체 신호전달과정에 미치는 영향을 분석하고자 인간의 TMP21 cDNA를 합성하고, CMV promoter 조절 하에 hTMP21를 클로닝하여, CMV/hTMP21 벡터를 제조하였다. 그리고 이들 벡터를 B35 neuroblastoma에서 과발현시킨 후 γ-secretase 구성단백질과 NGF 수용체 연관 단백질의 변화를 관찰하였다. 그 결과, 4종류의 γ-secretase 구성단백질의 발현은 vehicle transfectants보다 CMV/hTMP21 transfectants에서 유의적으로 감소하였다. 또한 NGF low affinity 수용체인 p75<SUP>NTR</SUP>과 downstream 단백질인 RhoA의 양은 NGF를 처리하지 않은 TMP21 transfectants에서 유의적으로 증가하였으나 NGF 처리에 의해 감소되었다. High affinity NGF 수용체인 TrkA의 인산화도 NGF 처리가 없는 경우 유의적으로 감소하였으나 NGF 처리에 의해 증가되었다. 또한 downstream 신호전달 과정 중에서 ERK의 인산화는 TrkA와 유사한 발현변화를 나타내었으나 Akt 인산화는 NGF의 처리에 의해 더욱 증가하였다. 이러한 결과는 TMP21이 neuroblastoma에서 NGF 수용체 신호전달과정를 조절하는 중요한 단백질로서 작용함을 제시하며, AD의 작용기전 연구에 중요한 기초자료를 제공할 것으로 사료된다. Transmembrane protein 21 (TMP21) is a member of the p24 cargo protein family and has been shown to modulate γ-secretase-mediated Aβ production which was specifically observed in the brains of subjects with Alzheimer’s disease (AD). In order to investigate whether TMP21 could affect nerve growth factor (NGF) receptor signaling pathway, the alteration of NGF receptors and their downstream proteins were detected in TMP21 over-expressed cells. CMV/hTMP21 vector used in this study was successfully expressed into TMP21 proteins in B35 cells after lipofectamin transfection. Expressed TMP21 proteins induced the down-regulation of γ-secretase complex components including Presenlin-1 (PS-1), PS-2, Nicastrin (NST), Pen-2 and APH-1. Also, the expression level of NGF receptor p75<SUP>NTR</SUP> and RhoA were significantly higher in CMV/hTMP21 transfectants than vehicle transfectants, while their levels returned to vehicle levels after NGF treatment. However, the phosphorylation of NGF receptor TrkA was dramtically decreased in NGF No-treated CMV/hTMP21 transfectants compared with vehicle transfectants, and increased in NGF treated CMV/hTMP21 transfectants. In TrkA downstream signaling pathway, the phosphorylation level of ERK was also decreased in CMV/hTMP21 transfectants, while the phosphorylation of Akt was increased in the same transfectants. Furthermore, NGF treatment induced the increase of phosphorylation level of Akt and ERK in CMV/hTMP21 transfectants. Therefore, these results suggested that over-expression of TMP21 may simultaneously induce the up-regulation of p75<SUP>NTR</SUP>/RhoA expression and the down-regulation of TrkA/ERK phosphorylation through the inhibition of γ-secretase activity.

        거대세포동맥염에 동반된 급성 골수성백혈병 1예

        Giant cell arteritis (GCA), the most common form of systemic vasculitis in adults, preferentially involves large and medium-sized arteries in patients over the age of 50. The classic manifestations are headaches, jaw claudication, polymyalgia rheumatica, and visual symptoms. Acute myeloid leukemia (AML) is a hematopoietic stem cell disorder characterized by a block in the differentiation of hematopoiesis, resulting in the growth of a clonal population of neoplastic cells or blasts. This malignant alteration in hematopoietic stem cells leads to a loss of normal hematopoietic function, which, if left untreated, typically leads to death within weeks to months of its clinical presentation. Although there have been reports of CLL or CML accompanied by several kinds of autoimmune vascular diseases, such as polymyalgia rheumatica, GCA, or necrotizing temporal arteritis, no studies have reported a case of AML with GCA. We experienced an 80-year-old male patient who developed AML 6 years after the diagnosis of GCA. He was under the use of oral glucocorticoid, hydroxychloroquine, and methotrexate at the time of the diagnosis of the AML. This is the first case in Korea to report GCA accompanied by AML.

        잠재적인 생균제제 Lactobacillus plantarum P1201을 이용한 항산화 활성이 증진된 두유-요구르트 제조

        잠재적인 생균제제 L. plantarum P1201의 두유 발효 중 이화학적 특성, 총 phenolics과 isoflavone 함량 및 항산화 활성 변화를 연구하였다. 균주 P1201는 pH 2.5의 산 및 인공위액산 환경에서 4 h 후 각각 58.14%와 62.22%의 생존율을 나타내었다. 두유 요구르트의 최적 발효 조건은 배양온도 35℃, 종균 접종량 5.0% 및 10%의 sucrose이었다. 한편 총 phenolic와 isoflavone 함량 및 항산화 활성은 SM 요구르트보다 SPM 요구르트가 우수하였다. 특히 SPM 발효 48 h 후, daidzein, glycitein 및 genistein의 함량은 각각 91.50, 12.34 및 61.39 ㎍/g이 검출되었다. 이러한 결과로 본 연구의 두유 요구르트 추출물은 잠재적인 천연 항산화제로 식품에서 사용할 수 있을 것으로 판단되었다. In this study, we evaluated changes in physicochemical properties, total phenol and isoflavone contents, and antioxidant activity during soymilk fermentation by a potential probiotic Lactobacillus plantarum P1201. The P1201 strain showed survival rates of 58.14% and 62.22% after 4 h exposure to acid and artificial gastric acid conditions (pH 2.5), respectively. The optimal conditions for soy-yogurt fermentation by L. plantarum were determined to be as follows: temperature of 35℃, seed volume of 5.0%, and sucrose content of 10.0%. The total phenol and isoflavone contents and antioxidant activity were higher in soy-powder milk (SPM) yogurt than soymilk yogurt. In particular, the highest levels of isoflavone-aglycones, such as daidzein, glycitein, and genistein (91.50, 12.34, and 61.39 ㎍/g, respectively), were observed after 48 h of SPM fermentation. Thus, these results suggest that the soy-yogurt extract could be used as a potential source of natural antioxidants in food.

The continuity and three-dimensional Reynolds-averaged Navier-Stoke equations were used as governing equations to analyze the flow characteristics around the rail-type rescue system. SST(Shear Stress Transport) turbulence model was used as turbulence closure. The unstructured tetrahedral meshs were used to construct the grid system for the computational domain. The boundary conditions were maintained as the same conditions of the measured data of our previous works. In front of EMM(Emergency Measures and Maintenance) system, the velocity and static pressure were distributed as the value 1~4㎧ and 112~126㎩, respectively.
