제주 파인애플 구아바 (Acca sellowiana) 추출물의 항산화 활성 및 피부 안전성

This study analyzed the antioxidant activity and skin primary irritation test by using hot-water extracts of Acca sellowiana leaves (WAL) and branches (WAB). Due to the recently warmed climate, Jeju Island produces subtropical plants, and the efficacy of one of them, A. sellowiana, has been confirmed. DPPH · ABTS · DMPD radical scavenging activity, ferrous-ion chelating activity, cupric reducing antioxidant capacity (CUPRAC), reducing power assay, ferric-reducing antioxidant power (FRAP) were teseted, and total phenol and phenol contents were analyzed. In addition, through skin primary irritation test, it was investigated whether it can be used as a cosmetic ingredient. As a result, in all antioxidant activity experiments, the activity of WAL and WAB increased in a concentration-dependent manner. In addition, the content of flavonoids was contained in the WAL more than WAB, and accordingly, the antioxidant activity of the WAL was better. In addition, WAL is hypoallergenic and WAB is a non-irritating ingredient, suggesting that it is possible as a natural material that can be used in cosmetics in the future.

To identify the natural and novel cosmeceutical ingredients which have whitening function, we examined the effects of tyrosinase activity and melanin production on halophyte Limonium tetragonum (Thunb.) A. A. Bullock extracts. As a result, the extracts showed whitening effect with no cytotoxicity. Melanin contents of B16F10 melanoma cells were decreased in a dose-dependent manner at 50, 100, 200 μg/mL treatment of the extracts. In tyrosinase activity inhibition test, L. tetragonum (Thunb.) A. A. Bullock extracts showed decreased tyrosinase activity as the concentration of α-MSH was increased. Furthermore, it was observed that the tyrosinase and MITF expression was significantly downregulated by adding L. tetragonum (Thunb.) A. A. Bullock extracts in α-MSH, a melanogenesis inducing material, treated B16F10 melanoma cells. These results indicate that L. tetragonum (Thunb.) A. A. Bullock extract might be an effective whitening agent by inhibit melanin formation.

The anti-inflammatory activity and ignorable toxicity of Rumex acetosella extract (J9) were investigated and confirmed by measuring the expression level of NO, PGE2, and pro-inflammatory cytokines (IL-1β, IL-6, TNF-α) in lipopolysaccharide (LPS)- and J9-treated in RAW264.7 macrophages. Relative to a LPS-only-treated group, the inhibition effect of J9 on the production of NO, PGE2, pro-inflammatory cytokines was increasing across J9-treated samples but the cell viabilities stayed constant. We also exhibited that the iNOS expression level in the J9-treated samples was dose-dependently decreased relative to its untreated control and the elevated expression level of COX-2 seemed to be equal across samples treated with LPS alone or in combination with J9.

Nucler factor-kappa B (NF-kappaB) 프로모터는 염증성 질환을 유도하는 호염증성 시토카인의 발현에 중요한 역할을 수행하는 전사인자 중의 하나이다. 본 실험에서는 200 여종의 식물추출물들로부터 항염효능이 있는 추출물을 선발하기 위해 NF-kappaB 리포터 실험을 수행하였다. NF-kappaB 리포터 실험결과, 12종의 식물추출물, 즉 개나리, 고추잎, 박하, 뱀딸기, 뽕나무, 삼백초, 솔잎, 양애줄기, 약쑥, 어성초, 왕벚꽃가지, 조릿대 등이 lipopolysaccharide (LPS)에 의해 유도된 NF-kappaB 프로모터 활성을 농도의존적으로 억제하는 것을 확인하였다. 이들 12종의 식물추출물이 호염증성 시토카인 발현에도 동일한 효과를 나타내는지 알아보기 위해 tumor necrosis factor-alpha (TNF alpha)와 인터루킨-8에 대한 ELISA실험을 실시하였다. ELISA실험 결과, NF-kappaB 리포터 실험결과와 동일하게, TNF-alpha와 인터루킨-8 생산이 12종 식물추출물 모두에서 감소됨을 관찰하였다. 이러한 실험결과는, 12종의 식물 추출물에서 보여지는 호염증성 시토카인 억제효과가 NF-kappaB 프로모터 활성억제를 통해 이루어지고 있음을 시사한다. 또한, 이들 12종 식물은 diphenyl-p-picrylhydrazyl (DPPH) assay를 통해 살펴본 결과 높은 항산화 활성도 있음을 확인하였다. 이상의 결과로부터, 12종의 식물 추출물은 염증성 피부질환 전용 화장품 제형에서 항염 및 자극완화 소재로 응용될 수 있음을 확인하였다. Nuclear factor-kappaB (NF-kappaB) is a critical transcription factor for maximal expression of many of the cytokines that are involved in the pathogenesis of inflammatory diseases. In this study, we found that 12 plant extracts among 200 plants, namely, Forsythia koreana, Capsicum annuum L, Mentha arvenis, Duchesnea chrysantha, Morus alba, Saururus Chinenis (Lour) Baill, Pine needle, Zingiber mioga (Thunb.), Roscoe, Houttuynia, Prunus yedoenis, Sasa quelpaertenis, significantly inhibited LPS- induced NF-kappaB activation in a concentration-dependent manner. Additionally, 12 plant extracts were found to have antioxidant activities in DPPH assay Therefore, we have attempted to determine whether 12 herbal extracts could inhibit the expression of cytokines possessing NF-kappaB promoter in their promoter regions. Consistently 12 herbal extracts inhibited LPS-induced production of TNF alpha and interleukin-8 (IL-8). These results show that 12 herbal extracts suppresses the production of pro-inflammatory mediators through the inhibition of the NF-kappaB signaling pathway, we suggest that 12 herbal extracts can be used as a anti-inflammatory and soothing agent.

Melanin is one of the most important factors affecting skin color. Melanogenesis is the bioprocess of melanin production by melanocytes in the skin and hair follicles and is mediated by several enzymes, such as tyrosinase, tyrosinase related protein (TRP)-1, and TRP-2, MITF. In this study, we investigated the effect of Artemisia fukudo Makino extracts on tyrosinase activity and melanin production as natural products of whitening functional cosmetics. Melanin content in murine B16F10 melanoma cells were decreased by Artemisia fukudo Makino extracts in a dose-dependently. In addition, the inhibition of tyrosinase activity of Artemisia fukudo Makino extracts showed to decrease tyrosinase activity as the concentration of α-MSH was increased. Furthermore, western blot analysis revealed that Artemisia fukudo Makino extracts significantly downregulated the expression of tyrosinase, TRP-1 which treat of α-MSH–induced melanogenesis in murine B16F10 melanoma cells. As a result, Artemisia fukudo Makino extract showed functionalities as an effective whitening agent to inhibit melanin formation.
애기수영, 방가지똥 및 암대극 추출물이 Melanoma Cell에서 멜라닌 합성에 미치는 영향

In this study, we investigated the effect of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts on tyrosinase activity and melanin production as natural products of whitening functional cosmetics. To measure the melanin production, 50, 100, 200 μg/mL of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts were treated on α-MSH treated B16F10 melanoma cells, respectively. Melanin contents in α-MSH treated B16F10 melanoma cells were decreased by 41.5, 51.11, and 61% in 200 μg/mL treatment compared to none treatment, respectively. In addition, the intracellular tyrosinase activity was decreased after treatments with all extracts. Furthermore, 100 μg/mL of Euphoibia jolkini extract was decreased 81.5% of melanin production in B16F10 melanoma cells. When the three extracts were compared, Euphoibia jolkini extract was considered to be the most functional material for whitening effect.

The present study aimed to investigate comparative anti-inflammatory effects of Syzygium samarangense (SS, Wax apple) leaf and branch extract considering the balance of safety and efficacy. SS is a plant species in the family Myrtaceae, native to an area that includes the Greater Sunda Islands, Malay Peninsula but now cultivated in Jeju Island. Dose response studies were conducted to determine the inhibitory effect of extracts on the expression of inflammatory enzymes: Prostaglandin (PGE₂) and nitric oxide (NO) inflammatory cytokine production, TNF-α, interleukin (IL)-6, interleukin (IL)-1β and inducible nitric oxide synthase (iNOS), and cyclooxygenase (COX-2) in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. As a result, SS were shown higher anti-inflammatory activity. SS inhibited production of PGE2, NO and mRNA expression of iNOS, COX-2 and decreased TNF-α, IL-6, IL-1β induced by LPS stimulation in RAW 264.7 cells. SS at 800 μg/mL showed inhibitory effects on almost all the inflammatory mediators we examined except for all study regulation. In conclusion, this study suggest that SS could be a potential source as a new natural anti-inflammatory agent.

피부미백 소재를 개발하기 위해 멜라닌 함량, 세포내 tyrosinase 활성의 측정 및 Western blotting 실험이 수행되었다. 백합(Lilium Oriental Hybrid ‘Siberia’) 뿌리의 80% 에탄올 추출물로부터 얻은 에틸아세테이트 분획물(R-EA)은 α-melanocyte stimulating hormone(α-MSH)에 의해 멜라닌 생성이 유도된 B16/F10 흑색종 세포에서 농도 의존적으로 멜라닌 생성을 저해하였다. 정확하게 세포내 tyrosinase 활성과 멜라닌 함량은 에틸아세데이트 분획물 100μg/mL 처리 시 α-MSH 단독 처리군에 비해 각각 45%와 74%의 저해율을 보였다. α-MSH에 의해 멜라닌 생성이 유도된 B16/F10 흑색종 세포에서 단백질 발현양상을 살펴본 결과 TRP-1이 가장 많이 억제된 양상을 확인할 수 있었고 이 결과는 세포내 tyrosinase 활성저해보다 멜라닌 생성저해가 더 많이 일어난 것과 일맥상통하는 것이다. 이를 종합해 볼 때 p-coumaric acid와 resveratrol 함량이 백합뿌리의 에탄올 추출물에 비해 많이 함유된 에틸아세테이트 분획물은 멜라닌 생성 유도물질에 의해 촉진된 ERK의 활성화를 억제하는 피부미백 소재로서 그 가치가 입증된다고 사료된다. In order to develop a skin-whitening agent, melanin contents and intracellular tyrosinase activity were determined by western blotting. Ethyl acetate fractions of 80% ethanol extracts from lily (Lilium Oriental Hybrid ‘Siberia’) bulbs (R-EA) inhibited melanin synthesis in a dose-dependent manner in α-melanocyte stimulating hormone (α-MSH)-treated B16/F10 murine melanoma cells. Intracellular tyrosinase activity and melanin contents were suppressed by 45% and 74%, respectively, in response to treatment with 100 μg/mL of R-EA. Examination of protein expression associated with α-MSH-induced melanogenesis showed that tyrosinase related protein (TRP)-1 was inhibited more strongly than tyrosinase, and these results were correlated with stronger inhibition of melanin synthesis than intracellular tyrosinase activity. Taken together, R-EA containing p-coumaric acid and resveratrol could be used as a hypopigmentation agent through suppression of sustained extracellular signal-regulated kinase (ERK) activation via melanogenic induction.
