This study was conducted to assess the chemopreventive effects of sodium selenite in the rat medium-term multi-organ bioassay using a DMBDD model (DEN+MNU+BBN+DMH+DHPN). Seventy five, 6-week-old, male SD rats were divided into 3 groups. The animals in group 1 received DEN(diethylnitrosamine, 100 ㎎/㎏ bw, single i.p., in saline), MNU (N-methyl-nitrosourea, 20 ㎎/㎏ bw, i.p., 4 times for 2 weeks), BBN (N-butyl-N-(4-hydroxybutyl) nitrosamine, 0.2% in drinking water for 2 weeks), DMH (1,2-dimethylhydrazine, 40 ㎎/㎏ bw, s.c., in saline, 4 times for 2 weeks), and DHPN (N-bis(2-hydroxy-propyl)nitrosamine, 0.1% in drinking water for 2 weeks), then were placed on sodium selenite (4 ppm in drinking water) for 22 weeks from weeks 4 to 26. The animals in group 2 were given DMBDD alone. The animals in group 3 were given sodium selenite alone. Animals were sacrificed at week 12 for ACF quantitative analysis and at week 26 for tumor induction. The body weights in the group 1 were significantly decreased compared with those of group 2. The tumor multiplicities of large intestine in the group 1 were significantly decreased compared with those of group 2 (P<0.05). These results indicate that sodium selenite may have a potential as chemopreventive agents of colon carcinogenesis.

This study was performed with investigation on current operation of foreign GLP regulation. The analysis of the foreign GLP regulation and operation was usually based on OECD GLP website and OECD mutual joint visiting (MJV) report. The MJV report of major OECD countries were contained information of GLP monitoring authority and program, organization including GLP regulation, number of inspector, type and method of inspection etc.. This study was also investigated comparative analysis of GLP regulation among domestic monitoring authorities. Improvement plan of domestic GLP regulation and preparations to the onsite evaluation of the OECD MJV to Korea in 2020 were suggested by the comparative analysis between foreign GLP and domestic GLP. These results in this study were summarized as GLP regulation characterization of 5 major OECD countries, analysis of recent OECD GLP inspection report, regulation difference of domestic GLP monitoring authorities, and preparation to the on-site evaluation of the OECD MJV. This study will be utilized as education material of domestic GLP inspector and preparation to the onsite evaluation of the OECD MJV to Korea in 2020.

This study was conducted to assess the modifying effects of diisodecyl phthalate (DIDP) and diethylhexyl phthalate (DEHP) in the rat medium-term multi-organ bioassay using five different carcinogens. Seventy five, 6-week-old, male F344 rats were divided into four groups. Animals were sacrificed at week 12. To evaluate modifying effects of preneoplastic lesions of DIDP and DEHP, we measured glutathione S transferase placental form (GST-P) and 5-bromodeoxyuridine (BrdU) expression level in liver, numbers of aberrant crypt foci (ACF) in colon and performed histopathological observation. The body weights of rats given 12,000 ppm DEHP and 8,000 ppm DIDP were significantly decreased compared to those of control group. Relative liver and kidney weights of rats given DIDP were significantly increased compared to those of control group in a dose dependent manner. There was no significance of GST-P positive foci and BrdU labeling indices in liver and numbers of ACF in colon among each group. Although hyperplasia of urinary bladder of rat given DEHP were significantly increased compared to that of control group, there were no significant changes of preneoplastic lesions on all organs of rats given DIDP. These results indicate that DIDP has no modifying effect on preneoplastic lesions in all organs.
실험적 대장암 모델에서 키토산의 발암 억제효과에 관한 연구

This study was conducted to assess the chemopreventive effects of chitosan in a rat colon carcinogenesis induced by azoxymathane (AOM). Ninety, 5-week-old, male F344 rats were divided into three groups. THe animals in group 1 received subcutaneous injections of 15㎎/㎏ AOM three times for two weeks, then were placed on powdered basal diet containing 2% chitosan for 37 weeks from weeks 3 to 40. The animals in group 2 were given AOM alone. The animals in group 3 were given 2% chitosan without prior carcinogen treatment. All animals were sacrificed at week 12 for quantitative analysis of aberrant crypt foci (ACF) and at week 40 for analysis of tumor induction. Total numbers of ACF and AC per colon of group 1 were not significantly different from those of group 2. Tumor incidences and multiplicities of small intestine in the group 1 were significantly decreased compared with those of the group 2 (P<0.05). According to pathological diagnoses, adenocarcinoma incidence and multiplicity in the small and large intestine in the group 1 were significantly decreased compared with those of the group 2 (p<0.05). No toxic effects were observed in animals given chitosan in terms of body weights, and liver or kidney histology. These results indicate that chitosan may have a potential as chemopreventive agents of colon carcinogenesis during the postinitiation stage.

Corrosion of reinforcement is a major problem affecting a large number of reinforced concrete structures, this paper presents the results of an experimental study on reinforced concrete beams to determine their residual flexural capacity after undergoing different corrosion degrees (corrosion length, corrosion location, corrosion rate) of reinforcing bars. Corrosion was induced in the laboratory by an accelerated corrosion technique using two sources of external power supply. The results show marked reductions in flexural strength due to reinforcement corrosion.
화학적 간발암모델에서 Bisphenol A가 간암의 전암성 및 종양성 병변에 미치는 영향

Bisphenol A(BPA) was known as an endocrine disrupting chemical. This study was conducted to assess the effect of BPA, weaker estrogen, on the preneoplastic and neoplastic lesions induced by diethylnitrosamine (DEN). One hundred male F344 rats were divided into four groups which were treated with DEN followed by BPA. To make liver tumor early, we conducted that DEN containing osmotic pump implanted into rat abdominal cavity. Then diet containing BPA were fed to the rats. All animals were sacrificed at 10 and 18 weeks. Body weights were significantly decreased in 4000ppm at 18 weeks. Relative Liver weights of 4000 ppm BPA treated group were significantly increased compared to that of DEN alone group at 18 weeks. There were no significant differences of liver tumor incidences. Sum area of GST-P positive foci and BrdU labeling indices of BPA treated group were not significantly different compared to those of control group. These results suggest that BPA have no effects in preneoplastic and neoplastic lesions on DEN-induced hepatocarcinogenesis.
