Thin Layer Chromatography 법(法)에 의한 ${\alpha}-Solanine$의 정량(定量)

Thin-layer chromatography을 이용(利用)하여 감자중(中)의 ${\alpha}-solanine$의 분리(分離), 정량방법(定量方法)을 모색하였다. Methanol-chloroform (2 : 1, v/v) 혼합용매를 이용한 감자 추출액에 0.2N HCl 용액을 가(加)하여 색소물질(色素物質)을 비롯한 불순물(不純物)을 제거한 후 진한 암모니아 용액(溶液)을 가(加)하여 생성(生成)된 침전을 원심분리(遠心分離)(25,000 rpm, $4^{\circ}C$, 10min.)함으로써 solanine의 침전을 얻었다. 얻어진 solanine 침전을 1% $NH_4OH-absolute$ ethanol-chloroform(1 : 2 : 2, v/v)의 용매 system을 사용(使用)한 TLC로 ${\alpha}-solanine$과 ${\alpha}-chaconine$을 분리(分離)하였다. 분리(分離)된 ${\alpha}-solanine$은 Infrared spectrophotometry에 의해서 순도(純度)가 확인(確認) 되었다. 분리(分離)된 ${\alpha}-solanine$을 진한 황산-1% paraformaldehyde(2:1, v/v) 용액(溶液)과 반응(反應)시킨 다음 310nm에서 흡광도(吸光度)를 측정(測定)하여 정량(定量)하였다. ${\alpha}-solanine$의 진한 황산-1% paraformaldehyde에 의한 발색용액(發色溶液)의 molar absorptivity는 2,090이었다. The thin Layer chromatographic determination of ${\alpha}-solanine $ in potato tuber was investigated. The solanine was extracted with methanol-chloroform mixture (2 : 1, v/v) and precipitated as the crude glycoalkaloid by the addition of ammonium hydroxide. The solanine in the crude precipitate was separated by TLC using a solvent system of 1% $NH_4OH-absolute$ ethanol-chloroform (1:2:2, v/v). The purity of ${\alpha}-solanine $ was confirmed by infrared spectrophotometry. The ${\alpha}-solanine $ separated by TLC was determined at the wavelength of 310nm after coloration with concentrated sulfuric acid-1% paraformaldehyde, with the molar absorptivity of 2,090.

Purpose: The effects of tocopherols on the iron-catalyzed lipid oxidative stability of soybean oil-in-water emulsion with a peppermint extract were studied. Methods: The emulsion consisted of tocopherol-stripped (TSSO) or refined, bleached and deodorized (RBDSO) soybean oil (40 g), citrate buffer (60 g, pH 4.0), and FeSO4 (0.5 mg) with/without a 75% ethanol extract (400 mg/kg) of peppermint (Mentha × piperita). Lipid oxidation was evaluated based on the hydroperoxide and p-anisidne values of the emulsion. Polyphenol compound and tocopherol contents were determined using a spectrophotometer and a HPLC, respectively. Results: Addition of peppermint extract significantly (p<0.05) decreased lipid oxidation of the TSSO emulsion, but showed no significant effect on the RBDSO emulsion. The polyphenol compounds in the emulsion with added peppermint extract were degraded, with higher degradation in the RBDSO emulsion with added peppermint extract than in the TSSO emulsion with the extract. On the other hand, tocopherol contents of the emulsions were not changed significantly, due possibly to the regeneration of tocopherols by polyphenol compounds. Conclusions: The peppermint extract decreased the lipid oxidation of a soybean oil-in-water emulsion via direct reaction of its polyphenol compounds on the TSSO emulsion lipids. The main role of polyphenol compounds of the extract in the RBDSO emulsion was to regenerate tocopherols, which indirectly reduced the level of lipid oxidation.

The aim of this study was to evaluate a methyl alcohol extract of Enteromorpha linza (MEEL) for its anti-obesity effects. We demonstrated the inhibitory effects of MEEL on adipogenesis by investigating the lipolytic effects of MEEL in 3T3-L1 preadipocytes. Treatment with MEEL significantly suppressed terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet content observed by Oil Red O staining. The concentrations of cellular triglycerides were also reduced in 3T3-L1 cells after treatment of MEEL as compared with the controls. Treatment with 300 and 500 Ռg/ml of MEEL caused 67 and 19% reduction in lipid droplet content, respectively. In addition, MEEL significantly reduced the expression levels of key pro-adipogenic transcription factors including peroxisome proliferator-activated receptor Ճ (PPARՃ), cytidine-cytidine-adinosine-adenosine- thymidine (CCAAT)/enhancer binding proteins Ձ (C/EBPՁ) and C/EBPՂ as compared with controls. Taken together, these findings demonstrated that MEEL showed strong anti-obesity effects.
Roles and action mechanisms of herbs added to the emulsion on its lipid oxidation

Quality of food emulsions is mainly determinedby their physicochemical stability such as lipid oxidation,and herbs as antioxidative food materials are added toimprove their quality and shelf-life. Despite the extensiveresearches, the chemistry and implications of herb additionin the lipid oxidation of emulsions are still confusing. Thisreview intended to provide the information on the roles andaction mechanisms of herbs in the lipid oxidation of foodemulsions, with focuses on polyphenols. Polyphenols act asantioxidants mainly via reactive oxygen species scavengingand metal chelating; however, their oxidation productsand reducing capacity to more reactive metal ions increasethe lipid oxidation. Factors such as structure, concentration,and distribution determine their anti- or prooxidant role. Interactions, synergism and antagonism, among polyphenolcompounds and the effects of tocopherols derived from oilon the antioxidant activity of herbs were also describedwith the involving action mechanisms.
