The concentrations of 4 kalosaponins from tissues of Kalopanax septemlobus (Thunb.)Koidz grown in 7 provenances in Korea were determined by HPLC. Kalosaponin contents in plant part were much higher in the inner bark(30.59 mg/g on the dry weight basis) than those of young leaves(22.74 mg/g on the dry weight basis) and root bark(18.02 mg/g on the dry weight basis). A considerable range of variation in the contents was observed among population. The kalosaponin contents in inner bark from each population were highest in the Mt. Barwang (30.37 mg/g on the dry weight basis) followed by Mt. Gariwang, Hanra II, Mangun, Paltan, and Hanra I population. A variation of kalosaponin contents among population may be affected by both environmental and genetic factors. Establishment of selection and propagation of high kalosaponin containing trees can be a good source for the development of valuable forest products.
유자의 성숙종자 배양 및 종자유래 배발생 현탁배양으로부터 체세포배발생을 통한 유자의 식물체 재생

유자의 성숙종자를 MS 기본배지에 치상하여 종자의 내피에서 주심조직 유래의 희고 부서지기 쉬운 배발생캘러스가 1.2%의 빈도로 형성되었다. 이 캘러스는 1 mg/L 2,4-D를 첨가한 MS배지에서 증식되었다. 증식된 캘러스를 0.1 mg/L kinetin을 첨가한 MS 배지에 옮겼을 때 많은 수의 체세포배가 형성되었다. 배발생캘러스를 1 mg/L 2,4-D를 첨가한 액체 배지에 넣어 배발생 현탁배양계를 확립하였다. 배양된 현탁배양세포를 0.5 mg/L ABA를 첨가한 고체배지에 평판하였을 때 높은 빈도로 체세포배로 발달하였으며 MS 기본배지 혹은 1 mg/L kinetin 첨가배지에서 소식물체로 발달하였다. 소식물체는 성공적으로 토양으로 옮겨서 온실에서 육성되었다. Off-white, friable embryogenic calluses were formed on the internal integument of mature seeds of yuzu (Citrus junos) cultured on Murashige and Skoog's basal medium at a frequency of 1.2%. Embryogenic calluses were proliferated when cultured on medium with 1 mg/L 2,4-D. Upon transfer to medium with 0.1 mg/L kinetin, embryogenic calluses produced numerous somatic embryos. Embryogenic suspension cultures were established by placing embryogenic calluses into liquid medium with 1 mg/L 2,4-D. When plated onto medium with 0.5 mg/L ABA, embryogenic cells developed into somatic embryos at a high frequency, and then regenerated into plantlets. Plantlets were successfully transplanted to potting soil and grown in a greenhouse.

A micropropagation system for black Locust(Rohinia pseudoacacia) was established by using shoots and pin-punctured leaves of in vitro germinated seedlings. The greatest number of shoots (an average of 10.5 shoots) was obtained when shoot tips were cultured on MS medium supplemented with 1.0 ㎎/ℓ BAP and 0.01 ㎎/ℓ NAA. When pin-punctured leaf explants were cultured on the same medium, mean number of 13.E shoots were produced. Shoot growth was accelerated by adding 50 ㎎/ℓ of silver nitrate (AgNO₃), an anti-ethylere compound to the culture medium. Each shoot was excised from the mass and transferred onto half strength MS medium for rooting. Zygotic embryos at different developmental stages were cultured on LS medium supplemented with various growth regulators to induce somatic embryos. When cultured on LS medium with 1.0 ㎎/ℓ 2,4-D. 14.3% of the zygotic embryos induced somatic embryos. Upon transfer onto the basal medium, somatic embryos sporadically converted into plantlets.

The influence of various levels of major medium components such as sucrose, nitrate, phosphate, plant growth regulators, and light intensity for cell growth and the production of anthocyanin content in cell cultures of Populus alba X P. glandulosa were investigated. Best results for anthocyanin yield were obtained using Mura.shige and Skoog(MS) medium containing 5% sucrose, 12.5% nitrate, 200% phosphate, 1.0㎎/ℓ indole-3-acetic acid(IAA), 1.0㎎/ℓ benaylaminopurine(BAP), and continuous illumination of 7,000 lux. On the other hand, maximum cell growth was achieved with 5% sucrose, 50% nitrate above 400% phosphate compare with that of MS basal mediumi, and 0.5mgj1 2, 4-dichlorophenoxyacetic acid(2, 4-D). Anthocyanin accumulation in a suspension cultured cells of given genotype was stimulated by subculturing onto the medium lacking 2, 4-D. Pigmented cell clusters were extracted with methanol containing 1% hydrochloric acid (HCl) and then anthocyanin was identified by thin layer chromatography (TLC) and U. V. spectrophotometer.
제주도 주목(Taxus cuspidata S. et Z.)의 Taxol 및 Cephalomannine의 함량분석

To seek for a stable supply source of taxol as an anticancer agent urgently needed for the treatment of ovarian cancer, the concentrations of taxol and cephalo mannine in bark and needles of 37 yew (Taxus cuspidata S. et Z.) trees grown in 6 locations of Cheju Island were analyzed by using HPLC. A considerable variation in the content was observed in plant part and the location. The average taxol content in the needles (220.3 ± 56 ㎍ per g dry wt) was ca. 4 times hgher than that in the bark, whereas the content of cephalomannine in the needles (353 ± 141 ㎍) was ca. 5 times higher than that in the bark. Particularly, even within Cheju Island, needles from trees grown near the coast including Sogwipo area contained much higher taxol contents than needles from trees grown at mountain locations, indicating that taxane level is affected by environmental effects. Taxol in the needles was confirmed by electrospray mass spectrometry. These results suggest that the needles of yew trees growing in Cheju Island may provide a renewable source for taxol.
Schima wallichii sp. liukiuensis 로 부터 분리된 β-Sitosterol Glycoside 항균물질의 안정성 및 돌연변이원성

Stability of the β-sitosterol glycoside from Schima wallichii sp. liukiuensis at various physical conditions were investigated, mutagenecity of the steroid saponin was determined by Ames test. When exposed in pH 3 to pH 8, the β-sitosterol glycoside was stable on antimicrobial activity against yeasts. The antimicrobial activity of the β- sitosterol glycoside also stable in high temperature, N₂, O₂ gas and light exposure, and metal ion. Ames test result revealed that (β-sitosterol glycoside did not have any mutagenic activity. These results suggest that the β-sitosterol glycoside might be a promising candidate as a natural antimicrobial compound.
