Small non-coding RNA를 발현하는 형질전환 벼의 환경위해성 평가 방법

Since the RNA interference (RNAi) had been discovered in many organisms, small non-coding RNA-mediated gene silencing technology, including RNAi have been widely applied to analysis of gene function, as well as crop improvement. Despite the usefulness of RNAi technology, RNAi transgenic crops have various potential environmental risks, including off-target and non-target effects. In this study, we developed methods that can be effectively applied to environmental risk assessment of RNAi transgenic crops and verified these methods in 35S::dsRNAi_eGFP rice transgenic plant we generated. Off-target genes, which can be non-specifically suppressed by the expression of dsRNAi_eGFP, were predicted by using the published web tool, pssRNAit, and verified by comparing their expressions between wild-type (WT) and 35S::dsRNAi_eGFP transgenic rice. Also, we verified the non-target effects of the 35S:: dsRNAi_eGFP plant by evaluating horizontal and vertical transfer of small interfering RNAs (siRNAs) produced in the 35S::dsRNAi_eGFP plant into neighboring WT rice and rhizosphere microorganisms, respectively. Our results suggested that the methods we developed, could be widely applied to various RNAi transgenic crops for their environmental risk assessment.

청소년의 게임과몰입과 고독감의 종단적 관계: 잠재성장모형을 적용하여

본 연구는 청소년을 대상으로 하여 청소년의 게임과몰입과 고독감의 변화양상과 두 변인의 상호적 관계를 살펴보고, 환경적 변인인 부모의 비일관적 양육방식, 과잉간섭적 양육방식과 개인적 변인인 사회기술, 자기통제, 성별이 어떠한 영향을 미치는지 검증하기 위해 잠재성장모형을 활용하였다. 이를 위하여 한국컨텐츠진흥원 게임이용자패널조사 4개년 청소년 848명의 응답 자료를 활용하였다. 연구결과를 요약하면, 첫째, 게임과몰입과 고독감은 모두 시간에 따라 선형적으로 성장하는 양상을 보였다. 둘째, 게임과몰입과 고독감의 초기치와 성장의 관계를 살펴보면, 게임과몰입의 초기값의 수준이 높을수록 고독감의 변화율은 감소하는 관계를 보였다. 한편, 고독감의 초기값의 수준은 게임과몰입의 변화율에 통계적으로 유의미한 영향을 미치지 않았다. 셋째, 게임과몰입의 초기치에 유의미한 영향을 준 공변인은 부모의 비일관적 양육방식, 부모의 과잉간섭적 양육방식, 개인적 변인인 사회기술, 자기통제, 성별이었다. 넷째, 게임과몰입의 변화율에 유의미한 영향을 준 공변인은 부모의 과잉간섭적 양육방식, 개인적 변인인 사회기술, 자기통제, 성별이었다. 다섯째, 고독감의 초기치에 유의미한 영향을 준 공변인은 부모의 비일관적 양육방식, 과잉간섭적 양육방식, 개인적 변인인 사회기술, 자기통제로 나타났다. 마지막으로 게임과몰입의 변화율에 유의미한 영향을 준 공변인은 부모의 비일관적 양육방식, 과잉간섭적 양융방식, 개인적 변인인 사회기술, 자기통제로 나타났다. 본 연구의 결과를 바탕으로 청소년의 게임과몰입과 고독감에 대한 개입방법과 관련된 시사점을 도출하였다. This study examined the temporal change of game over-indulgence and loneliness and their reciprocal relationships. Latent growth curve models(LGCM) were used to examine the effect of environmental factors (i.e. inconsistent or over-controlling parenting) alongside personal factors (i.e. social skills, self-control and gender) on the initial value and the change rate of game over-indulgence and loneliness. A four-year longitudinal panel data consisting of 848 adolescents were recruited for the purposes of data analyses. Results are as follows. First, both game over-indulgence and loneliness showed linear temporal growth. Second, initial levels of game over-indulgence inversely predicted loneliness over time, while vice versa for loneliness was not significant. Third, inconsistent and over-controlling parenting, social skills, self-control and gender affected the initial value of game over-indulgence. Fourth, over-controlling parenting, social skills, self-control and gender influenced the change rate of game over-indulgence. Fifth, inconsistent and over-controlling parenting, social skills, self-control influenced the initial value of loneliness. Lastly, a number of suggestions for intervention were made.
