홍삼사포닌 Rg3가 함유된 샴푸의 탈모방지와 양모개선 효과

최근 들어 홍삼의 발모효과가 실험적으로 입증이 되었으나 임상적인 효능에 대해서는 아직 연구가 부족한 실정이다. 본 임상시험에서는 홍삼사포닌 Rg3 0.003%가 함유된 샴푸(Somang Co., Korea)의 사용에 따른 탈모방지 및 발모 탈모 및 발모효과를 인체적용시험을 통해 확인하고자 한다. 탈모로 진단된 42명의 환자가 참여하였으며, 홍삼사포닌 Rg3 0.003%가 함유된 헤어샴푸를 사용하는 군과 홍삼사포닌 Rg3 0.003%를 제외한 헤어샴푸를 사용하는 군으로 나누어 16주간 제품 사용 후 모발의 굵기, 밀도, 성장속도를 측정하였고, 탈모 개선 정도의 전문가 육안평가 및 피험자의 주관적 만족도로 평가하였다. 또한 제품사용의 피험자 주관적 기호도를 추가적으로 조사하였다. 시험 결과 시험제품 사용 후 16주에서 모발의 굵기, 모발의 밀도 및 모발 성장속도가 모두 통계적으로 유의성 있게 증가하였으며 전문가 육안평가와 피험자의 주관적 설문평가, 환자의 기호도 면에서도 시험제품이 더 우수한 결과를 보였다. 결론적으로 홍삼사포닌 Rg3가 함유된 샴푸는 피부에 자극 없이 탈모 방지 및 양모개선 효과에 도움을 주는 제품으로 판단된다. Recently, hair growth effect of red ginseng was proved by many in vitro experiments, but there are not enough clinical studies. In the present study, we evaluated the efficacy of Rg3 hair shampoo (Somang Co., Korea) containing Rg3 0.003% extracted from ginseng radix on preventing hair loss and promoting hair growth by clinical study. 42 volunteers were enrolled present study and devided into two groups; one group used the Rg3 hair shampoo and another group used shampoo not containg Rg3. Before and after 8 and 16 weeks use of Rg3 hair shampoo, we measured the density, thickness and growth rate of hair. At the same time, an assessment of improvement of hair loss and subjective satisfaction of patients were conducted by clinicians and patients. Results obtained from the group used Rg3 hair shampoo showed statistically significant improvement in the density, thickness and growth rate of hair. Results from the clinicians, patients and subjective satisfaction of patients also showed better responses for the group used Rg3 hair shampoo. In conclusion, Rg3 hair shampoo is an effective product on preventing hair loss and promoting hair growth.

In this study, ethosomes loaded with Eucommia ulmoides extracts that have anti-inflammatory and anti-aging activities on the skin were prepared as cosmetic materials. The particle size, physical property, and entrapment efficiency of ethosomes to identify their stabilities during 4 weeks were measured. Ethosomes containing 0.01∼0.05% E. ulmoides extracts did not show either precipitation or separation. The mean particle size of these ethosomes was 136.4 nm, and monodisperse. The particle size of ethosome containing 0.05% E. ulmoides extracts was 151.15 nm and the entrapment efficiency was 81.79%. Therefore, in vitro skin permeation experiment was performed with ethosome containing 0.05% E. ulmoides extracts that showed the high loading efficiency and stability during 4 weeks. The order of skin permeation of E. ulmoides extracts was ethosome (77.16%) > liposome (62.80%) > ethanolic solutoin (41.59%). These results suggest that ethosomes containing 0.05% E. ulmoides extracts are effective for the skin permeation and may be used as a cosmetic formulation containing plant extracts.
β-hexosaminidase 분비 억제 및 각질형성세포 분화에 대한 두충(Eucommia ulmoides Oliver) 추출물의 효과

본 연구에서는 두충 추출물이 RBL-2H3 세포의 β-hexosaminidase의 분비 억제와 HaCaT keratinocytes 피부장벽의 회복과 관련한 filaggrin, transglutaminase-1 (TGase-1), cornified cell envelope (CE)의 발현에 미치는 영향에 관하여 연구하였다. β-hexosaminidase 방출 억제 능은 13% 효능을 확인하였고, 피부장벽기능의 회복과 관련된 인자들은 발현과 활성의 정도가 매우 우수한 것을 확인하였다. 각질형성세포의 분화를 판단할 수 있는 CE 측정에서는 두충추출물이 양성대조군보다 더 좋은 효능을 나타내기도 하였다. 따라서 두충 추출물은 β-hexosaminidase 분비 억제에 효과가 있으며, 손상된 피부장벽강화에 영향을 미치는 각질형성세포의 분화 촉진에 효과가 있음을 확인하였다. In this study, Eucommia ulmoides Oliver extracts was studied in order to see any effects on the β-hexosaminidase release suppression of RBL-2H3 cells and on the expression of filaggrin, transglutaminase-1 (TGase-1) and cornified cell envelope (CE) related to the recovery of HaCaT keratinocyte skin barrier. Results showed that Eucommia ulmoides Oliver extracts reduced β-hexosaminidase release in RBL-2H3 cells and increased the effects of Eucommia ulmoides Oliver extract on the expression of filaggrin, transglutaminase-1 (TGase-1) and cornified cell envelope (CE) in HaCaT keratinocytes. Taken together, these results suggested that Eucommia ulmoides Oliver extract may be applicable for keratinocyte differentiation.

Collagen is decomposed by MMP-1, an enzyme induced by transcription factor activator protein 1 (AP-1) and matrix metalloproteinase-9 (MMP-9). Action of MMPs in inflammatory response promotes inflammatory cell movement and secretion, resulting in wrinkles on the skin. After using Bacillus sp. fermentation system and water, Artemisia vulgaris was fermented to prepare different solvent fractions using water, dichloromethane, hex ane, n-butanol, and ethyl acetate. These fractions were used to assess their effects on cell survival, wound healing, MMP-1/MMP-9 and procollagen type I C-peptide (PICP) expression, and skin turnover. MTT assay showed that cell viability of each treated group was 103% to 121%, indicating that A. vulgaris fractions were not toxic compared to control (cell viability: 100%). Wound healing assay revealed that wound healing ability in each treated group was 51% to 61%. This was lower than wound healing area in the control. Using RT-PCR, inhibition of MMP-1/MMP-9 gene expression was examined. As a result, each group treated with fraction showed reduced expression of both MMP-1 and MMP-9 compared to tumor necrosis factor-α (TNF-α) treatment group. Effects on collagen biosynthesis were analyzed using a PICP ELISA Kit. The group in which Artemisia vulgaris was treated increased collagen synthesis from 141 to 262ng/mL compared to the control group. Three-dimensional cell culture revealed that each fraction could increase skin wall formation. These results suggest that each fractions has anti-aging and anti-wrinkle effects on the skin, indicating its suitability as a functional material.
