Mesenchymal stem cells derived from adult bone marrow or adipose tissue are known to be less immunogenic and besides immunosuppressive. But the detail of immunologic effect of adipose-derived stem cells(ADSCs) is not revealed yet. This study is aimed to investigate the effect of ADSCs on lymphocytes proliferation and the production of cytokines in vitro. The ADSCs extracted from omental or subcutaneous fat were expanded during third to fifth passages. The phenotype of the ADSCs was identified by the conventional cell surface markers using flow cytometry: positive for CD29 and CD44, but negative for CD34, CD45, CD117 and HLA-DR. The ADSCs were able to differentiate into the osteoblast or adipocytes with induction media. Third-party ADSCs added to the mixed lymphocyte reactions(MLR) induced overall suppressive effect on lymphocytes proliferation, but in part stimulated, whereas the suppressive effect of ADSCs on the nonspecific lymphocyte proliferation provoked by phytohemagglutinin-A(PHA-A) was totally profound. The stimulated index(SI = [(responder cell + stimulator cell + ADSC)/responder cell + stimulator cell] × 100) was 11.69% to 156.5% in MLR and 0.99% to 14.44% in PHA-A assay. The changes of cytokines production were analyzed from the supernatant collected during the lymphocyte proliferation reactions using ELISA. The production of TNF- α, IL-2, IFN- γ was increased during the MLR and PHA-A stimulation. Adding ADSCs, only the level of TNF- α was decreased in MLR, whereas all of three cytokines was decreased in PHA-A stimulation. The level of Th1 cytokines such as IL-2 and IFN- γ in MLR was significantly increased during co-culture with ADSCs regardless of suppressive or stimulated response. The changes of Th2 reaction such as IL-4, IL-5, IL-13 were minimal and statistically insignificant. The effects of ADSCs on immunologic or nonspecific lymphocyte stimulation are different in extent and cytokine regulation. Further studies will be needed before the clinical application of ADSCs to regenerative medicine.

현재 일부 정부기관에서 정책홍보평가를 별도 용역사업으로 시행하고 있으나 홍보성과에 평가에 대한 평가모형이나 지표가 확립되어 있지 않고 있다. 정부 정책홍보 성과관리 측면에서의 홍보성과의 목적, 과정, 방법 등의 논의에 비해, 정책홍보 효과관리 측면에서의 홍보효과측정 모형과 방법에 대한 연구가 더 필요한 시점이다. 본 연구는 정부 정책홍보 효과관리 모형 개발을 위해, 기존 통합마케팅커뮤니케이션(IMC:Integrated Marketing Communication) 효과측정 이론 중 가장 널리 알려진 이론들이 정책홍보 효과측정 모형화에 어떻게 적용가능한지 알아보기 위해 기존 MacKenzie, Lutz, and Belch(1986)의 구조방정식모형 연구결과를 바탕으로 연구가설을 설정했다. 가설검정 연구결과와 모형적합도 비교분석을 통해, 기존 광고효과과정모형 중 가장 많이 알려진 MacKenzie, Lutz, Belch(1986)의 모형이 홍보효과과정모형 특히 정책홍보효과과정모형에 적용가능함을 알 수 있었다. 이러한 연구결과는 광고효과과정모형을 활용해 정책홍보효과과정모형을 개념화하고 홍보효과의 정교화된 측정지표를 제시하였다는 점에서 의의가 있다. Currently a few government agencies conducted the evaluations on policy public relations; however, it was found that the evaluations lack in the evaluation model and scales. Thus, government management of policy public relations need more elaborated discussion of setting PR goals, process, methods for improving the effectiveness and accomplishment. This study applied to MacKenzie, Lutz, and Belch(1996)"s model of advertising effect evaluations for government policy public relations based on structural equation model method. Based on the survey method, the results showed that policy knowledge and policy preference directly influenced the policy preference, which also directly influences the policy acceptance. It is well established that the advertising evaluation is well suited to evolution of policy public relation process. This study contributes to the elaboration of public relations process and model in government public relation evaluation.
