우리나라 양식 자주복, Takifugu rubripes (Temminck & Schlegel, 1850)에서 발생한 Vibrio atlanticus의 첫 감염 사례 보고

본 연구는 육상 순환 양식장, 약 $20^{\circ}C$의 수온에서 양식 중이던 자주복에서 체표에 백탁과 궤양을 보이는 개체가 다수 발생하여 그 원인을 밝히고자 하였다. TCBS 선택배지에서 순수 배양된 균체를 확인하고 계대 배양하여 16S rRNA와 rpoA 유전자의 염기서열 분석을 실시하였으며, 그 결과는 Vibrio atlanticus와 99.77%의 상동성을 가지는 것으로 나타났다. 생화학적 특징을 이미 보고된 같은 균종과 비교한 결과, Voges-Proskauer test와 Gelatin 가수분해능에 대한 결과에서 차이를 나타내는 것으로 확인하였다. A case of disease was occurred in tiger puffer, Takifugu rubripes, reared in circulated system (water temperature was about $20^{\circ}C$) on land in Korea. Diseased fish showed a unique external sign, the cloudy skin ulcer in various sizes. We investigated the cause of disease, and isolated one pure cultured bacterium, TP01. This had 99.7% homology with Vibrio atlanticus by sequence analysis of the 16S rRNA and rpoA genes. TP01 showed some differences with other reported V. atlanticus strains in the biochemical characteristics as like Voges-Proskauer test and gelatin hydrolysis.

On April of 2000 and February of 2001, a disease characterized by darkening, ascites and enlargement of the spleen occurred in flounder, Paralichthys olivaceus, farmed near northern and eastern coasts of Korea. Various degree of necrosis including liquefactive-type was observed mainly in hemopoietic tissue of spleen and kindney. Certain tissue such as branchial soft tissue including epithelium showed a similar necrotic findings. But some tissues such as intentine had several necrotic clusters of wandering cells, being possibly leukocytes. Necrotic evidences of blood cells were evident in various tissues including hemopoietic tissues. Necrobiotic bodies were frequently encountered in blood smear preparations. Bullet type-virious were detected in the spleenic cells. Each virion was about 500 nm in diameter and 170nm in length. From these results, this disease was thought to be a rhabdoviral disease.
관상용 금붕어(Carassius auratus)의 비결핵항산균 감염증에 관한 고찰

In October 2021, goldfish (Carassius auratus) that reared in an aquarium continued to die and the cause analysis has begun. For diagnosis, biological, molecular and histopathological analysis were conducted, and samples are divided with symptomatic and asymptomatic objects. The 16s rRNA target region was identified from tissue (kidney+spleen) sample of asymptomatic fish, and the sequenced result shown 100% homology with Mycobacterium fortuitum. In histopathological studies, various size of granuloma and inflammatory regions are observed in kidney tissue of all tested fish, and acid-fast bacteria staining positive reactions are observed inside or outside the granuloma. In order to diagnose mycobacteriosis, accurate preparation is required along with various diagnostic methods, and more attention is needed in handling aquarium fish management to prevent mycobacteriosis.
가물치에 대한 Nocardia seriolae의 인위감염과 조직학적 관찰

Snakehead, Channa argus were intraperitoneally infected with Nocardia seriolae. at the concentrations of 1.5×10<SUP>7</SUP> cfu/㎖ or 1.5×10<SUP>8</SUP> cfu/㎖. The infected fish were kept in aquaria at 25℃ and 30℃ for 3 weeks. Clinical signs and mortality were monitored daily to evaluate the virulence. All artificially infected fish showed the same clinical sign found in naturally infected fish. All the fish infected with 1.5×10<SUP>8</SUP> cfu/㎖ of N. seriolae died within 24days. N. seriolae showed higher virulence to snakehead at the temperature 30℃. Internal lesions such as whitish nodules in the infected internal organs were not correlated with mortality but some degenerative changes were observed in all the infected organs within a week. Whitish nodules in the infected organs which are the typical character in nocardial infection was initially found at two weeks after the artificial infection in snakehead.

Recently, import supply of glass eels from Philippines has been increased in South Korea, and the species distinction was very important to import traders and Anguilla marmorata and A. bicolor pacifica are major species at Philippines. In this study, the species distinction and the rate were researched targeting imported glass eels from the estuaries of North Luzon and the South Mindanao in May and July, 2014. Dorsal fin position and pigmentation pattern at caudal fin are the trait standard for species distinction. The results about morphological identification were coincide with the result of the genetic identification, and the regional distribution of A. marmorata rate was over 90% at North Ruzon in May. However, the rates of A. bicolor pacifica and A. mamorata were 63.3% and 36.7% in May, and were 53.3% and 46.7% in July respectively at South Mindanao.

This study about morphological observation of pre- or post ovulated eggs obtained from artificially maturated female eels. Female eels were divided with two groups as wild eels from nature and feminized eels from farm. Artificial maturation had been conducted with the established methods in this laboratory, and then maturate eggs sampled at fixed 3 times and these were observed with stereomicroscope. Rate of increased body weight (RIW) were measured with 2 times. Egg diameters and development of oil droplets are determined for standardization of egg maturation degree, and the transparency of egg cytoplasm and the homogeneous degrees of egg size were referred to determine of egg quality. Rate of increased body weight (RIW) were good in range about 10 % at final salmon pituitary extracts (SPE) injection time and in range about 20 % at 17α, 20β-dihydroxy-4-pregnen-3-one (DHP) injection time. Good matured egg for fertilization had 900-1000 ㎛ diameter, and they had about 50 oil droplets in size about 100 ㎛ diameter. There were not predominant differences at egg size and oil droplet development between wild female and feminized eels.
나일틸라피아의 임파조직내 대식세포 분포변화에 관한 병리조직학적 연구

To elucidate the distributional pattern of macrophages within lymphomyeloid tissues according to the disease process, tilapias, teleostean fish, were intraperitoneally injected with live Edwardsiellatarda and its extracellular product(ECP) respectively. And then histopathological examination for the spleen and gead kidney were carried out for the individuals which had not any clinical signs. In the group injected with live E. tarda, macrophages were densely organized into MMC-like structures with showing some degree of recovery in histological arrangement. At the 2nd week, overall structures of the lymphomyeloid tissues became normal, accompanying the disappearance of most of macrophage groups. Also in case of ECP injection, quite similar findings were observed. Moreover, macrophage collections and hypertrophied ellipsoids were recognized at 1hour after the injection of ECP in head kidney and pleen, respectively. These results suggested that characteristic behaviors of macrophages in lymphomyeloid tissues would be used as important morphological criteria for early diagnosis of edwarsiellosis or possibly of other infectious diseases.

Hot water extracts of 5 herbs were tested in-vitro for anti-bacterial, anti-fungal, anti-parasitical effects for possible use against fish diseases. Skullcap, Scutellaria baicalensis, was the most effective herb in these 5. The effects of skullcap in cultured flounder were examined for various physiological responses and bacterial disease-prevention follow as feeding skullcap absorbed diet for 4, 8, 12weeks. There were not any significant effects in physiological responses, except beneficial action of growth promotion. No definitive preventive activity was observed with experimental feeding of the extract against infected flounder. As we could not confirm in-vivo antibacterial effects of skullcap in flounder despite its in-vitro efficacy, further studies are needed to define the in-vivo efficacy.
해수 순치 중의 뱀장어, Anguilla japonica에서 나타나는 Vibrio scophthalmi 감염증

Vibriosis is one of the most prevalent fish disease belonging to the genus Vibrio. In present study, Vibrio sp. isolated from Japanese eel was confirmed as Vibrio scophthalmi using analysis of the genomic sequence of 16S rRNA. The major signs were hemorrhage of body surface and inner surface of abdomen, severe enteritis and retention of ascitis. Histopathological examination revealed blood cell degenerations in various organs (gills, liver, spleen, kidney, heart, intestine), exfoliate of intestinal epithelium, and congestion and hemorrhage in intestinal lamina propria. This is the first case report on V. scophthalmi infection in Japanese eel Anguilla japonica.
PCR 기법을 이용한 2009년 우리나라 서해안과 남해안 바지락, Ruditapes philippinarum의 Perkinsus olseni 감염에 관한 보고

Prevalence of a protozoan parasite Perkinsus olseni in Manila clam Ruditapes philippinarum was surveyed from July to December 2009 on the west and south coast of Korea. P.olseni infection was diagnosed using two primer sets, P.olseni NTS Forward/P.olseni NTS Reverse set and PolsITS-140F/PolsITS-600R set in polymerase chain reaction(PCR). The results using PolsITS-140F and PolsITS-600R primer set was retained up to 60% at all stations from July to December, except for Padori. Especially, Goheung showed 100% prevalence from October to December. The results about comparison of the 4 station`s DNA sequences which were analyzed from PCR products(457bp) using PolsITS-140F and PolsITS-600R primer set, there were only 2base differences at Sunjedo.
