        Simulation of sloshing in a bi-lobe tank under arbitrary rotation using the FDS scheme and the HCIB method

        Three-dimensional sloshing in a bi-lobe tank under arbitrary rotation is simulated using a code developed using the flux-difference splitting scheme for variable density incompressible fluids and the hybrid Cartesian/immersed boundary method. The material interface is regarded as a moving contact discontinuity and is captured using a free surface capturing method derived from the Riemann solver,without any additional treatment along the interface. The boundary condition for the arbitrary motion of the bi-lobe tank, which contains a thin partition between two partially overlapping cylindrical tanks, is handled with ease by using the hybrid Cartesian/immersed boundary method. The computed time evolution of the interface is compared with the snapshots taken during the experiments on sloshing caused by the sway motion of the bi-lobe tank. Good agreement is observed between the computational and experimental results. The validated code is used to simulate three-dimensional sloshing in the bi-lobe tank that is subject to combined pitch and roll motions. A rotational vector is used to locate the Lagrangian points of the unstructured surface grid according to the motion of the tank. Grid independence tests are carried out using three different size grids. Violent three-dimensional sloshing ensues with an increase in the angular velocity of rotation.

        파랑중 선박의 운동을 고려한 갑판적 컨테이너의 Securing 및 Lashing 하중 계산 (I)

        파랑중을 운항하는 선박은 항상 파도의 영향을 받아 운동을 하게 된다. 특히 컨테이너선과 같이 갑판위에 화물을 많이 운송하는 선박의 경우에는 여러 방향의 운동형태 중에서 횡동요가 화물의 안전에 가장 큰 영향을 미친다. 이러한 선박에서는 컨테이너의 이동 및 전도를 방지하기 위하여 Securing과 Lashing을 하게 되는데, 적절한 시스템의 선정을 위하여 각각에 작용하는 하중들의 정확한 평가가 요구되고 있다. 따라서 본 논문에서는 갑판적 컨테이너들의 연결부에 작용하는 Securing 하중과 Lashing 와이어에 작용하는 힘에 대한 계산식을 유도하고, 몇 가지 Lashing 패턴에 대하여 계산을 수행하였다.       • Improvement of Position of Label (POL) Influence by Fluorescently-labeled Helper Probe

        DNA microarray-based technologies provide for high-throughput methods and the simultaneous analysis of a large number of genetic information at one assay. However, several important aspects of microarray technology such as the selection of optimal oligonucleotide probes and improvement of hybridization kinetics still have to be resolved. Recently, Lei Zhang et al. proposed that one of the important things in applications of a short oligonucleotide microarray is the position of label (POL) effect. In this study, we fabricated a microarray for the identification of polymorphisms in human mitochondrial DNA hypervariable region II (HVII), as well as a fluorescently-labeled helper probe applied to a fabricated microarray. In conclusion, we report on the results obtained with additional fluorescently-labeled oligonucleotide probes, called helper probes, in an attempt to improve the relative POL influences. Microarray results were improved and reduced the POL effect. These results may contribute to better hybridization patterns, data interpretation, and a new aspect for the selection of an efficient probe in microarray applications.

        선박의 운동 운항환경 모니터링 시스템 개발

        A ship in a sea cruises with rolling, pitching, heaving etc because of environmental causes such as wind and wave. Those motions make crews or passengers feel inconvenience and they feel acceleration changes. Therefore, if lateral and vertical accelerations can be measured at a specific position in a ship, it can be known how discomfortable crews or passengers are. The motion monitoring system developed in this paper consists of measuring and communicating part including five accelerometers and gyro and a main computer which acquires measuring data and calculates motion indices. MSI(Motion Sickness Incidence) and MII(Motion Induced Interrupt) are calculated in real time using measured acceleration and angular rate. The validity of the developed system was confirmed through the real ship test of Hannara which is the school ship of Korea Maritime University. 바다에서 운항하는 선박은 바람과 파도 등의 외란 때문에 횡동요(Rolling), 종동요(Pitching), 상하동요(Heaving) 등의 운동을 하게 되며, 이러한 운동은 가속도 형태로 승객이 느끼게 된다. 따라서, 선박내의 특정 지점에서 좌우방향, 상하방향 가속도와 각속도 등을 계측하면 선원 또는 승객이 선박 운동에 어느 정도 피폭되었는가를 알 수 있다. 본 연구에서 개발한 운동 운항환경 모니터링 시스템은 4개의 가속도계와 가속도계, 자이로가 포함된 관성 자세계측장치, 데이터 취득장치를 포함한 계측 및 통신부, 중앙에서 데이터를 관리하고, 운항환경 지수를 계산하는 전산기로 구성되고, 계측된 가속도와 각속도를 이용하여 운항환경을 나타내는 정량적 지수인 뱃멀미 지수(Motion Sickness Incidence, MSI), 운동유발 작업방해회수(Motion Induced Interrupt, MII)를 실시간으로 계산한다. 개발된 시스템은 한국해양대학교 실습선인 한나라호의 부산-목포, 부산-제주 연안항해시 실선시험을 통하여 유효성을 확인하였다.

        파랑수면 근처에서 항주하는 수중운동체의 운동해석

        A cylinder-type underwater vehicle for military use that is running near the free surface at the final homing stage to hit a surface ship target is affected by wave force and moment. Since wave can affect an underwater vehicle running at the depth less than half of the modal wave length, it is important to confirm that the underwater vehicle can work well in such a situation. In this paper, wave force and moment per unit wave amplitude depending on wave frequency, wave direction, and vehicle's running depth were calculated by 3-Dimensional panel method, and the numerical results were modeled in external force terms of six degrees of freedom equations of motion. Motion simulation of the underwater vehicle running in various speed, depth, and sea state were performed.

        Development of simple and rapid HLA-C genotyping method using an oligonucleotide microarray

        Human Leukocyte Antigen (HLA), called human Major Histocompatibility Complex (MHC), performs the crucial function of antigen presentation in human immunity, and correctly matched HLA antigens improves survival and reduces immunological complications. Therefore, a precise HLA typing method is necessary for successful transplantation between donor and recipient. Until now, many methods have been developed for HLA typing, including serological and DNA-based methods. However, these previous methods have been somewhat limited due to their requirements for a large number of samples and complex experimental procedures. To resolve this problem, in this study, we developed rapid genotyping methods for HLA-C using an oligonucleotide microarray. To evaluate the performance of the HLA-C genotyping microarray, we analyzed 50 unrelated Korean individuals. The genotyping results obtained by the oligonucleotide microarray were identical with those of PCR-SBT and PCR-SSPs, at a low-resolution, 2-digit level. Therefore, this microarray method for genotyping HLA constitutes a rapid, simultaneous, and high-throughput method that can be used as an alternative to current methods.

      • Development of Salmon Identification DNA Chip based on Mitochondrial COIII-ND3-ND4L Variations

        Salmon are born in fresh water, migrate to the sea, and then return to their natal river for reproduction. This feature has been utilized for their enrichment as commercial resources because the artificial rearing and release of fry and the catching of returning salmon become easy tasks. For proper management of salmon resources and fair trade of salmon products, a reliable identification of salmon species is necessary. However, species identification based on morphological features has often been inaccurate and time consuming in many cases. In particular, for processed salmon products in the market, the application of morphological features toward species identification is impossible. To resolve this problem, we have developed a DNA chip that distinguishes 9 species of salmonid fishes. DNA sequence variations in the mitochondrial DNA COIII-ND3-ND4L region were utilized for designing species-specific oligonucleotide probes. The probes were synthesized and covalently linked onto a glass slide in duplicate. Hybridization of the probes with single-stranded DNA target molecules labeled with a fluorescent dye produced species-specific hybridization spot patterns. The whole process of species identification took only about 4 hours. This DNA chip system provides a sensitive, objective, and high-throughput method for salmonid species identification, being especially useful for processed products in the market. This technique can be easily applied to other fish species.
