Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on ${\beta}-glucuronidase$. Among water extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepatoprotective effect on $CCl_4-induced$ hepatotoxicity of rats. These results suggest that the ${\beta}-glucuronidase$ seems to be closely related to the liver injury, which could be prevented by the inhibitor of ${\beta}-glucuronidase$.

Propolis is a lipophilic, natural product prepared by mixing the exudates collected from various plants by honeybees with beeswax for the purpose of using to seal hive walls and to strengthen the borders of combs. Because of its versatile bioactivities, propolis has been attracting many investigators` interest. But the pharmacological studies on propolis has, to date, been exclusively performed for an alcohol extract, there is few information of water extract. Therefore, in this study, we investigated the various effects of water extract of Chinese propolis. The water extract of propolis and its fractions of organic solvents showed strong antioxidative activities, especially ether and ethylacetate fractions, and reduced the lipid peroxidation of rat liver in vitro. Additionally the ether fraction of propolis (10 mg/ml) inhibited the activity of hyaluronidase by 50%. In vivo, the water extract of propolis considerably decreased s-GOT, s-GPT and s-LDH activities which represent for the hepatotoxicity induced by CCl₄4 in rats, and prolonged the MST (Medium sevival tinme) and ILS (Increasing in MST over control) by 18% in mice which inoculated with sarcoma 180 ascites cells. These results suggest that the water extract of propolis has various bioactivities as well as the alcohol extract.

Bioequivalence of two flouxetine capsule, Prozac® (Lilly korea Ltd.) and Lanclic® (Samsung Pharm. IND. Co.), was evaluated according to the guideline of KFDA. Twenty four healthy male volunteers (21-26 years old) were divided into two groups and a randomized 2×2 cross-over study was employed. After 60㎎ of fluoxetine was orally administered, blood was taken at 1, 2, 3, 4, 5, 6, 7, 8, 10, 12, 24, 32, 48 and 72 hours after administration and just before administration. Plasma was analyzed for fluoxetine and internal standard (clomipramine) by a sensitive and validated HPLC assay. The pharmacokinetic parameters (AUC_(t), C_(max) and T_(max)) were calculated and ANOVA test was used for the statistical analysis of parameters. Differences in AUC_(t), C_(max) and T_(max) between two capsules were -0.90, 3.46 and -14.08% respectively. All powers (1-β) for AUC_(t), C_(max) and T_(max) were more than 0.9. Detectable differences (Δ) and confidence interval were all less than ±20%. All the parameters above met the criteria of KFDA for bioequivalence and indicated that Lanclic® capsules are bioequivalent to Prozac® capsules.

β-Glucuronidase-Inhibitory Activity and Hepatoprotective Effect of Ganoderma lucidum

To prove the relationship between the fluctuation in serum β-glucuronidase level and hepatoprotective activity was investigated. The ether fraction of G.lucidum, which had potent β-glucuronidase-inhibitory activity, protected against CCI₄-induced liver injury. From this ether fraction, ganoderenic acid A, was isolated as the potent inhibitor of β-glucuronidase. It had a potent hepatoprotective effect against CCI₄-induced liver injury. These results suggest that the glucuronidase seems to be closely related to liver injury, which could be prevented by glucuronidase inhibitors.
