Monascus sp. CS-2가 生産하는 黃色色素에 關한 硏究 : (第3報) 黃色色素의 安全性 試驗 (Part3) Safety Test of Yellow Pigment

분리 정제한 黃色色素의 安全性을 검토하기 위여 in vivo 및 in vitro 시험을 한 결과는 다음과 같다. 1. Mouse를 이용한 安全性 시험은 經口 투여시 LSD_50은 체중 20g당 0.13245g이었다. 2. 본 色素를 이용한 발열성물질 시험결과 발열 한도량은 體內투여시 ㎏당 5㎎이었다. 3. 본 色素를 이용한 histamine물질 시험은 혈압강하물질 표준품으로 비교할 때 시험동물 ㎏당 10㎎까지 安定하였다. 4. 본 色素의 병원성菌에 대한 감수성 시험결과 500mcg/㎖ 농도에서 Bacillus subtilis (ATCC 6633), Sarcina lutea (ATCC 8341) 및 Staphylococcus aureus (ATCC 6538-P)에 대하여 미량의 저해 작용을 나타내었다. Safety of yellow pigment produced by Monascus sp. CS-2 was evaluated. Acute oral toxicity, pyrogen test, and histamine test, as well as antimicrobial activity were determined. The results obtained were; LD oral in mice was 132.5㎎/20g, pyrogen test in rabbit was 5㎎/㎏, and histamine test in cat was 10㎎/㎏. Also the pigment was particularly sensitive to Bacillus subtilis (ATCC 6633), Sarcina lutea (ATCC 9341) and Staphylococcus aureus (ATCC 6538 P), whereas not sensitive to Pseudomonas pyosyanea (ACTC 10490), Bacillus var. mycoides (ATCC 11778), Bordetella bronchiseptica (ATCC 4617) and Staphylococcus epidermidis (ATCC 12228).

Growth responses of Brevibadterium ammoniagenes BA 17-2, which had been obtained by the treatment of several mutagens in our previous report, were investigated to select the preliminary optimal concentrations of phosphate, Mg^(++), Mn^(++) and thiamine for the production of 5'-XMP aminase. In this experiment it was shown that the concentration of phosphate in the medium has an important effect on the growth of microorganism. Using the medium containing 0.2% of MgSO₄·7H₂O, 3㎎/ℓ of MnSO₄·H₂O, and 1㎎/ℓ of thiamine-HCl, the maximum cell mass was obtained at the concentration of 0.4% of KH₂PO₄ and KH₂PO₄, respectively. Above the concentration of these phosphates, cell growth was inhibited as the phosphate concentration increased to 1?0, but the inhibition was overcome by the addition of 1% of MgSO₄·7H₂O and 3㎎/ℓ of thiamine-HCl. The 5'-XMP aminase activity was also influenced by the concentration of phosphate, Mg^(++), Mn^(++), and thiamine. In addition, the optimal culture pH and temperature for the enzyme activity were found to be 6.8 and 32℃, respectively.
