소의 뇌하수체 조직에서 분리한 mRNA로부터 소 성장호르몬 cDNA를 합성 분리하였다. 분리된 bGH cDNA는 mRNA product의 2차 구조를 변화시킬 목적으로 N-terminal 부위를 변형시킨뒤,pIN III-Al 플라스미드에 삽입시켜 대장균내에서 발현할 수 있는 재조합 플라스미드 pIG-B를 제조하였다. 플라스미드 pIG-B를 갖는 E. coil JM103은 N-terminal이 변형된 소 성장호르몬을 생성하였다. 또한 분리된 bGH cDNA를 probe로 사용하여 소의 뇌하수체 조직으로부터 소 성장 호르몬 유전자를 분리하였다. cDNA was prepared from $poly(A)^+$ mRNA isolated from pituitaries and cloned in E. coli. It contains the coding sequence for bovine growth hormone(bGH). The cloned bGH cDNA was confirmed by colony hybridization, restriction enzyme mapping, and nucleotide sequencing. The cloned cDNA was modified to construct the plasmid that expresses the bovine growth hormone in bacteria. A genomic gene of bovine growth hormone was also cloned using the bGH cDNA as a probe.
