        유용림목의 (有用林木) 동위효소에 관한 조림 (造林) 및 유전학적 연구 - × Pinus taeda · rigida 와 P . densiflora 에 있어서 Leucine Aminopeptidase 와 Peroxidase 동위효소의 유전 -

        Twelve Pinus taeda were used as mother trees and one P. rigida plus tree was used as pollen tree for 12 cross combinations. Nine P. densiflora plus trees (4 mother and 7 pollen trees) were used as parents for 11 cross combinations. Those parents and 10 progenies were analyzed for LAP of ×P. taeda·rigida hybrid and P. densiflora and for peroxidase of ×P. taeda·rigida. The analysis, based on the banding patterns, indicate three alleles for LAP-A locus (A1, A2, A3) and two alleles for LAP-B locus (B1, B2) in ×P. taeda·rigida hybrids. Chi-square test on the segregation for progenies did not show significant differences. The results indicated good agreement with monohybrid Mendelian inheritance. Independence test for occurrence frequency of 2 alleles (LAP-A3, LAP-B2) illustrated that there is neither linkage nor repulsion relationship between LAP-A3 and LAP-B2 alleles. Three band at LAP-A locus were always exhibited from all parents and their progenies of P. densiflora. However, the occurrence of two bands at LAP-B locus was variable, one bands assumed as homozygous alleles (B2/B2) and two bands as heterozygous alleles (B1/B2). The segregation ratio for progenies of P. densiflora suggested that LAP-B locus may be controlled by two alleles (B1 and B2). Three Peroxidase loci (Px-A, Px-B, Px-C) assumed to be controlled by allozyme in ×P. taeda·rigida hybrid. The Px-B and Px-C loci could not find out the variations from banding patterns of parents and their progenies, while the Px-A locus showed the variations of occurrence frequency by two bands. The segregation ratio for A1/A2 at LAP-A locus suggest that the peroxidase allozymes of ×P. taeda·rigida hybrid appeare to be monomeric products; that is, Px-A locus may be controlled by two alleles (A1 and A2).

        비파 부위별 에탄올 추출물의 생리활성

        비파의 소비촉진 및 부가가치를 향상시키기 위하여 2007년6월 전남 완도군에서 재배중인 비파‘전중(田中)’을 수확하여 부위별(잎, 과육 및 씨) 에탄올 추출물의 생리활성 효과를 조사하였다. 총 페놀함량은 잎(60.6㎍·mL<SUP>-1</SUP>), 종자(22.9㎍·mL<SUP>-1</SUP>)그리고 과육(19.4㎍·mL<SUP>-1</SUP>)순으로 많았다. 추출물의 농도가 500㎎·mL<SUP>-1</SUP>일 때 부위별 전자공여능은 잎 추출물(91.8%), 종자 추출물(25.0%), 과육 추출물(14.2%)순이었다. 아질산염 소거는 추출물의 농도가 500㎎·mL<SUP>-1</SUP> 일 때 잎추출물은 73.0%, 과 육추출물은 66.9%, 종자 추출물은 64.9%를 나타내었다. Tyrosinase 활성 저해 효과는 과육, 잎,종자 추출물 순으로 높게 나타났는데,과육 추출물 1,000㎎·mL<SUP>-1</SUP>에서는 16.9%, 4,000㎎·mL<SUP>-1</SUP>에서는 33.3%의 저해율을 나타내었다. 항균활성은 잎 추출물에서 높게 나타났는데, 그 중 Streptococcus mutans 균에서 저해환의 직경이 12.1(500㎎·mL<SUP>-1</SUP>)∼15.4㎜(2,000㎎·mL<SUP>-1</SUP>)으로 크게 나타났다. This study was conducted to promote the consumption and to improve the values of the loquat (Eriobotrya japonica Lindl., cv. “Jeonjung”). We also harvested loquat grown in Wando-gun, Jeonnam Province, on June 10, 2007, and examined its physiological activities with the ethanol extracts from different plant parts. Total phenol content was high in the order of leaves (60.6 ㎍·mL<SUP>-1</SUP>), seeds (22.9 ㎍·mL<SUP>-1</SUP>) and fruits (19.4 ㎍·mL<SUP>-1</SUP>). When the concentration of extracts was 500 ㎎·mL<SUP>-1</SUP>, leaves showed highest DPPH (α,α-diphenyl-β-picryl-hydrazyl) radical scavenging activity (91.8%) and followed by seeds (25.0%) and fruits (14.2%). And nitrite scavenging activity was high in the order of leaves (73.0%), fruits (66.9%) and seeds (64.9%). Fruit extracts showed highest mushroom tyrosinase inhibition activity, especially the activity at 4,000 ㎎·mL<SUP>-1</SUP> extracts was 33.3%. The highest anti-microbial activity exhibited in the leaf extracts, especially showing the largest inhibition diameter (12.14 ㎜) against Streptococcus mutans at 500 ㎎·mL<SUP>-1</SUP> extracts.

        수확시기 및 저장온도에 따른 비파의 저장력

        비파 수확기 및 저장온도에 따른 저장중 품질변화를 조사한 결과를 요약하면 다음과 같다. 저장중 과실 경도는 처리간 차이가 없었으나 외관은 수확기가 늦을수록, 온도가 높을수록 저하되는 경향을 보였다. 식미는 수확기가 빠를수록 저장온도가 높을수록 감소하는 경향을 보였다. 과피의 Hunter L* 및 b* 값은 저장후기 심하게 감소하는 경향을 보였는데 수확시기나 저장온도에 따른 차이는 없었다. 반면, a* 값은 수확시기에 따라 심한 차이를 나타냈다. 부패과율은 수확시기가 빠를수록, 저장온도가 높을수록 높은 경향을 보였다. 호흡량 및 에틸렌농도는 수확시기가 빠를수록, 저장온도가 높을수록 높았다. 아세트알드하이드와 알콜함량은 6월 21일 수확과와 0℃ 저장과에서 낮아지는 경향을 보였다. 비파를 수확 적기(6월 21일)에 수확해서 0℃에 저장시 외관증진과 함께 부패과율 감소로 저장력이 증진되었다. This experiment was conducted to examine the effects of harvest date (June 17, June 21, and June 25) and storage temperature (0, 5, 10℃) on the shelf life in loquat fruits (Eriobotrya japonica Lindly cv. Tanaka). Changes in flesh firmness were not different among treatments, but visual appearance was affected by harvest date and storage temperature. Sensory value decreased with storage time in fruits harvested early and stored at high temperature. Skin Hunter L* and b* values significantly decreased at the end stage of storage regardless of harvest dates or storage temperatures, while a* value increased with delaying harvest date. Decay rate increased with delaying harvest date and rising storage temperature. Ethylene and CO₂ concentrations were significantly increased in early harvest (June 17) and at high storage temperature (10℃). Acetaldehyde and ethanol concentrations were reduced when fruits were harvested on June 21 and stored at 0℃. Storage of fruits harvested on June 21 at 0℃ extended the shelf life by improving visual quality and reducing fruit decay.

        한국의 대차수(大茶樹)

        Nine large Korean wild tea trees, Tongdosa, Whagea, Milyang, Gule Tojimyn, Bongsan, Duklang, Boriam, Whaumsa were measured the hight and collar diameter from 1995 to 2008. Among them the largest hight is 4.2m at Dosim tea plantation of Whagea, and the biggest collar diameter is 32.0cm at the Boseong Tea Museum. There are variance from 4.5cm to 32.0cm at collar diameter and from 0.5m to 4.2m at hight among trees. It is very difficult to measure the growth years because the plant growth is represented by growth environments for example soil quality, water conditions etc. It is simply can checked to be measured the ring of year on the wood disc. The stem of 5.0 cm branch of Dosim plantation at Whagae was measured the 32 year rings. Therefore the years of the 5.0 cm branch was estimated 34 years old by added 2 years of pith growth. On the basis of this data, the age of the largest tree in this place is estimated approximately 100 years. Out of these places, three places, Tongdosa, Whaumsa, and Boriam, have the long period of tea culture and history. With the tea cultural studies, the scientific studies are required continuously for an authentic evidence.

        하드 아노다이징 (N) EV 경량화 브레이크 디스크 성능 평가

        The power of the vehicle is rocking epicenter occurred. The first cause is carbon dioxide emissions owing to environmental changes on Earth due to the global warming disaster, second is usage of raw material of various internal combustion engines. In this paper, focusing on electric vehicle, especially the light disc rake system is condidered to be a fairly important brake system. High proportion of the weight of the brake disc is occupied by car. Therefore, the lightweight brake disc is urgently required in the car. Due to increasing traffic accident and an upsurge of vehicles with braking devices, the importance of braking device development is fairly significantly ahead. In the present study conventional cast iron brake disk was replaced with a lightweight aluminum parts in hard anodizing. By the use of hard anodizing of aluminum 6061 series through minimizing the inconvenience by a little faster dissemination of electric vehicle through the design of light weight disk.

        Isoperoxidase 변이형에 (變異型) 의한 소나무 Clone 감별

        Using of zymography for identification of same clone of Pinus densiflora, the two year old needle leaves of 48 ramtes including 8 clones (6 ramets per clone) were collected in the clone bank which has been established on the near campus of Institute of Forest Genetics, Suwon, in 1962. All 8 bands are named from cathod to anode, G, H, K, M, N, Q, S and Y. Only CB-1 clone shows all bands, while KW-3 clone reveals only 5 bands. Other 6 clones were found 7 bands but the occurred frequencies of those bands are variable among those clones. Though the grafting stock are used various individuals grown seed propagation of P. densiflora, moreover, three stocks have been different species and that one has been P. rigida and two individuals have been P. koraiensis, the zymograms of the ramets belonging to the same clone reveals the identified patterns. The results show that the stocks for grafting have not been affected on the isoperoxidase patterns of their scions in P. densiflora. Among 48 ramets of 8 clones, 4 ramets are found the different isoperoxidase patterns from that of the remained rametes within same clone. Thus, it is conluded that zymography is useful for testing genuineness of the grafted clones of P. densiflora.

        This paper proposes a new training system for equilibrium sense and postural control using unstable platform and force plate. This system consists of unstable platform, force plate, computer interface, software and the computer. Using this system and training programs, we perform the experiment to train the equilibrium sense and postural control of subject. To evaluate the effects of balance training, we measured some parameters such as the maintaining time in the target, the moving time to the target and the mean absolute deviation of the trace before and after training. The result shows that this system can improve the equilibrium sense and balance ability of subject. This study shows that proposed system had an effect on improving equilibrium sense and postural control and might be applied to clinical rehabilitation training as a new effective balance training system.

        기존의 첨단주택은 IT 차원에서 접근되어 에너지 절약을 위한 기술은 시도하지 않았고 에너지 환경관리 기술이 미흡하였다. 본 논문에서는 실제 사용자 생활 패턴을 파악하여 에너지 소모를 얼마나 줄일 수 있는지 테스트 하였다. 복도, 출입구 그리고 현재 근무 중인 테이블에 조도 센서와 인체 감지 센서를 설치하였는데 조도 센서와 인체감지 센서는 하나 의 그룹을 이루며 생활패턴을 파악하고 조도 값에 따라 실내등을 소등할 수 있도록 한다. 모든 센서 노드는 스타 토폴로지(one-hop) 형태로 연결되며 데이터는 USN 브릿지에 있는 베이스노드로 수집되고 이더넷을 통해 서버로 전달하면 DB에 저장된다. 그러므로 실시간으로 실내 환경 데이터를 수집하여 실내 환경 통합 감시 및 모니터링이 가능하고 에너지 소 모를 줄일 수 있다.

        타액내 구강질환 원인 균의 세균배양법, SYBR green qPCR법, MRT-PCR법 간의 정량분석

        Objectives: The purpose of this study was to compare SYBR Green qPCR, TaqMan, and bacterial selective medium cultures for accurate quantitative analysis of oral microorganisms. Methods: The SYBR Green method is widely used to analyze the total amount of oral microorganisms in oral saliva. However, in this study, MTR-PCR method based on TaqMan method was performed using newly developed primers and probes. In addition, it was designed to confirm the detection agreement of bacteria among bacteria detection method. Results: As a result of MRT-PCR and SYBR Green qPCR analysis, more than 40 times (0.9-362.9 times) bacterium was detected by MRT-PCR. In addition, more bacteria were detected in saliva in the order of MRT-PCR, SYBR Green qPCR, and bacterium culture, and the results of MRB-PCR and SYBR Green qPCR showed the highest agreement. The agreement between the three methods for detecting P. intermedia was similar between 71.4 and 88.6%, but the agreement between MRT-PCR and SYBR Green qPCR was 80% for S. mutans. Among them, the number of total bacteria, P. intermedia and S. mutans bacteria in saliva was higher than that of SYBR Green qPCR method, and bacterium culture method by MRT-PCR method. P. intermedia and S. mutans in saliva were detected by MRT-PCR and MRT-PCR in 88.6% of cases, followed by the SYBR Green qPCR method (80.0%). Conclusions: The SYBR Green qPCR method is the same molecular biology method, but it can not analyze the germs at the same time. Bacterial culturing takes a lot of time if there is no selective culture medium. Therefore, the MRT-PCR method using newly developed primers and probes is considered to be the best method.
