        넙치(Paralichthys olivaceus)의 활어 수송 시 수온, 염분 및 마취제 영향

        Transport is essential in the farming process of farmed fish and is one of the physical stress factors such as sorting. The effect of water temperature and anesthesia during low salinity transport was confirmed. In the experimental group, the water temperature was set to 20℃ (natural water temperature, NWT), 15℃ (cooling water temperature, CWT) respectively, in water with a salinity concentration of 35‰, 15‰ and an anesthetic (anesthesia, Anes., Sigma USA) was diluted and mixed to 50 ppm. A styrofoam box (66× 42×20 cm) was used as a transport container, and 8 flounder were accommodated and transported in a plastic bag injected with 3 ℓ of seawater and liquid oxygen. As a result of the study, the concentration of cortisol before transport increased significantly from 2.4±0.1 ng ml-1 in the experimental groups except for the CWT+35‰ group (16.7±12.8 ng ml-1). The K+ concentration slightly increased from 3.1±0.0 mEq l-1 before transport to 4.5±1.1 mEq l-1 in the NWT+15‰ group, showing no difference, and significantly increased in all other experimental groups. There was no effect on changes in blood characteristics, and water temperature and anesthetic had a negative effect on osmotic control due to stress. AST and ALT were not affected.

        넙치(Paralichthys olivaceus) 암컷과 강도다리(Platichthys stellatus) 수컷 잡종에서 수정란의 수온별 발생속도 및 자어 성장

        To investigate the characteristics of hybrid eggs and larva produced by olive flounder Paralichthys olivaceus femalesand starry flounder Platichthys stellatus males, we examined the developmental speed of hybrid eggs at differentwater temperatures. The developmental speed of hybrid eggs tended to increase with increasing water temperature. Specifically, the hatching times were 91 hrs, 62 hrs and 43 hrs at 10℃, 15℃and 20℃, respectively. The mean biologicalminimum temperature of the hybrid was 1.3℃, which is in between that of the olive flounder and the starryflounder. In high water temperatureseasons, slower growth was observed in hybrids of the starry flounder which is acoldwater fish.

        대구 (Gadus macrocephalus)의 정자 동결보존

        본 연구는 대구 (Gadus macrocephalus) 정자의 동결보존 기술을 개발하기 위해 적정 동해방지제 및 동결보존 방법을 탐색하고, 동결보존된 정자를 해동하여 효과를 조사하고자 한다. 동결보존에 사용된 동해방지제는 demethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol, methanol 및 propylene glycol (PG)였으며, marine fish ringer’s solution (MFRS)를 희석액으로 사용하였다. 적정 동결보존 방법을 탐색하기 위하여 동해 방지제 농도별 (10%, 20%), 평형시간 (3분, 5분, 10분), 1차 동결 (액체 질소 표면 위 3, 8, 12 cm) 별로 총 90개 실험구를 설정하였다. 해동 후 가장 높은 정자 생존율을 보인 실험구는 PG 10%, 평형시간 3분, 1차동결 8 cm 방법으로21.3±1.8%의 생존율을 보였다. 신선한 정자와 해동한 정자를 사용하여 인공수정시킨 수정란의 부화율에서 유의한 차이는 없었다. An experiment was performed to obtain cryopreservation techniques of Pacific cod Gadus macrocephalus sperm. Milt were cryopreservation using five cryoprotectant demethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol, methanol and propylene glycol (PG) with marine fish ringer’s solution (MFRS) as diluent. Milt were cryopreserved each experimental methods like cryoprotectants (10% and 20%), equilibration time (3, 5 and 10 min) and freezing protocols (liquid nitrogen vapor above 3, 8 and 12 cm). Post-thaw sperm survival rate revealed the highest in 10% PG with optimum methods of equilibration time (3 min) and freezing protocol (liquid nitrogen vapor above 8 cm) about 21.3±1.8%. Hatching rate of fertilization eggs using fresh and cryopreserved sperm were no significantly different.

        먹장어(Eptatretus burgeri)의 혈액성상과 산소소비에 미치는 수온 및 염분의 영향

        Hagfish Eptatretus burgeri is classified as a agnathans and has many different physiological properties compared with vertebrates. In this study, we examined effects of water temperature and salinity on blood properties and oxygen consumption in hagfish. In the experiment of water temperature change, hematocrit (Ht), red blood cell (RBC) and glucose of hagfish blood revealed the lowest values at 15℃. Oxygen consumption of hagfish had significantly increased with rising water temperature, and the increasing rate was twice as much when the temperature was manipulated every 5℃. Also, oxygen consumption during the night time (a short photoperiod) was significantly higher than that of the daytime. Q10 level was 3.50 in the light period and 3.92 in the dark period. No significant change in plasma glucose level was showned in changing salinity from 30 psu to 22 psu, while it had rapidly increased at 20 psu (13.7±4.0 mg/dL) and thereafter all hagfish were dead at 18 psu. However, osmolarity, Na+, K+ and Cl- levels had significantly decreased when salinity decreased. This results are expected to develop the artificial rearing techniques of natural hagfish.

        고수온에서 조피볼락(Sebastes schlegeli)의 산소소비 및 혈액성상 변화

        Aquaculture production of Korean rockfish Sebastes schlegeli has been continuously increased from 2000s and the fish has become the second most important mariculture fish in Korea. However, there are some environmental problems in aquaculture of Korean rockfish recently. In this regards, stress responses to high water temperature was examined via oxygen consumption, blood physiological parameter and endocrinological method. Oxygen consumption of Korean rockfish had significantly increased with rising water temperature. And oxygen consumption during the light time was no different with the dark time. The levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) showed no difference until 27℃, but it had rapidly increased at 30℃. Moreover high water temperature affected to increase in plasma glucose and cortisol levels.

        수온별 저산소 노출에 따른 조피볼락 (Sebastes schlgeli)의 혈액성상 변화

        The objective of this study was to evaluate the effects of acute hypoxia on the physiological stress responses (plasma cortisol as the primary response, and hematocrit, hemoglobin, plasma glucose, sodium, chloride, osmolality, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) as the secondary responses) of Korean rockfish. The mortality in Korean rockfish started at 0.7 mg L-1 and 0.8 mg L-1 levels at 12℃ and 20℃, respectively. And the time required for the mortality in experimental fish was 274 and 148 minutes at 12℃ and 20℃, respectively. Levels of hematocrit, hemoglobin, AST, ALT, ion concentration, osmolality, glucose and cortisol had significantly increased with decreasing dissolved oxygen at 12℃ and 20℃.

        암모니아 노출에 따른 조피볼락(Sebastes schlegeli)의 생리학적 반응

        The aim of the present study was to assess the effects of ammonia on physiological responses in Korean rockfish (Sebastes schlegeli). Interestingly, no mortality were observed when the specimens (301.1±8.0 g) were exposed to five levels of un-ionized ammonia (NH3) (control, 1, 2, 4, 8 mg L-1) for 3 hours. Furthermore, a significantly higher increase in gill Na+/K+-ATPase (NKA) pump activity with was detected due to the ammonia exposure. The activity of the fishes were found to be 4 and 8 mg L-1 NH3, which was significantly high compared to normal 1 mg L-1 NH3. Although ammonia exposure had no effect on plasma Cl-, exposure to both 4 and 8 mg L-1 NH3, however it led to increase in the plasma Na+, K+ and osmolality levels. Also, prolong ammonia exposure cause increase of plasma cortisol and glucose levels. The increase in glucose was accompanied by an increase in cortisol. The fish exposed to 4 and 8 mg L-1 NH3 showed significantly higher hematocrit than control group than those exposed to 1 and 2 mg L-1 NH3. The intensity of cell damage increased with the increase concentration and exposure to ammonia. Furthermore, hyperplasia, separation and epithelial necrosis were also observed in gill tissues. Taken together, the results showed that direction of changes to the investigated parameters can be used to determine the physiological responses of Korean rockfish to ammonia.

        넙치 Paralichthys olivaceus 수정란의 수온별 발생 속도

        This study was performed to examine the influence of water temperature on egg developmental speed for determining the required time and optimum water temperature for hatching of olive flounder Paralichthys olivaceus eggs. The fertilized eggs were collected from the naturally spawned adults in November 2007. The eggs were randomly divided into 6 groups of temperature (5, 10, 15, 20, 25 and 30℃) and transferred in 1ℓ beaker, respectively. The fertilized eggs of the olive flounder did not hatched at 5℃ and 30℃ and hatching rates at 10, 15, 20 and 25℃ were 3, 12, 25 and 50%, respectively. The relationships between the water temperature (T, ℃) and required time (1/t, hour) from egg to each developmental stage were given as follows ; Blastula: 1/t=0.0208T?0.0951 (r2=0.8593)Kupffer's vesicle: 1/t=0.0052T?0.0176 (r2=0.9819)Myotome: 1/t=0.0034T?0.0172 (r2=0.8508)Hatching: 1/t=0.0016T?0.0068 (r2=0.9915) Biological minimum temperature in egg development was calculated to be 4.3℃.
