        Bak-like단백질을 code하는 cDNA의 동정

        Cells are eliminated in a variety of physiological settings by apoptosis, a genetically encoded process of cellular suicide. Bak, a member of the Bcl-2 protein fami1y; accelerates apoptosis by an unknown mechanism. We have found a novel cDNA encoding a 101 amino acid protein possessing a Bak-like in our full-length cDNA bank. Bak-like shares the conserved domains BHI and 2 with other proapoptotic proteins but lacks the BH3 domain. Bak-like is expressed in a wide variety of tissues. Like Bak, Bak-like gene product primarily enhances apoptotic cell death following an appropriate stimulus.

        A variety of genetic alterations in human glioblastomas comprises signal transduction and cell cycle arrest control of cellular processes. Subtractive hybridization is potentially a faster method for identifying differentially expressed genes associated with a particular disease state. Using the technique of subtraction, we isolated a novel gene that is overexpressed in glioblastoma as compared to normal brain tissue. Glioblastoma was used as tester and brain normal was used as driver. We identified 23 novel genes by BLAST of the digested 130 clones. In this study, we selected that this 571 bp cDNA of “clone 25” was not homologous to any of the known genes in the Genbank database. The expressions of mRNA of “clone 25” of brain tumors were significantly higher than normal brain tissue. “Clone 25” is clearly more frequently and more strongly expressed in several carcinoma cell lines compared to corresponding normal tissues of lung, colon, and prostate. To test the time course for G0-phase arrest, serum stimulation and expression at various times for RT-PCR was performed. We confirmed the mRNA expression of “clone25” was up-regulation for 0.5, 1, 2, and 4hr of WI-38 cell differentiation. The novel gene, “clone 25” was upregulated in glioblastoma tissue and several cancer cell lines. This gene is time dependent activation during time course of serum stimulation. Expression and activation of “clone 25” may be important and specific development roles of glioblastoma.

        가미소암산(加味消癌散)의 혈관신생(血管新生) 억제에 관한 항암효과 연구

        Soamsan is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. Extracts of Gamisoamsan inhibited the growth of cultured CT-26 cells, mouse colon adenocarcinoma, in a dose-dependent manner $(1\;to\;50{\mu}g/ml)$, and $ID_{50}$ was estimated approximately $16.7{\mu}g/ml$. Using tumor-bearing mouse model, in which was produced by subcutaneous injection of CT-26 cells ($1{\times}10^5$cells). the effects of Gamisoamsan on tumor growth and host survival were examined by evaluating tumor volume and increase in life span. When Gamisoamsan extracts in variable doses of 100, 200 and 500mg/kg body weight per day were orally administered to tumor-bearing mice, following results were obtained: Improvement in the hematological parameters following Gamisoamsan treatment such as hemoglobin contents, red blood cells and white blood cells of the tumor-bearing mice have been observed. Gamisoamsan treatment also showed a prolongation of life span and a reduction of tumor volume in the CT-26 tumor hosts. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.

        태반 내 Immortalization-upregulated Proteins-2 (IMUP-2) 발현

        목 적: Immortalization-upregulated proteins (IMUPs) family는 SV40의 유전자 도입을 통한 불사화된 인간 섬유아세포에서 새로이 분리 동정된 핵 내 단백질로써, 세포의 증식과 종양형성에 관여하는 것으로 알려져 있으나, 태반 발달과정에 따른 발현 양상과 기능에 대해서는 알려져 있지 않은 실정이다. 본 연구의 목적은 정상 태반과 자간전증 태반조직에서의 IMUPs 유전자의 발현을 분석하고, IMUPs 유전자의 HTR-8/SVneo trophoblast cells 내로 도입 후 IMUPs의 기능을 분석하고자 하였다. 연구방법: IMUPs 발현을 분석하고자, 정상 태반 (n=15), 중기 자간전증 태반 (n=11), 그리고 말기 자간전증 태반 (n=15)조직을 수집하여 RT-PCR, RNA in situ hybridization, 면역조직화학법, 그리고 Western blot 등을 실시하였다. IMUP-2의 기능을 확인하고자 HTR-8/SVneo trophoblast cells에 IMUP-2 plasmids를 transfection한 뒤 24시간 후에 각 그룹간의 세포 수를 계수하였으며, 세포사멸 관련 유전자들의 발현을 분석하고자 RT-PCR, 그리고 Western blot 분석 등을 실시하였다. 결 과: IMUPs는 주로 태반 내 합포영양막세포와 포합체결절에서 주로 발현되었다. IMUP-1의 경우 정상과 자간전증 태반에서의 발현의 차이가 관찰되지 않았으나, IMUP-2의 경우 정상 태반에서 매우 약한 발현을 보였으며, 자간전증에서는 발현의 증가가 통계학적으로 유의하게 관찰되었으며 (p<0.001), 특히, 중기 자간전증의 태반조직 내에서는 강한 발현이 관찰되었다. 또한, IMUP-2의 유전자 도입에 의해 과발현된 HTR-8/SVneo trophoblast cells에서는 세포사멸 관련 유전자들의 증가로 영양막세포의 수가 감소됨이 관찰되었다. 결 론: 이러한 결과들은 IMUP-2의 발현이 정상 태반의 발달에 관여할 뿐 아니라 증가된 IMUP-2는 영양막세포의 세포사멸을 증가시킴으로써 자간전증과도 상관성이 있음이 관찰되었다. 따라서, IMUP-2는 자간전증을 예측 및 진단 할 수 있는 마커로 유용하게 활용 가능하다고 사료된다. Objectives: Members of the immortalization-upregulated protein (IMUP) family are nuclear proteins implicated in SV40-mediated immortalization and cellular proliferation, but the mechanisms by which their expression is regulated are still unknown in placenta. To investigate to expression and functions of IMUPs in placenta, we conducted to compare IMUPs expression in normal and preeclamptic placenta tissues and analyzed the function of IMUP-2 in HTR-8/SVneo trophoblast cells after IMUP-2 gene transfection. Methods: The expression of IMUPs was analyzed in placental tissues from the following groups of patients (none underwent labor): 1) term normal placenta (n=15); 2) term with preeclamptic placeneta (n=15); and 3) pre-term with preeclamptic placenta (n=11) using semi-quantitative RT-PCR, RNA in situ hybiridization, immunohistochemistry, and Western blot. In order to evaluate the function of IMUP-2 in HTR-8/SVneo trophoblast cells, IMUP-2 plasmids were transfected into HTR-8/SVneo trophoblast cells for 24 hours. Results: We observed that IMUPs are mainly expressed in the syncytiotrophoblasts and syncytial knot of placental villi. The expression of IMUP-1 was not differences between normal and preeclamptic placenta tissues. However, IMUP-2 expression was significantly higher in preterm preeclamptic placenta tissues than in normal placenta tissues without labor (p<0.001). Furthermore, we confirmed overexpression of IMUP-2 induced apoptosis in HTR-8/SVneo trophoblast cells through up-regulation of pro-apoptotic proteins. Conclusions: These results suggest that the expression of IMUP-2 is involved in placental development as well as increased IMUP-2 expression is associated with preeclampsia through the inducing of trophoblast apoptosis.

        사회혁신기업의 마케팅을 위한 SNS 활동 특성이 브랜드 이미지와 이용의도에 미치는 영향

        본 연구는 비대면 환경하에서 어려움을 겪고 있는 사회혁신기업들의 마케팅 수단으로 활용하고 있는 SNS활동 특성이 브랜드 이미지와 이용의도에 미치는 영향에 관하여 실증하였다. SNS활동 특성은 정보접근성과 정보품질성 및 상호작용성으로 구성하였으며, 종속변수를 구매의도가 아닌 이용의도로 설정한 이유는 사회혁신기업의 특성이 사회서비스와 같은 서비스 이용자가 많기 때문이다. 설문은 350명의 사회혁신기업 고객들을 대상으로 이루어졌다. 실증결과 정보품질성과 상호작용성은 브랜드 이미지에 유의한 영향을 미치며, 브랜드 이미지는 이용의도에 유의한 영향을 미치는 것으로 나타났다. 또한 정보품질성은 유일하게 이용의도에 유의한 영향을 미치고 있어 정보의 콘텐츠의 중요성을 살펴볼 수 있었다. 본 연구는 약 3만 여 개에 달하는 중소규모의 사회혁신기업에게 SNS를 활용한 마케팅활동의 중요성과 좀 더 실효성 있는 콘텐츠가 중요함을 시사하고 있다. This study demonstrated the effect of SNS activity characteristics, which are being used as marketing means by social innovation companies that are having difficulties in a non-face-to-face environment, on brand image and intention to use. The characteristics of SNS activity consisted of information accessibility, information quality, and interactivity, and the reason why the dependent variable was set to use intention rather than purchase intention is because social innovation companies have many service users such as social services. The survey was conducted with 350 customers of social innovation companies. As a result of empirical results, it was found that information quality and interactivity had a significant effect on brand image, and that brand image had a significant effect on intention to use. In addition, information quality was the only one that had a significant effect on intention to use, so it was possible to examine the importance of information content. This study suggests the importance of marketing activities using SNS and more effective contents for about 30,000 small and medium-sized social innovation companies.

        A variety of genetic alterations in human glioblastomas comprises signal transduction and cell cycle arrest control of cellular processes. In an attempt to identify novel genes implicated in glioblastoma development, we performed experiments using suppression subtraction hybridization(SSH) with human glioblastoma tissue, as well as with cloned genes, that are expressed more than in normal brain tissues. After the identification of a novel gene, RT-PCR was performed to determine its mRNA expression in human glioblastoma tissues. In order to learn more about the expression profile of this gene, PCR was performed using various commercially available normal or carcinoma cell lines. The novel gene was found to be strongly expressed in glioblastoma tissues and other carcinoma cell lines. To determine whether this novel gene was associated with cell cycle regulation, a serum stimulation study with normal WI-38 fibroblast cells treated medium containing 0.1% FBS for 48hr was used for its examination. The novel clone was identified as being expressed in several glioblastoma tissues and carcinoma cell lines. The time-course expression of this novel gene in the WI-38(8PDL) normal lung cell line indicated a significant increase for G1-phase arrest. We have used suppression subtractive hybridization(SSH) to identify cDNA of a gene “Clone 15” overexpressed in human glioblastoma. When the expression of “Clone 15” was screened in numerous cell types, we could confirm that this novel gene was overexpressed in tumor compared to matched normal tissue.
