Giant Synovial Chondromatosis of the Knee Mimicking a Parosteal Osteosarcoma: A Case Report

Synovial chondromatosis is a benign nodular cartilaginous proliferation arising in the synovium of joints. The radiolographic features of this condition are variable. Rarely, it would be confused with malignancy such as chondrosarcoma, osteosarcoma or synovial sarcoma. We report a case of primary synovial chondromatosis of the posterior aspect of the proximal tibia mimicking a parosteal osteoarcoma on the radiography, which showed a homogeneously radiopaque juxtacortical mass. However, subsequent computed tomography (CT) showed multiple intra-articular masses containing chondroid mineralization, suggesting synovial chondromatosis. 활액막성 연골종증은 관절 활액막으로부터 기원하는 증식성의 결절성 연골성 양성 종양이다. 영상의학적 소견은 다양하게 나타나는데 드물게 연골육종, 골육종 그리고 활액 육종과 같은 악성 종양과의 감별을 요하는 수가 있다. 저자들은 컴퓨터 단층 촬영에서는 관절 내 다 발성의 연골성 무기질침착이 뚜렷한 종괴로 인해 활액막성 연골종증을 예상할 수 있으나, 단순 방사선 소견 상 피질골에 근접하고 균질한 방사선비투과의 모습을 보여 방골성 골육종과 유사해 보이는 근위 경골의 후면에 발생한 원발성 활액막성 연골종증 1예를 보고한다.

비브리오균은 해양 및 하구 환경에서 상재하는 균으로 그람 음성 간균, 호염성, 비포자형성의 특징을 가지고 있다. 질병과 관련된 균주로는 Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio cholera 등이 있다. 매년 어패류의 섭취 등을 통해 식중독 사고가 빈번하게 발생하고 있으며, 이에 본 연구에서는 서해안 6곳에서 패류를 채취하여 비브리오균의 분포도를 모니터링하고, 이들의 항생제 내성을 분석하였다. 총 120건의 패류를 TCBS 평판배지와 API 20E kit 기법으로 동정하였을 때 23.3%의 시료에서 비브리오균이 검출되었다. 이들 분리균주들의 항생제 내성을 16종의 항생제에 대해 분석한 결과 82.1%가 각각 vancomycin과 ampicillin에 대해 내성을 보였고, rifampin (71.4%)과 cephalothin (53.6%)에 대한 내성비율도 다른 항생제들에 비해 상대적으로 높은 것으로 나타났다. 분리균주들의 항생제 감수성은 chloramphenicol (92.9%), sulfamethoxazole/trimethoprim (92.9%)과 tetracycline (96.4%)에 대해 가장 높았다. 분리균주들의 약 71.4%가 vancomycin과 ampicillin을 포함하여 3개 이상의 항생제에 대해 다중내성을 보였다. Bacteria of genus Vibrio are Gram-negative, curved, halophilic, nonspore-forming bacteria, autochthonous inhabitans of the marine and estuarine environments. Some of the Vibrio species such as V. parahaemolyticus, V. vulnificus, and V. cholerae are associated with human disease. Each year many people have been suffering from food-borne disease caused by the ingestion of seafood. In this study, we have monitored antibiotic resistance of this microorganism in 6 coastal areas of West Sea by sampling shellfish monthly. Vibrio spp. were detected from 23.3% of 120 samples analyzed using TCBS agar plates as well as API 20E kit. Among 16 antibiotics tested, resistance to vancomycin and ampicillin was observed in 82.1% of the isolates, and Vibrio spp. resistant to rifampin (71.4%) and cephalothin (53.6%) were also high. Most of the isolates were sensitive to chloramphenicol (92.9%), sulfamethoxazole/trimethoprim (92.9%), and tetracycline (96.4%). About 71.4% of the isolates showed multiple drug resistance toward 3 antibiotics including vancomycin and ampicillin.
딱지꽃 (Potentilla chinensis) 추출물의 항염증 효과

In this study, we investigated the anti-inflammation effect of Potentilla chinensis (PC) on Raw264.7 macrophage cells. Ethanol extract of PC decreased the production of nitric oxide (NO) in LPS-stimulated RAW264.7 cells. Ethanol extract was fractioned by n-hexane, chloroform, ethyl acetate, n-butanol, water and each fraction was tested for inhibitory effects on inflammation. Among the sequential solvent fractions, PC chloroform extracts (50, 100, 300, and 500 μg/mL) significantly suppressed LPS-stimulated production of NO. During the entire experimental period, 200 and 300 μg/mL of PC chloroform extracts had no cytotoxicity. LPSinduced NO and prostaglandin E2 (PGE2) production were inhibited by PC chloroform extracts up to 50% and 90% of these productions, respectively. PC chloroform extracts reduced the expression of iNOS and COX-2 gene. These results suggest that PC chloroform extracts exhibit strong effects of anti-inflammation and can be a potential candidate in the treatment of acute and chronic inflammatory diseases.
유산균의 생존율 향상을 위한 동결건조보호제로서 호박분말의 효과

Food-grade protective agents such as sucrose, skim milk, and pumpkin powder were studied for their ability to improve the viability of freeze-dried lactic acid bacteria, including Lactobacillus plantarum MG989, L. fermentum MG901, Streptococcus thermophilus MG5139, Lactococcus lactis MG534, Enterococcus faecium MG89-2, and Bifidobacterium animalis ssp. lactis MG741. The best results were obtained with 5% sucrose contained 10% skim milk and 2% pumpkin powder; approximately 43.1 to 86.6% cell viability was observed during the freeze-drying process. During accelerated storage of freeze-dried lactic acid bacteria, those survival rate decrease rapidly, reaching 9.8~35.2% in 1 month. As a result, the group (5% sucrose, 10% skim milk, and 2% pumpkin powder) showed the protective effect much higher by 639.5% than the control group (5% sucrose and 10% skim milk). These results suggested that pumpkin powder was good candidate as a potential cryoprotectant agent.

Lactic acid is an important product arising from the anaerobic fermentation by lactic acid bacteria (LAB). It is used in the pharmaceutical, cosmetic, chemical, and food industries as well as for biodegradable polymer and green solvent production. The poly lactic acid (PLA) is an important material for bio-plastic manufacturing process. For PLA production by new LAB, we screened LAB isolates from shellfish. A total of 28 LAB were isolated from various shellfishes. They were all Gram positive, oxidase and catalase negative. Based on API 50CHL kit, 7 strains among the 28 isolates were identified as Lactobacillus plantarum, 6 strains as Lactobacillus delbrueckii, 5 strains as Leuconostoc mesenteroides, 3 strains as Lactobacillus brevis, 2 strains as Lactococcus lactis, 1 strain as Lactobacillus salivarius, 1 strain as Lactobacillus paracasei, 1 strain as Lactobacillus pentosus, 1 strain as Lactobacillus fermentum and 1 strain as Pediococcus pentosaceu. Also, we examined the amount of total lactic acid produced by these new strains by HPLC analysis with Chiralpak MA column. One strain E-3 from Mytilus edulis was indentified as Lactobacillus plantarum and found to produce 20.0 g/L of D-form lactic acid from 20 g/L of dextrose. Further studies are underway to increase the D-lactic acid production by E-3.
길경, 황기와 오미자 혼합추출물의 NO 억제활성과 Hyaluronidase 억제활성 효과

본 연구에서는 천연식물자원으로부터 항염증, 항산화 및 항관절염에 활성을 갖는 유용한 물질을 검색하기 위해 문헌조사 및 기존의 연구결과를 통하여 선정된 53가지의 생약재로부터 NO와 HAse(hyaluronidase)의 억제활성 여부를 측정하였다. 측정 결과 억제효과가 우수하다고 확인된 황기(Astragalus membranaceus)와 오미자(Schisandra chinensis), 길경(Platycodon grandiflorum)을 최종 시료로 선정하였고, 이를 단독으로 사용하기보다는 유효성분의 항염증 및 항산화 효능에 대한 상승효과를 알아보기 위하여 혼합추출물의 최적조건을 확인하였다. 각 소재들을 1:2:1로 혼합한 혼합추출물에서 다양한 추출조건에 따라 단일 생약재 추출물에 비해 2배에서 4배까지의 수율 증가를 보였으며, NO 억제활성 효과도 증가한 것을 확인할 수 있었다. 수율과 NO 억제활성을 고려할 경우 혼합추출물의 최적 추출 조건은 황기, 오미자, 길경의 혼합비율 1:2:1, 추출시간 12시간, 추출온도 30oC, 혼합 속도 50 rpm, 추출용매로 ethyl acetate이었다. 위의 최적 조건을 적용한 혼합추출물은 HAse 저해 활성 실험에서 최초 추출물(not grinded, 95% EtOH, 24시간, 실온, 0 rpm)보다 약 16% 정도의 저해 상승효과가 나타났다. 이는 길경과 황기, 오미자의 상승작용 때문으로 사료된다. 이를 통해 황기와 오미자, 길경의 혼합추출물이 항염증, 항산화 및 항관절염에 활성을 갖는 유용한 물질로 사용 가능할 것이라 사료된다. In this study, the optimal extraction conditions for three medicinal herbs as functional sources against inflammatory and arthritic diseases were developed. Traditional medicinal herbs were screened for their inhibition of hyaluronidase (HAse) activity and nitric oxide (NO) synthesis. For the screening of anti-inflammatory properties, ethanolic extracts of 53 species of traditional medicinal herb were examined. We confirmed that Astragalus membranaceus (A.R.), Schisandra chinensis (S.F.), and Platycodon grandiflorum (P.G.) inhibit NO production. For extraction from all three herbs simultaneously, an ethanol concentration of 95%, a 1:2:1 mixture ratio, and at 50 rpm mixing speed, for over 12 h and at 30oC was the best condition for optimal extract yield and NO inhibition effects. HAse inhibition from the three herb extraction was three fold higher than single samples. The ethanol extracts were fractionated with various solvents (n-hexane, chloroform, ethyl acetate, n-butanol, and water). The ethyl acetate-soluble fraction of the herb mixture showed the highest extract yield (13%) and NO inhibition effects (73%). In conclusion, this study provides experimental evidence that a mixture of P.G., A.R., and S.F. could be used as a source of antioxidant ingredients in the food industry.
독활(Aralia cordata Thunb) 추출물의 Nitric Oxide Synthesis 저해효과

Assessment was made of the effects of Aralia cordata Thunb (DH) on the cell proliferation, inducible nitric oxide synthase (iNOS) mRNA gene expression and nitric oxide (NO) production in RAW 264.7 macrophage cells. For the screening of anti-inflammatory activities, ethanolic extracts of 55 species of traditional herbal medicines were examined for inhibitory effects, and it was confirmed that DH possessed inhibitory effects on NO production. As a result, DH significantly decreased the production of NO and iNOS gene expression at a concentration of 250 μg/mL. The chloroformsoluble fractionates have the strongest No synthesis inhibitory effect. It is presumed that the inhibition of NO production in LPS-stimulated RAW 264.7 cells by DH components occurred via the modulation of iNOS and DH, and that the active compound from DH may be useful for therapeutic management of inflammatory-associate diseases.
전통발효식품에서 분리한 유산균의 항균활성 및 프로바이오틱스 기능성 연구

The aim of this study was to investigate probiotic characteristics and fermentation profile of selected lactic acid bacteria (LAB) isolated from traditional fermented foods. Antibacterial activity against various pathogens, acid and bile salt tolerance, cell hydrophobicity, and antibiotic resistance were examined. 16S rRNA sequencing was carried out to identify eight presumptive LAB isolates. In general, all identified LAB (Enterococcus faecium MG89-2, Lactobacillus plantarum MG207, L. paracasei MG310, L. casei MG311, Streptococcus thermophilus MG510, L. bulgaricus MG515, L. helveticus MG585, and L. fermentum MG590) showed strong antimicrobial activity. Also, the selected strains were resistant to bile acid up to 3% and their autoaggregation rates were as high as 60%. All selected strains were sensitive to chloramphenicol, tetracycline, and ampicillin, whereas resistant to nalidixic acid and kanamycin.

Lactic acid and its derivatives are widely used in the food, pharmaceutical, and cosmetic industries. It is also a major raw material for the production of poly-lactic acid (PLA), a biodegradable and environmentally friendly polymer and a possible alternative to synthetic plastics derived from petroleum. For PLA production by new strains of lactic acid bacteria (LAB), we screened LAB isolates from shellfish. A total of 51 LAB were isolated from 7 types of shellfishes. Lactic acid production of individual isolates was examined using high-performance liquid chromatography using a Chiralpak MA column and an ultraviolet detector. Lactobacillus plantarum T-3 was selected as the most stress-resistant strain, with minimal inhibition concentrations of 1.2 M NaCl, 15% ethanol, and 0.0020% hydrogen peroxide. In a 1 L fermentation experiment, D-lactic acid production of 19.91 g/L fermentation broth was achieved after 9 h cultivation, whereas the maximum production of total lactic acid was 41.37 g/L at 24 h.
