        This study was conducted to evaluate the prediction of sensory attributes of tobacco smoke by the use of volatile oil components of tobacco. For analytical and sensory evaluations, twelve aging tobaccos (i.e., 3 crop years; 1998, 1999, 2000, and 4 stalk positions) were prepared in flue-cured and burley tobaccos. 61 volatile oil components and 5 sensory attributes such as irritation, impact, after taste, bitter and green were investigated. Irritation of flue-cured tobacco, and irritation and impact of burley tobacco were significantly increased with the ascending stalk position, whereas after taste of burley tobacco was significantly decreased. Significantly positive correlations among irritation, impact, and bitter were observed in flue-cured tobacco. A significantly positive correlation between irritation and bitter was observed, significantly negative correlations between after taste and irritation and between after taste and impact were observed in burley tobacco. Except for green of burley tobacco, all probabilities of multiple linear regression equations between volatile oil components of tobacco and sensory attributes of tobacco smoke were significant(P$\leq$0.05). This study suggests that the multiple linear regression equations may be useful to predict the sensory attributes of tobacco smoke with a few selected volatile oil components of tobacco.

      • 황색종 담배에서 생육형질, 품질 및 화학성분과의 관계

        This study was conducted to relate among growth characteristics, quality, and chemical components for flue-cored tobacco. The results are summarized as follows : 1. Starch content in harvested green leaf was correlated negatively with leaf length, leaf width, leaf area, and leaf weight of harvested green leaf, respectively, while positively with total sugar content in cured leaf, and the ratio of leaf length to leaf width of harvested green leaf. 2. Organic matter and total nitrogen contents in the soil were correlated positively with nicotine content in cured leaf, respectively, and total nitrogen in the soil negatively with total sugar content in cured leaf. 3, Amount of fertilizer, application date of MH, priod of harvesting, and yield were correlated positively with nicotine content in cured leaf, respectively , while negatively with total sugar content. Application amount of MH was correlated positively with total sugar content, but negatively with nicotine content in cured leaf. Also amount of compost was correlated negatively with nicotine content in cured leaf. 4. The ratio of total sugar to nicotine (TS/N) per plant was correlated positively with price (Won/kg) and specific leaf area, but negatively with leaf length. leaf width, leaf area, harvested leaves, and leaf weight in cured leaves, respectively. Yield was correlated negatively with TS/N per plant. 5. TS/N of the best quality tobacco per plant was 12.0. Those of the best quality tobacco in each stalk position were 42.1 for first~third leaf, 28.4 for 4th~6th leaf , 23.7 for 7th~9th leaf, 7.7 for l0th~12th leaf , and 7.8 for over 13th leaf from bottom, respectively. 6, When TS/N was 12.0, optimum values of growth characteristics per plant were 100.5 $\pm$ 10.3g for leaf dry weight, 755.1$\pm$53.2cm for leaf length, 294.4$\pm$25.1cm for leaf width, 8, 892$\pm$111cm2 for leaf area, 16.0$\pm$0.6 leaves for harvested leaves, and 7.32$\pm$0.44mg/cm2 for specific leaf area, respectively. 7. When TS/N was 12.0, optimum values of chemical components per plant were 1.92$\pm$0.28% for nicotine content, and 20.4$\pm$ 1.4 % for total sugar content, and that of yield was 238.3$\pm$ 9.8kg/10a.

      • 잎담배의 적심시기와 정도에 따른 엽중 비휘발성유기산 및 지방산의 함량변화

        This study was conducted to investigate the influence of the topping time and height on the nonvolatile organic and higher fatty acid contents and the correlation among acid contents of tobacco plants. Within the topping time, the flowering (normal) stage of topping was the lowest in total analyzed acid (TAA), total nonvolatile organic acid (TNOA) and magic acid contents of NC2326, however, the highest in those of Burley 21 on 30 days after budding. Total higher fatty acid (THFA) and linoleic acid contents tended to be low with normal stage of topping. The lower the topping height, the lower the TAA, THFA and linoleic acid contents in middle and upper leaves (or NC2326 and Burley 21 were observed. On the other hand, the lower the topping height, the lower the TNOA and malic acid contents in middle and upper leaves for Burley 21 and upper leaves for NC2326 were found. There was a significant positive correlation between magic acid and TNOA contents. Similarly, TAA content showed a significant positive correlation with TNOA, oxalic acid and citric acid contents. But there was a negative correlation between malic acid and THFA contents. There were highly positive correlations among oleic acid, linoleic acid and THFA contents, respectively.

        Killer 효모와 알콜 발효효모간의 원형질체 융합주의 특성

        원형질체 융합을 통하여 killer 효모의 유전형질을 기존의 ethanol 발효효묘에 도입하므로서 야생의 killer 효모에 저항성을 가지고, 오염효모를 치사시킬 수 있으며, ethanol 발효능도 유지하는 새로운 효모균주를 개발하였다. 먼저 융합주의 생리적인 특성을 검토한 바, 융합주는 양천주에 비하여 세 적이 크고, DNA 함량이 높았으며, 증식도는 친주와 유사한 경향이었다. 또 융합주를 최소배지에서 보존하는 것이 안정성을 높이는 방법이었고, 7일 간격으로 7회 계대배양하므로서 유전적으로 안정화시킨 융합주를 6개월 간 계속 사용한 결과 segregant가 전혀 나타나지 않았으므로 매우 안정하였다. 또한 융합주는 핵 및 포자를 형성함을 관찰 할 수 있었고, TTC 정색반응에서 적색 및 pink 색을 띄었으며, 탄소원의 자화성 및 발효능은 천주와 유사하였는데, KCI, NaCl, sodium propionate alc cycloheximide 등에 대한 내성도 양친주의 한쪽을 따랐다. 그리고, 융합주를 20% glucose와 sucrose에서 72시간 및 60시간 배양했을 때, FWKS 260의 경우 각각 9.6v/v% 및 9.8v/v%의 ethanol을 생성하였고, 감수성주와 혼합배양한 결과 FWKS260의 경우 48시간 이후에는 감수성주를 거의 발견할 수 없었으며, 전주 및 융합주의 dsRNA plasmid를 추출하여 전기 2.5kb의 L 및 M dsRNA plasmidsms는 서로 연관성이 있으며, killer toxin 분비 및 저항성을 나타내는 유전자를 지배하는 것은 M dsRNA plasmid임을 확인할 수 있었다. 수 있었다. Cell volume and DNA contents of the fusants were similar to those of parents. Genetic stability of the fusants was increased when they were cultured on minimal medium (MM) rather than on complete medium (CM), and the fusants were stabilized by subculturing 7 generations each 7 day on MM agar. The finally selected fusants after being cultured for 6 months on CM were stable without segregation. The fusants could also form nuclein and ascospores, and show red and pink colors by the test of TTC colorization. Assimilability and fermentability of carbon sources of the fusants were similarto those of parents. The tolerance of KCl, NaCl, sodium propionate and cycloheximide showed the traits of one strain of parents. When the fusants were cultured for 72 hr and 60 hr in the medium containing 20% glucose and sucrose, respectively, the yield of ethanol for FWKS 260 was reached to 9.6 v/v% and 9.8v/v%, respectively. The sensitive strain Kyokai 7 was found to be killed entirely after cultivation of 48 hr by the killer toxin from the fusants. The recipient S 29 and Kyokai 7 were found to have neither L nor M dsRNA plasmid. However, K 52 and fusants had both L and M dsRNA plasmid of 4.7 kb and 2.5 kb, respectively. The curants treated by heat and cycloheximide did not contain M dsRNA plasmid, but had large amounts of L dsRNA plasmid of those of killer yeasts.

        .betha.-1, 3-glucanase 생성균의 분리 및 효소 생성 조건

        The bacteria, which were capable of producing ${\beta}-1$, 3-glucanase inducibly by utilizing cell wall of Aspergillus fumigatus as a sole carbon source, were isolated from soil in the campus of Kyungpook National University. Among them, the strain which produced the enzyme excellently was selected and identified to be Pseudomonas stutzeri KF 13 by morphological, cultural and physiological examination. The optimal conditions for the enzyme production from Pseudomonas stutzeri KF 13 were investigated. the enzyme production was reached maximum state shen the broth cultured for 72hr at $30^{\circ}C$. And the enzyme showed the highest activity in the medium containing 3.5% cell wall as an inducer, 15% yeast autolysate as a nitrogen source and 0.05% $MnSO_4$ at pH 7.5.

        아세트 알데히드(특수산업) 공장폐수의 성분과 이용균주의 분리

        아세트알데히드 페수를 생물학적으로 처리할 목적으로 폐수의 성상을 조사하였으며, 아세트알데히드 폐수에서 잘 생육하는 53개 균주를 분리 고정하고 그 중 가장 COD 제거율이 우수한 4개 균주 AW-6, AW-22, AW-38, AW-41을 선별하고 이들을 동정하였다. 아세트알데히드 폐수의 성상은 CODmn 5,260ppm, BODs 6,452ppm, pH 1.85였으며 유기성분의 주체는 초산으로 6.76%를 함유하였다. 균의 동정 결과, AW-6은 Micrococcus 갠뎐, AW-22는 Micrococcus luteus, AW-38은 Microbacterium lacticum, AW-41은 Microbacterium laevaniformans 또는 그 우연균으로 동정되었다. As a research for treatment of waste water from acetaldehyde plant by biological method, we investigated general characteristics of the waste water, and isolated and identified some useful bacteria which effectively treated its waste water. Among the total number of 53 strains which were grown in waste water from an acetaldehyde plant, the strains AW-6, AW-22, AW-38 and AW-41 were found to be useful for COD removal of waste water. $COD_{Mn}$ and $BOD_{5}$ of the waste water were 5260 ppm and 6452 ppm, respectively, and pH was 1.85. And the main organic component in waste water was acetic acid which was contained 6.76%. By the taxonomical characteristics, the strains AW-6, AW-22, AW-38 and AW-41 were identified as Micrococcus roseus, Micrococcus luteus, Microbacterium lacticum and Microbacterium laevanifromans or similar strain, respectively.

        아세트 알데히드(특수산업) 폐수의 생물학적 처리

        아세트알데히드 공장폐수의 생물학적 처리를 위하여 중화제 및 응집제의 효과를 검토하고, 4개 균주 Micrococcus roseus AW-6. Micrococcus luteus, AW -22. Microbacterium lacticum AW-38. Microbacterium laevaniformans AW-41을 혼합 배양하여 미생물의 영양요구량 및 성장에 미치는 환경인자 둥을 조사한 결과는 다음과 같다. 아세트알데히드 폐수를 잘 자화시키는데 필요한 온도는 $30^{\circ}C$였고 pH는 7.0이였으며 96시간째에 COD 제거율이 최대에 달하였다. 폐수처리를 위한 중화제로서는 NaOH가 가장 좋았고 응집제의 처리 효과는 인정되지 않았으며 폐수처리를 위한 최적 희석배수는 10배 정도로 나타났다. 균 생육을 위한 영양원으로서는 $NH_{2}NO_{3}$ 200ppm, $KH_{2}PO_{4}$, 50ppm, $CaCl_{2}$ 15ppm. $MgSO_{4} \cdot 7H_{2}O $ ppm을 보충하였을 때 COD 제거율이 향상되었다. 회분배양 실험결과 속도계수 Kl은 4. 5X 10-6이었으며, BOD 제거융을 85%로 하기 위한 체류시간은 45시간 이었다. 연속배양 결과 COD 제거율은 85%로 나타났으며 처리수의 수질은 COD 60ppm. BOD 40ppm, pH7.0 정도로서 환경기준치 이하였다. In order to establish the biological treatment system which can be used for treatment of waste aster from acetaldehyde plant, it was investigated optimum nutrient requirements and growth conditions by mixed culture of Micrococcus roseus AW-6, Micrococcus luteus AW-22, Microbacterium lacticum AW-38 and Microbacterium laevaniformans AW-41 as well as the effect of coagulants and neutralization reagents. Also, it was carried out the continuous culture as well as batch culture to treat the waste water by mixed culture of these strains. The COD removal rate was reached to maximum state for 96hrs culture at pH7.0 and $30^{\circ}C$ NaOH as the neutralization reagents was the most effective, but the coagulants had no effect on the COD remonal rate and the optimum dilution times for treatment were 10 fold. The COD removal rate was also increased by supplimenting 200 ppm $NH_{2}NO_{3}$, 50 ppm $KH_{2}PO_{4}$, 15 ppm $CaCl_{2}$ and 1 ppm $MgSO_{4} \cdot 7H_{2}O $ as additional nutrients. The removal rate coefficient $K_{1}$ on the batch culture was $4.5\times 10^{-6}$, and the detention time for BOD removal rate of 85% was approximately 45hrs. The COD of waste water was reduced to 15% of its initial value by the continuous culture. The COD and BOD of the effluents were to be about 60 ppm and 40 ppm, respectively, and final pH was 7.0.
