수종의 한약재가 HepG 2.2.15 Cell의 HBeAg발현 억제에 미치는 효과(效果)

Purpose : Hepatitis B virus DNA transfected cell line(HepG2.2.15) was cultured to evaluate the effect of herbs on the expression of HBeAg and the replication of HBV. HepG2.2.15 produces HBV particles as well as viral proteins into cell culture media. Methods : Extracts of herbs were adminitered to the cells on the proper concentration. Culture media was collected 48 hours after the herbal administration and HBeAg level in the media was examined by ELISA method. To confirm that the anti-viral effect was not due to direct cytotocixity of the extracts, normal cell proliferation was shown by cell counting. And as of the interference in protein synthesis of HepG2.2.15 by herb-extracts, we used the result of study that we performed before by ${\alpha}FP$ assay using EIA method. Results& Conclusion : Herb medicines like 地楡(Sanguisorbae Radix) and 覆盆子(Rubi Frusctus) showed significant inhibitory effect on HBeAg expression at p<0.01 and 五味子(Acanthopanacis Cortex) at p<0.05. Whereas, though some herbs such as ?草根(Rubiae Radix), 山査(Crataegii Fructus), 白芍藥(Paeoniae Radix Alba), and 大黃(Rhei Radix et Rhizoma) showed the tendecy to suppress HBeAg. most of them were not significant statistically. From the above, we could conclude that those herb medicines can be applied to patients effectively and further studies on effective fraction of some herbs are thought to be needed.

Objectives : Consumption is a chronic wasting disease and major portion of Oriental Medicine's therapy. However, there is no standard diagnostic method for consumption that is $q{\grave{i}}-x{\bar{u}}$, $xu{\grave{e}}-x{\bar{u}}$, $yang-x{\bar{u}}$, $y{\bar{i}}n-x{\bar{u}}$. Methods : A questionnaire which includes symptoms and signs for diagnosis of $q{\grave{i}}-x{\bar{u}}$, $xu{\grave{e}}-x{\bar{u}}$, $yang-x{\bar{u}}$, $y{\bar{i}}n-x{\bar{u}}$ was evaluated by Delphi technique. Each question was valuated by interviewing 27 oriental medicine doctors. Then. we choose questions given over 5 points and reorganized some items according to the recommendations by interviewed-doctors. We then accessed the value of re-organized questions composing of the questionnaires. Conclusion : We finally chose each 9 items of $q{\grave{i}}-x{\bar{u}}$, $xu{\grave{e}}-x{\bar{u}}$, $yang-x{\bar{u}}$, $y{\bar{i}}n-x{\bar{u}}$'s questionnaire. Further study is necessary for modification of questionnaire by statistics and certification by clinical trial.
YBR의 간섬유화(肝纖維化)억제 효과(效果)에 관한 연구(硏究)

Objective : This study was performed to investigate the anti-fibrogenic effect and changes of inflammation-related genes by YBR I and YBR II (YBR I: Arteisiae Capillaris Herba, Atractylodis Rhizoma Alba, Hoelen/ YBR II: YBR I +Sanguisorbae Radix, Biotae Cacumen, Cirsii Japonici Herba) on HSC(hepatic stellate cells)-T6 and TAA-induced rat liver tissue. Materials and Methods : HSC-T6 were treated with various concentrations of distilled-water extract YBR I and YBR II extract for 24, 48 and 72 hours. After the treatment, cell viability, proliferation, procollagen levels and IL-6 levels were measured by using MTT Assay, BrdU Assay, Procollagen Type 1 C-peptide EIA kit, and Murine IL-6 ELISA Development kit. Rat liver fibrosis was induced by intraperitoneal TAA injection of 150mg/kg 3 times a week for 6 weeks. After the treatment, body weight, liver & spleen weights, liver function test, complete blood cell count and change of portal pressure were studied. In addition, gene expressions of ASMA, IL-6, MMP-2, TIMP-1 and TIMP-2, all of which are known to be associated with liver fibrosis, were analyzed by using Real-Time PCR. After YBR I and YBR IItreatment, percentages of collagen in TAA-induced rat liver tissue were measured. Results : The viability and proliferation of the HSC-T6 decreased as the concentration increased. The production of procollagen decreased as the concentration increased. The production of IL-6 was little influenced by YBR I and YBR II. There was no difference in rat body weight between the TAA-only group and the YBR groups. Compared with rat liver weight of TAA-only group, that of the YBR groups increased. In the YBR I group, the serum level of AST elevated by TAA injection significantly decreased and in the YBR I and II group, the serum level of ALP and ALT elevated by TAA injection decreased. In the YBR I group, white blood cell count elevated by TAA injection decreased but platelets increased. In the YBR I group, the portal pressure elevated by TAA injection significantly decreased. Decreases in the gene expression of ASMA and MMP-2 were observed in the YBR I group. The gene expression of IL-6 was little influenced by YBR I and YBR II -treated groups. In the histological finding, TAA injections caused severe fibrosis, but YBR I and YBR II treatment significantly reduced the amounts of hepatic collagens. Conclusions : These results suggest that YBR I and II have inhibitory effects on the hepatic fibrogenesis.

Objectives : This study was performed to investigate the anti-fibrogenic effect of Curcumae Longae Radix on human hepatic stellate cells. Materials and Methods: Hepatic stellate cells (LX-2) were treated with various concentrations of Curcumae Longae Radix extract for 24, 48, and 72 hours. It was extracted with distilled water. After the treatment, cell viability, proliferation, cell cycle analysis, procollagen levels and the mRNA of the ASMA, TIMPl, TIMP2, MMP2, collagen type la, PDGF-receptor-beta and TGF-beta were measured by using MTT assay, BrdU assay, RT-PCR, and procollagen type 1 C-peptide EIA kit. Results : The viability of HSCs decreased in the 48 hours group, and proliferation of HSCs decreased as the concentration increased. In the cell cycle analysis, Curcumae Longae Radix decreased the ratio of M phase, and increased the ratio of apoptosis, G0/G1 and S phase. In the RT-PCR, the mRNA expression of the collagen type la and ASMA decreased with the Curcumae Longae Radix treatment. The production of procollagen by the HSCs was decreased by the treatment of Curcumae Longae Radix with high dose. Conclusion : These results suggest that Curcumae Longae Radix is helpful in the treatment of liver fibrosis as well as liver cirrhosis.
NASH 동물모델에서 인진청간탕과 인진사령산의 효과규명

Objective : The aim of this study is to investigate the preventive effect of Injinchunggan-tang (YJCGT) & Injinysaryung-san (YJSRS) on MCD-diet-induced NASH in A/J mice. Methods : A/J mice were divided into 4 groups: Normal group (normal diet without any treatment). Control group (MCD diet only), YJCGT group (MCD diet with YJCGT), and YJSRS group (MCD diet with YJSRS). After 5 weeks, body weight, liver weight, biochemical parameters for liver function test, histological changes, and real-time PCR were assessed. Results : Mice lost body weight with the MCD diet and the YJCGT and YJSRS groups lost less than the control group, though showed no statistical significance. Liver weights were decreased by the MCD diet, but not significantly. In the liver function test, all the values were increased with the MCD diet, though some did not show significance. Alp and ALT levels were significantly less increased by YJCGT compared to the normal (p<0.05). All values were decreased or increased compared to the control by treatment though showed no significance possibly due to insufficient sample numbers. In histological findings of the livers. MCD-diet induced severe fatty change and collagen accumulation in the livers, but this fatty change was reduced in the YJCGT and YJYRS groups and fibrogenesis was inhibited significantly with p<0.05 and p<0.01, respectively. In real-time PCR analysis, YJCGT and YJYRS showed inhibitory effect on liver fibrogenesis by reducing associated gene expressions caused by MCD diet. Conclusion : YJCGT and YJSRS are considered to be possible candidates for the treatment of patients with NASH and/or liver fibrosis.

Objectives : This study was performed to investigate the anti-fibrogenic effect of Artemisiae Capillaris Herba on cultured rat hepatic stellate cells. Materials and Methods : Hepatic stellate cells(HSC-T6) were treated with various concentrations of Artemisiae Capillaris Herba extract for 24 hours. The extraction was done either with distilled water or 50% EtOH. After the treatment, cell viability, proliferation, procollagen levels and the mRNA of the collagen type 1a2 and ASMA were measured by using MTT assay, BrdU assay, RT-PCR, and Procollagen Type I C-peptide EIA Kit. Results : The viability and proliferation of the hepatic stellate cells were decreased as the concentration increased. The mRNA expression decreased consistently with the volume of the secreted procollagen with the extraction made with distilled water, which indicates the herb has inhibitory effect on fibrogenesis of the liver by regulating one of the fibrosis associated genes in transcription. However, it increased in 50% EtOH extraction, which shows that a more stable reaction is expected of the extraction made with distilled water than the extraction made with 50% EtOH. The production of procollagen was decreased by a low-concentration treatment with Artemisiae Capillaris Herba, but increased by a high concentration. It seemed that the cells were responding to Artemisiae Capillaris Herba in low- concentrations, thus producing small amounts of collagen. When the drug was administered at high enough concentration to give direct toxicity to cells, the ability of cells to produce collagen was activated, and the overproduction of collagen was observed as an undesirable results. Conclusion : These results suggest that Artemisiae Capillaris Herba is beneficial in the treatment of cirrhotic patients as well as for the patients with chronic hepatitis when extracted with water in the proper concentrations.
