The complete mitochondrial genome of a threatened loach (Sinibotia reevesae) and its phylogeny

In present study, the complete mitochondrial genome of Sinibotia reevesae was first sequenced using the next-generation sequencing technology and annotated using bioinformatic tools. The circular mitochondrial genome was 16,572 bp in length, and contained 13 proteincoding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and 1 displacement loop locus. It presents a typical gene organization and order for completely sequenced cypriniformes mitogenomes. The control region could be divided into three parts included the extended termination associated sequence domain, the central conserved domain and the conserved sequence block. Interestingly, two stemloop domains were found in control region and OL region, respectively. Furthermore, phylogenetic analyses using concatenated amino acid and nucleotide sequences of the 13 protein-coding genes with two different methods (Maximum likelihood and Bayesian analysis) both highly supported the close relationship of S. reevesae and Sinibotia superciliaris, which was in line with the previous classifications based on morphological and molecular studies. These data provide useful information for a better understanding of the mitogenomic diversities and evolution in fish as well as novel genetic markers for studying population genetics and species identification.

The purpose of this study is to investigate the changes of α-actin metabolism, α-actin mRNA and MHC mRNA expression in skeletal muscles of rats after one set exhaustive eccentric exercise and the effects of acupuncture on them. 130 adult male SD rats were randomly divided into the sedentary group and the exercise group. The rats of the exercise group were again divided into the acupuncture group and non-acupuncture group. According to the sampling time, the exercise group (the acupuncture and non-acupuncture group) was divided into 12 groups, that is the 0 hours, 6 hours, 12 hours, 24 hours, 48 hours and 72 hours groups. In each group there were ten rats. The rats weighting 205±23.3 (mean±SE) were maintained at 23±1℃. They were fed in different chests taking food and drink freely. The rats of the exercise group carried out continuous downhill running with the speed of 16m/min for about 4 hours until exhaustion on the animal treadmill (degree -16°). The rats of the acupuncture group were treated with acupuncture immediately after exhaustive eccentric exercise, but those of the sedentary group and the non-acupuncture group were treated without acupuncture. They were the control groups. The muscle samples were taken immediately, 6 hours, 12 hours, 24 hours, 48 hours and 72 hours after exercise. The contents of G-actin, F-actin and T-actin in skeletal muscles of rats were measured by means of Dnase I assay. The gene expression of α-actin m RNA and MHC isoforms mRNA were determined by using the quantitative reverse transcription PCR technology. Compared with the sedentary group, there was no remarkable change of T-actin after exercise. The decrease G-actin could be detected only immediately after exercise. The content of F-actin was significantly higher than that of the sedentary group 0~6 hours after exercise, but 12~24 hours after exercise it decreased to a lower level than that of the sedentary group. A delayed increase of F-actin depolymerization presented after exercise. The F-actin value of the acupuncture group was improved in a lower range than that of the non-acupuncture group 0~6 hours after exercise, but 12~24 hours after exercise it was remarkably higher than those of the non-acupuncture group. It was shown that acupuncture could regulate the F-actin concentration in skeletal muscles of rats after exercise. Furthermore, the regulation might be bilateral. Acupuncture could stop the increase of F-actin delayed depolymerization induced by high loading exercise. The gene expression of α-actin mRNA in skeletal muscles of rats decreased significantly after one set exhaustive exercise. The duration of its recovery was longer than 72 hours. It might be that acupuncture accelerated the recovery process of α-actin mRNA expression after exercise. The MHC mRNA expression of the non-acupuncture group was lower than that of the sedentary group 6~12 hours after exercise. There appeared a delayed excess expression of MHC mRNA 48 hours after exercise. The curve of the change of the total MHC mRNA expression in the acupuncture groups was similar to that in the non-acupuncture group. The duration of recovery of the acupuncture group was shorter, the delayed excess expression of MHC mRNA was ahead of time and its range was higher than that of non-acupuncture group. Molecular dynamics, an eff ective method to gain an insight into nanometric behaviour of materials, was employed to studythe nano-cutting behaviour of single crystal Ni 3 Al in nanometric scale. In this paper, comparisons were made for compressive/tensile stress, subsurface damage and surface roughness with three rake angles of a diamond tool. Subsurface damage waspartitioned by region and studied with work hardening in detail. A model for precise characterization of surface roughnesswas established with consideration of local surface fl uctuation. Simulation results showed that the chip thickness increasedas rake angle changed from negative to positive, and the boundary formed between tensile and compressive stress was inconsistent with the glide direction of stacking fault. Subsurface damage decreased as the increase of rake angle, and regularglide planes of stacking faults were found in front of the cutting tool. Further, the pinned dissociated 1/2 < 110 > superpartialdislocation with anti-phase boundary was demonstrated. The model was tested and characterized by implanted pits onperfect surface. Results showed that surface roughness can be well characterized, and an evident discrepancy was observedamong three rake angles, especially for 30° rake angle, which showed an distinct smooth surface compared with the others.

Objective To investigate the relation between nicotinic acetylcholine receptor subunit (nAChR) genes and schizophrenia, and the relation between tag single nucleotide polymorphism (rs1317286, rs1044396, rs6494212, rs16969968, and rs684513) and schizophrenia in Han Chinese people. Methods The protein-protein interaction (PPI) network among nAChR protein and 350 proteins encoded by schizophrenia-related susceptibility genes was constructed through the String database to explore whether nAChR genes were associated with schizophrenia in these known databases. Then, five single nucleotide polymorphisms (SNPs) of CHRNA3 (rs1317286), CHRNA4 (rs1044396), CHRNA7 (rs6494212), and CHRNA5 (rs16969968, rs684513) were analyzed in a sample of 1,035 schizophrenic patients and 816 healthy controls. The interaction between the markers was analyzed using multifactor dimensionality reduction (MDR) software. Power analysis was performed using the Quanto program. Results There are no significant differences in genotype or allele distribution were identified between the patients and controls (p>0.05). The haplotypes constructed by four markers rs1317286, rs6494212, rs16969968, and rs684513 were not associated with schizophrenia either. However, a significant association between models made of rs1317286, rs1044396, rs6494212, and rs684513 and schizophrenia was revealed in interaction analysis (p<0.05). Conclusion The nAChR protein may have effects on the development of schizophrenia through the interaction with proteins encoded by schizophrenia-related susceptibility genes, but no relation was found between selected polymorphisms and schizophrenia in the collected Han Chinese people. However, interaction analysis suggested four-SNP model has an important effect on schizophrenia.

Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is an unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes. In the present work, the entire encoding region for Oryza sativa PHGPx was expressed in Escherichia coli M15, and the purified fusion protein showed a single band with 21.0 kD and pI = 8.5 on SDS- and IFE-PAGE, respectively. Judging from CD and fluorescence spectroscopy, this protein is considered to have a well-ordered structure with 12.2% $\alpha$-helix, 30.7%$\beta$-sheet, 18.5% $\delta$-turn, and 38.5% random coil. The optimum pH and temperature of the enzyme activity were pH 9.3 and 27$^{\circ}C$. The enzyme exhibited the highest affinity and catalytical efficiency to phospholipid hydroperoxide employing GSH or Trx as electron donor. Moreover, the protein displayed higher GSH-dependent activity towards t-Butyl-OOH and $H_2O_2$. These results show that OsPHGPx is an enzyme with broad specificity for hydroperoxide substrates and yielded significant insight into the physicochemical properties and the dynamics of OsPHGPx.

The snakehead fish, Channa siamensis, belongs to the genus of Channa (perciformes: Channidae) and was first reported by Günther in 1861. Despite it has been described approximately for 15 decades, the genetic information is limited and the taxon status of this kind of fish is still unclear. The primary objective of this study is to get more genomic data and calculate the taxon location of this kind of fish. The next generation sequencing method was used to obtain the whole mitochondrial DNA information, and bioinformatic analysis was performed to investigate the evolutionary status and taxon location of C. siamensis. The circular mitochondrial DNA was 16,570 bp in length, and which showed typical piscine structure and arrangement. The overall nucleotide composition was 29.28% A, 24.72% T, 30.71% C, 15.29% G, with 54.1% AT, respectively. Phylogenetic analyses using concatenated amino acid and nucleotide sequences of the 13 protein-coding genes with two different methods (Maximum likelihood and Bayesian analysis) both highly supported C. siamensis belongs to the genus Channa and shows a close relationship with C. micropeltes. These data will provide more useful information for a better understanding of the mitochondrial genomic diversities and evolution in fish as well as novel genetic markers for studying population genetics and species identification.

When using high energy shot peening (HESP) technology, the surface layer of the welded joints produced grain breakage and residual compressive stress. When the shot peening strength is the optimal parameter 0.10 MPa, the residual compressive stress reaches a maximum value of 74.02 MPa, dislocation density in welded joints increased significantly and the tensile shear strength of the joint was increased by 18.6% compared to the joint without HESP treatment (205 MPa), and the fracture position of the welded joint was located in the Al alloy base material, these were brought by fi ne grain strengthening and strain strengthening. When the shot peening strength was 0.05 MPa, the strength of the joint was also improved, but the fracture position of the joint was the same as that without HESP treatment, and they were all in the fusion zone. However, when the shot peening strength was further increased to 0.15 MPa, severe cracks appeared inside the joint, which deteriorates the strength of the Mg/steel joint.
