        The Contemporary Value of Xunzi`s Political Ethics Thought for the Social Administration

        Xunzi`s political ethical thought, including gentleman personality, people-oriented, prospering etiquettes and respecting law and so on, still has a relational meaning for the modern social political moral construction. Gentleman personality comes with an obligation to improve the leading officials` morality; the idea of people-oriented has a vital significance for establishing“ put people first“ thought throughout society and insisting the mass line; This statecraft that focuses both on ritual and law has a very important revelation for improving the foundation for anti-corruption and a clean government. Xunzi`s political ethical thought provides the value for our country`s conducting a harmonious and efficient social administration through the three dimensions mentioned above.

        There are many key parts in the field of aerospace with aluminum alloy thin-walled parts, the problem of breaking the low efficiency in production that’s also followed. There will be a lot of deformation in processing because of its poor rigidity, poor process, which will lead to low processing efficiency. It is of great significance to improve the machining efficiency of the aluminum alloy thin-walled parts, and a series of analysis of the surface quality of aluminum alloy thin-walled parts there are carried out clamping, cutting tools, take the knife route so on, and the problems of low machining efficiency and poor surface quality are solved.

        Effect of teeth bending and mushrooming damages on leakage performance of a labyrinth seal

        To achieve higher aerodynamic performance, turbine usually works at tight clearance, which results in inevitable rub between the rotor and stator parts of labyrinth seal due to vibrations, misalignment, mechanical force, thermal stress, etc. In the rubbing events, contact between labyrinth fin and rotor part will commonly induce the teeth bending and mushrooming damages, which significantly affect the discharge performance of a labyrinth seal. To account for the influence of teeth bending and mushrooming on leakage performance of a straight-through labyrinth seal, the leakage rates and flow fields in the worn labyrinth seal are measured and also compared with the original design cases. With numerical methods, the discharge behaviors of the labyrinth seal with different degrees of bending and mushroom damages are analyzed. It shows that the predicted leakage performance and flow fields in the labyrinth seals match well with the experimental tests. For the bending cases, the leakage rates and flow patterns in labyrinth seals are dependent on the effective clearance and bending angle. The leakage ratio of forward bending case is smaller than that of backward bending case with the same geometrical dimensions. However, for the mushroomed labyrinth seals, the leakage rates and flow patterns are much dependent on the effective clearance but slightly dependent on the mushroom radius.

        <P><B>Abstract</B></P> <P>pH sensor is very important in various fields, and has triggered many types of devices based on different materials and mechanisms. How to improve the performances of the sensors is one of major challenges now. In the present paper, we developed an idea to improve the sensitivity of pH sensors based on AlGaN/GaN high-electron-mobility transistors (HEMTs) by introducing multi-sensing segments, and experimentally fabricated prototype devicesand then investigated their responses to aqueous solutions under different pH values. The optimized sensor exhibits a very high linear sensitivity of 1.35 mA/pH at drain-source voltage of 1.5 V, which is much higher than those of conventional pH sensors with single open gate sensing area. This indicates that the multi-sensing segments is very effective in increasing the sensitivity of the pH sensor, rather than simply increasing the sensing area. The sensors degrade after measurement in solution for a long working time, but they can recover to their initial states in a certain duration after washing, typically ∼24 h. Our finding paves new strategy for the future design of high sensitive and stable pH sensors based on HEMTs.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Novel multi-sensing segments pH sensor is designed and fabricated by using AlGaN/GaN high electron mobility transistor. </LI> <LI> Realizing very high sensitivity on the multi-sensing sensor, which is much higher than that of conventional geometric pH sensor. </LI> <LI> Achieved excellent performance stability with good recovery feature on the multi-sensing sensor. </LI> </UL> </P>

      • Integration Sites and Genotype Distributions of Human Papillomavirus in Cervical Intraepithelial Neoplasia

        Objectives: To analyse HPV integration prevalence and genotype distributions in cervical intraepithelial neoplasia (CIN) in east part of China, furthermore to assess preferential sites for common HPV integrations and provide baseline information for cervical abnormality screening and prevention. Methods: Integration of HPV in 113 paraffin-embedded cervical intraepithelial neoplasia samples was assessed using Gencap technology in Key Laboratory of Biotechnologies in BGI-Shenzhen. Results: 64 samples were HPV-integrated and as the cervical lesions increased, the integration rate became higher significantly (P=0.002). Fifteen different HPV genotypes were detected, 14 high-risk (16, 18, 31, 33, 51, 52, 56, 58, 66, 68) and 1 low-risk (11). The most common genotypes were HPV-16, 58, 33, 52, 66, and 56. Thirteen patients had co-integration involving mainly HPV-16 and 58. The frequency of HPV gene disruption was higher in L1 and E1 genes than in other regions of the viral genomes. Conclusion: Some 56.6% of CIN lesions in Qingdao had HPV integrations, and 67.2% of HPV-integrated patients were HPV-16 and 58, more prone to be integrated in younger patients below 45 years old. There exist preferential sites for HPV-16 and HPV-58 integration, and they are more likely to be disrupted in the L1 and E1 loci.

        Systemic administration of low dosage of tetanus toxin decreases cell proliferation and neuroblast differentiation in the mouse hippocampal dentate gyrus

        In the present study, we investigated the effect of Tetaus toxin (TeT) on cell proliferation and neuroblast differentiation using specific markers: 5-bromo-2-deoxyuridine (BrdU) as an exogenous marker for cell proliferation, Ki-67 as an endogenous marker for cell proliferation and doublecortin (DCX) as a marker for neuroblasts in the mouse hippocampal dentate gyrus (DG) after TeT treatment. Mice were intraperitoneally administered 2.5 and 10 ng/kg TeT and sacrificed 15 days after the treatment. In both the TeT-treated groups, no neuronal death occurred in any layers of the DG using neuronal nuclei (NeuN, a neuron nuclei maker) and Fluoro-Jade B (F-J B, a high-affinity fluorescent marker for the localization of neuronal degeneration). In addition, no significant change in glial activation in both the 2.5 and 10 ng/kg TeT-treated-groups was found by GFAP (a marker for astrocytes) and Iba-1 (a marker for microglia) immunohistochemistry. However, in the 2.5 ng/kg TeT-treated-group, the mean number of BrdU, Ki-67 and DCX immunoreactive cells, respectively, were apparently decreased compared to the control group, and the mean number of each in the 10 ng/kg TeT-treated-group was much more decreased. In addition, processes of DCX-immunoreactive cells, which projected into the molecular layer, were short compared to those in the control group. In brief, our present results show that low dosage (10 ng/kg) TeT treatment apparently decreased cell proliferation and neuroblast differentiation in the mouse hippocampal DG without distinct gliosis as well as any loss of adult neurons.

      • Prognostic Value of MGMT Promoter Methylation and TP53 Mutation in Glioblastomas Depends on IDH1 Mutation

        Several molecular markers have been proposed as predictors of outcome in patients with glioblastomas. We investigated the prognostic significance of $O^6$-methylguanine-DNA methyltransferase (MGMT) promoter methylation and TP53 mutation status dependent on isocitrate dehydrogenase 1 (IDH1) mutation in glioblastoma patients. A cohort of 78 patients with histologically confirmed glioblastomas treated with radiation therapy and chemotherapy were reviewed retrospectively. We evaluated the prognostic value of MGMT promoter methylation and TP53 mutation status with regard to progression-free survival (PFS) and overall survival (OS). It was revealed that mutations in IDH1, promoter methylation of MGMT, TP53 mutation, age, Karnofsky performance status (KFS), and extension of resection were independent prognostic factors. In patients with an IDH1 mutation, those with an MGMT methylation were associated with longer PFS (p=0.016) and OS (p=0.013). Nevertheless, the presence of TP53 mutation could stratify the PFS and OS of patients with IDH1 wild type (p=0.003 and 0.029 respectively, log-rank). The MGMT promoter methylation and TP53 mutation were associated with a favorable outcome of patients with and without mutant IDH1, respectively. The results indicate that glioblastomas with MGMT methylation or TP53 mutations have improved survival that may be influenced by IDH1 mutation status.

        A High-Throughput Method Based on Microculture Technology for Screening of High-Yield Strains of Tylosin-Producing Streptomyces fradiae

        Tylosin is a potent veterinary macrolide antibiotic produced by the fermentation of Streptomyces fradiae; however, it is necessary to modify S. fradiae strains to improve tylosin production. In this study, we established a high-throughput, 24-well plate screening method for identifying S. fradiae strains that produce increased yields of tylosin. Additionally, we constructed mutant libraries of S. fradiae via ultraviolet (UV) irradiation and/or sodium nitrite mutagenesis. A primary screening of the libraries in 24-well plates and UV spectrophotometry identified S. fradiae mutants producing increased yields of tylosin. Mutants with tylosin yield 10% higher than the wild-type strain were inoculated into shake flasks, and the tylosin concentrations produced were determined by highperformance liquid chromatography (HPLC). Joint (UV irradiation and sodium nitrite) mutagenesis resulted in higher yields of mutants with enhanced tylosin production. Finally, 10 mutants showing higher tylosin yield were re-screened in shake flasks. The yield of tylosin A by strains UN-C183 (6767.64 ± 82.43 μg/ml) and UN-C137 (6889.72 ± 70.25 μg/ml) was significantly higher than that of the wild-type strain (6617.99 ± 22.67 μg/ml). These mutant strains will form the basis for further strain breeding in tylosin production.

        OrMKK3 Influences Morphology and Grain Size in Rice

        Although morphology and grain size are important to rice growth and yield, the identity of abundant natural allelic variations that determine agronomically important differences in crops is unknown. Here, we characterized the function of mitogen-activated protein kinase 3 from Oryza officinalis Wall. ex Watt encoded by OrMKK3. Different alternative splicing variants occurred in OrMKK3. Green fluorescent protein (GFP)–OrMKK3 fusion proteins localized to the cell membrane and nuclei of rice protoplasts. Overexpression of OrMKK3 influenced the expression levels of the grain size-related genes SMG1, GW8, GL3, GW2, and DEP3. Phylogenetic analysis showed that OrMKK3 is well conserved in plants while showing large amounts of variation between indica, japonica, and wild rice. In addition, OrMKK3 slightly influenced brassinosteroid (BR) responses and the expression levels of BR-related genes. Our findings thus identify a new gene, OrMKK3, influencing morphology and grain size and that represents a possible link between mitogen-activated protein kinase and BR response pathways in grain growth.

      • Pretreated <i>Glehnia littoralis</i> Extract Prevents Neuronal Death Following Transient Global Cerebral Ischemia through Increases of Superoxide Dismutase 1 and Brain-derived Neurotrophic Factor Expressions in the Gerbil Hippocampal Cornu Ammonis 1 Area

        <P><B>Background:</B></P><P><I>Glehnia littoralis</I>, as a traditional herbal medicine to heal various health ailments in East Asia, displays various therapeutic properties including antioxidant effects. However, neuroprotective effects of <I>G. littoralis</I> against cerebral ischemic insults have not yet been addressed. Therefore, in this study, we first examined its neuroprotective effects in the hippocampus using a gerbil model of transient global cerebral ischemia (TGCI).</P><P><B>Methods:</B></P><P>Gerbils were subjected to TGCI for 5 min. <I>G. littoralis</I> extract (GLE; 100 and 200 mg/kg) was administrated orally once daily for 7 days before ischemic surgery. Neuroprotection was examined by neuronal nuclear antigen immunohistochemistry and Fluoro-Jade B histofluorescence staining. Gliosis was observed by immunohistochemistry for glial fibrillary acidic protein and ionized calcium-binding adapter molecule 1. For neuroprotective mechanisms, immunohistochemistry for superoxide dismutase (SOD) 1 and brain-derived neurotrophic factor (BDNF) was done.</P><P><B>Results:</B></P><P>Pretreatment with 200 mg/kg of GLE protected pyramidal neurons in the cornu ammonis 1 (CA1) area from ischemic insult area (<I>F</I> = 29.770, <I>P</I> < 0.05) and significantly inhibited activations of astrocytes (<I>F</I> = 22.959, <I>P</I> < 0.05) and microglia (<I>F</I> = 44.135, <I>P</I> < 0.05) in the ischemic CA1 area. In addition, pretreatment with GLE significantly increased expressions of SOD1 (<I>F</I> = 28.561, <I>P</I> < 0.05) and BDNF (<I>F</I> = 55.298, <I>P</I> < 0.05) in CA1 pyramidal neurons of the sham- and ischemia-operated groups.</P><P><B>Conclusions:</B></P><P>Our findings indicate that pretreatment with GLE can protect neurons from ischemic insults, and we suggest that its neuroprotective mechanism may be closely associated with increases of SOD1 and BDNF expressions as well as attenuation of glial activation.</P>
