Aims: Alpha-fetoprotein (AFP) is a well-known biomarker of liver cancer and liver injury. However, apparently healthy individuals having AFP elevation on heath check-ups were encountered in clinics. Their clinical features and cause of AFP elevation were not clear. Thus, we aim to investigate the clinical characteristics of the patients with an “elevated AFP level” (>7 ng/ mL cutoff), and its relation with body fat deposition in terms of hepatic steatosis and visceral adiposity. Methods: Patients having a diagnostic code of “elevated AFP level” (R772) were searched from 2009 to 2018 in a tertiary hospital. After excluding patients with any malignancies, liver cirrhosis, or viral hepatitis, 146 patients were included in the case group. As a control group, age and sex-matched 146 hepatic hemangioma (<3cm) patients were selected. Among the subgroup of case (n=49) and controls (n=49) who underwent liver CT, hepatic fat and visceral fat were measured using the pre-contrast CT image. Results: The case group showed a mean age of 48.7 years, male proportion of 64.4%, and a higher mean serum AFP level (12.76 ng/mL) than the control (2.85 ng/mL). The case group showed a higher prevalence of dyslipidemia (21.2 vs 3.4%, P<0.001) and hypertension (11.6 vs 4.8%, P=0.033), but a lower body mass index (BMI) (22.68 vs 23.85 kg/m2, P=0.003) than the control. Total-bilirubin level was higher in the case group (0.8 vs 0.5 mg/dL, P<0.001), but other laboratory results were similar to the control group. Hepatic fat measured by Hounsfield units of 8 regions of liver segments and visceral adiposity index in the subgroups of case and control were not different (59.40 vs 61.76, P=0.158 and 28.53 vs 27.32 ㎠/㎡, P=0.788, respectively). Conclusions: Asymptomatic patients with an elevated AFP level showed a higher prevalence of hypertension and dyslipidemia despite lower BMI than controls. Its relationship with abnormal fat metabolism warrants further study.

This study was performed to examine the skin phototoxicity of purified bee venom (Apis mellifera L.) collected using bee venom collector. To confirm whether the gel containing purified bee venom (BV gel) causes photototoxicity when used for the skin medicinal products, phototoxicity testing was conducted using guinea pig models. The BV gel (0.1 ml/site) was administered transdermally to guinea pigs. 8-MOP was used to introduce positive control response. After administration, the guinea pigs were irradiated with UVA (15 J/cm2) with doses based on standard phototoxicity study guidelines. In the weight measurement and clinical observation, BV gel groups didn’t show any significant changes compared with control group. BV gel groups did not show any symptoms such as erythema and edema formation of skin. This study demonstrated that BV gel has promising potential external treatment for topical uses that do not induce significant levels of skin phototoxicity.

<P>In this study, a divergent and practical solid-phase parallel diversity-oriented synthesis (DOS) strategy was successfully applied for the construction of five discrete core skeletons embedded with privileged benzopyranyl substructure. The diversity of these core skeletons was expanded through the introduction of various substituents at the R, R(1), and R(2) positions from a single key intermediate in five different pathways. More importantly, we efficiently maximized the molecular diversity through the transformation of the core skeleton itself by using the library-to-library concept and created a distinctively different collection of small molecules with the same building blocks. A 434-member polyheterocyclic benzopyran library was constructed on a scale of about 10 mg with the potential for further diversification. Without further purification, the average purity of the library is 85%.</P>

Oleuropein induces apoptosis in colorectal tumor spheres via mitochondrial fission

Background 5-Fluorouracil (5-FU) has been used as a standard chemotherapeutic agent for colorectal cancer (CRC). However, chemo-resistant cancer cells can survive under chemotherapeutic regimens, resulting in cancer recurrence. Thus, a novel approach is required to overcome chemo-resistance in CRC. Objective Recently, oleuropein (OLP), an extract derived from olive leaves, has shown anticancer eff ects and induces apoptosis in cancer cells; however, the role of OLP in 5-FU-resistant cells remains to be elucidated. Therefore, we have assessed the anticancer eff ect of OLP on tumor spheres and 5-FU-resistant cells, but also synergistic eff ect in combination of OLP and 5-FU. Results Our fi ndings revealed that OLP had a marked suppressive eff ect on tumor sphere formation capacity. Annexin V-PI staining data showed that OLP-induced apoptosis occurred in a dose-dependent manner. Mitochondrial fragmentation and mitochondrial superoxide production were observed following treatment with OLP. OLP treatment drastically suppressed the metastatic potential, including cell migration ability and anchorage-independent growth capability. In addition, combination treatment with 5-FU and OLP revealed a synergistic eff ect on cell viability in DLD-1 cells and 5-FU-resistant cells. Conclusion These fi ndings showed the suppressive eff ects of OLP against the colorectal tumor spheres and 5-FU-resistant cells. In addition, these results off er the new insights into novel therapeutic strategies for combining 5-FU and OLP in patients with chemo-resistant CRC.
