      • Slide Session : OS-RES-10 ; Oncology : Intensive Care Unit Admission is Associated with Poor Outcome in Patients with Mitochondrial DNA M.3243a>g Mutation: A Multicenter Retrospective Study

        Background: Mitochondrial DNA 3243A>G, one of the most common pathogenic mitochondrial mutations, often presents in adulthood and is associated with mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes (MELAS) and other syndromes. Due to multi-organ involvement, patients with m.3243A>G frequently require critical care, but the diagnosis often remains unrecognized. This multicenter study aimed to evaluate the prognostic factors of m.3243A>G in Taiwan, and focused on those who had been admitted to ICU. Methods: Patients diagnosed with m.3243A>G from January 1st 1997 to August 1st 2014 at the National Taiwan University Hospital were identified. Clinical features, laboratory study results, and outcomes were recorded. ICU admission course was reviewed. Prognostic factors for ICU admission, severe disability (defi ned by modifi ed Rankin Scale, mRS≥4), and death were analyzed. Results: A total of 33 patients were identifi ed, with 19 patients (58%) demonstrating the MELAS phenotype. Thirteen patients (39%) had been admitted to ICU, and 7 (54%) were diagnosed with the mutation after ICU admission. Seizure was the most common cause for ICU admission (n=8), followed by lactic acidosis (n=2), diabetic ketoacidosis (n=1), stroke (n=1), and respiratory failure (n=1). In total, 6 patients (18%) died while 11 patients (33%) developed mRS≥4. Logistic regression analysis identifi ed symptomatic CNS involvement as a signifi cant predictor of both ICU admission (OR 8.25, 95% CI 1.43-47.58, p=0.018) and development of mRS≥4 (OR 14.44, 95% CI 1.56-133.58, p=0.019); while Kaplan Meier analysis showed that ICU admission was signifi cantly associated with mortality (log-rank test p=0.012, Fig.).

        The protective effect of Acanthopanax senticosus (AS) against ethanol (EtOH)-induced apoptosis of the human neuroblastoma cell line SK-N-MC was investigated via 3-(4,5-dimothylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, flow cytometric analysis, DNA fragmentation assay, reverse transcription-polymerase chain reaction (RT-PCR), and caspase-3 assay. It was shown that cells treated with EtOH exhibit classical apoptotic features, while cells pre-treated with Acanthopanax senticosus prior to EtOH exposure showed decreased occurrence of apoptotic features. In addition, Acanthopanax senticosus pre-treatment was shown to inhibit EtOH-induced increase in caspase-3 mRNA expression and activity. These results suggest that Acanthopanax senticosus may exert a protective effect against EtOH-induced apoptosis of human neuroblastoma cells.

        중국 소비자들의 게이밍 커머스 이용 심리 및 행동에 관한 연구 : 기본심리욕구 충족감을 중심으로

        본 연구는 게이밍 커머스를 이용하는 중국 소비자의 심리 및 행동의 본질을 파악하기 위하여 기본심리욕구이론을 이론적 토대로 하여, 게이밍 커머스 이용 소비자의 ‘기본심리욕구 충족감’에 대한 선행요인과 결과요인을 밝히고자 하였다. 질적연구(연구 1)와 양적연구(연구 2)의 혼합연구방법이 사용되었는데, 연구 1은 게이밍 커머스 사용경험이 있는 총 226명의 중국 소비자들을 대상으로 게이밍 커머스에 대한 인식 및 이용동기를 서술하게 하여 수집된 텍스트 자료를 주제적 내용분석법으로 분석하였고, 연구 2에서는, 총 360명의 게이밍 커머스 사용 경험이 있는 중국 소비자들을 대상으로 설문조사를 하여 연구1에서 밝혀진 동기들이 소비자들의 ‘기본심리욕구 충족감’에 유의미하고 긍정적인 영향을 미치는지, 나아가 ‘기본심리욕구 충족감’이 긍정적 소비행동(지속사용의도 및 인앱결제의도)으로 연결되는지 검증하였다. 연구1의 주제적 내용분석 결과, 중국 소비자들이 게이밍 커머스를 이용하는 동기로 ‘가상현실’, ‘게임’, ‘소셜 네트워킹’, ‘쇼핑’, ‘기타’의 다섯 가지 차원이 도출되었다. 이차적으로, 하위 주제적 내용분석을 실시한 결과 가상현실 경험 관련 동기로는 ‘일상으로부터의 해방감’, ‘아바타 맞춤화’, ‘자아표현’, 게임 경험 관련 동기로는 ‘오락성’, ‘자아성취감’, 소셜 네트워킹 경험 관련 동기로는 ‘상호작용성’이 귀납법 비율 5% 이상의 주요하고 반복적인 이용동기로 나타났다. 연구 2에서 수집된 양적자료를 구조방적식모델을 활용해 분석한 결과, ‘아바타 맞춤화’, ‘자아표현’, ‘오락성’, ‘상호작용성’은 ‘기본심리욕구 충족감’에 유의미한 영향을 미친 반면, ‘일상으로부터의 해방감’과 ‘자아성취감’의 영향은 유의하지 않았다. 또한 ‘기본심리욕구 충족감’은 게이밍 커머스 ‘지속사용의도’와 ‘인앱결제의도’에 정적 영향을 미쳤다. 마지막으로 ‘지속사용의도’가 ‘인앱결제의도’에 유의한 정적 영향을 미쳤다. 본 연구는 글로벌 게임 시장에서 가장 큰 규모와 영향력을 행사하는 중국 소비자들을 대상으로 게이밍 커머스 이용동기, 기본심리욕구 충족감 및 행동을 밝혔다는 점에서 학문적, 실무적 의의가 있다. This study attempted to understand the nature of Chinese consumers psychological and behavior of using gaming commerce and to clarify the antecedents and consequences of ‘satisfaction of basic psychological needs’ based on the Basic Psychological Need Theory. We used a mixed-method approach of qualitative research(study 1) and quantitative research(study 2). In Study 1, a total of 226 Chinese consumers who experienced using gaming commerce were asked to describe their perception and motivation for using gaming commerce, and the collected textual data were analyzed by thematic content analysis. In study 2, an online survey of 360 Chinese users of gaming commerce was conducted to verify whether the motivations identified in study 1 have a significant, positive effect on consumers basic psychological needs satisfaction and whether basic psychological needs satisfaction leads to positive consumption behavior (i.e., continuous use intention, in-app purchase intention). As a result of study 1, the motives of Chinese consumers to use gaming commerce were classified into five dimensions: virtual reality- related experience, game-related experience, social networking-related experience, shopping- related experience, and others. With sub-thematic content analysis, liberation from daily life, avatar customization, self-expression, entertainment, self-achievement, and interaction was found to be the main motivations for using gaming commerce, with the induction ratio of 5% or above. The quantitative data collected in study 2 were analyzed using a structural equation modeling and the results are as follows. First, avatar customization, self-expression, entertainment, and interaction had a significant effect on basic psychological needs satisfaction, while the effect of liberation from daily life and self-achievement were not significant. Second, basic psychological needs satisfaction had significant effects on continuous use intention and in-app purchase intention. Lastly, continuous use intention had a significant effect on in-app purchase intention. This study has academic and practical significance in that it identified the motivations for using gaming commerce, the satisfaction of basic psychological needs, and the behavior of Chinese consumers, who have the largest scale and influence in the global game market.

        Identification of aldehyde reductase catalyzing the terminal step for conversion of xylose to butanetriol in engineered Escherichia coli

        <P>Biosynthetic pathways for the production of biofuels often rely on inherent aldehyde reductases (ALRs) of the microbial host. These native ALRs play vital roles in the success of the microbial production of 1,3-propanediol, 1,4-butanediol, and isobutanol. In the present study, the main ALR for 1,2,4-butanetriol (BT) production in Escherichia coli was identified. Results of real-time PCR analysis for ALRs in EWBT305 revealed the increased expression of adhP, fucO, adhE, and yqhD genes during BT production. The highest increase of expression was observed up to four times in yqhD. Singular deletion of adhP, fucO, or adhE gene showed marginal differences in BT production compared to that of the parent strain, EWBT305. Remarkably, yqhD gene deletion (KBTA4 strain) almost completely abolished BT production while its re-introduction (wild-type gene with its native promoter) on a low copy plasmid restored 75?% of BT production (KBTA4-2 strain). This suggests that yqhD gene is the main ALR of the BT pathway. In addition, KBTA4 showed almost no NADPH-dependent ALR activity, but was also restored upon re-introduction of the yqhD gene (KBTA4-2 strain). Therefore, the required ALR activity to complete the BT pathway was mainly contributed by YqhD. Increased gene expression and promiscuity of YqhD were both found essential factors to render YqhD as the key ALR for the BT pathway.</P>

        <P>Development of sustainable technologies for the production of 3-hydroxypropionic acid (3HP) as a platform chemical has recently been gaining much attention owing to its versatility in applications for the synthesis of other specialty chemicals. Several proposed biological synthesis routes and strategies for producing 3HP from glucose and glycerol are reviewed presently. Ten proposed routes for 3HP production from glucose are described and one of which was recently constructed successfully in Escherichia coli with malonyl-Coenzyme A as a precursor. This resulted in a yield still far from the required level for industrial application. On the other hand, strategies employing engineered E. coli and Klebsiella pneumoniae capable of producing 3HP from glycerol are also evaluated. The titers produced by these recombinant strains reached around 3 %. At its current state, it is evident that a bulk of engineering works is yet to be done to acquire a biosynthesis route for 3HP that is acceptable for industrial-scale production.</P>

      • Engineering Escherichia coli for glycolic acid production from D-xylose through the Dahms pathway and glyoxylate bypass

        <P>Glycolic acid (GA) is an ai-hydroxy acid used in cosmetics, packaging, and medical industries due to its excellent properties, especially in its polymeric form. In this study, Escherichia coli was engineered to produce GA from D-xylose by linking the Dahms pathway, the glyoxylate bypass, and the partial reverse glyoxylate pathway (RGP). Initially, a GA-producing strain was constructed by disrupting the xylAB and glcD genes in the E. coli genome and overexpressing the xdh(Cc) from Caulobacter crescentus. This strain was further improved through modular optimization of the Dahms pathway and the glyoxylate bypass. Results for module 1 showed that the rate-limiting step of the Dahms pathway was the xylonate dehydratase reaction, and the overexpression of yagF was sufficient to overcome this bottleneck. Furthermore, the appropriate aldolase gene for module 1 was proven to be yagE. The results also show that overexpression of the lactaldehyde dehydrogenase gene, aldA, is needed to increase the GA production while the overexpression of glyoxylate reductase gene, ycdW, was only essential when the glyoxylate bypass was active. On the other hand, the module 2 enzymes AceA and AceK were vital in activating the glyoxylate bypass, while the RGP enzymes were dispensable. The final strain (GA19) produced 4.57 g/L GA with a yield of 0.46 g/g from D-xylose. So far, this is the highest value achieved for GA production in engineered E. coli through the Dahms pathway.</P>

      • Identification and characterization of a thermostable endolytic β-agarase Aga2 from a newly isolated marine agarolytic bacteria <i>Cellulophaga omnivescoria</i> W5C

        <P><B>Abstract</B></P> <P>Research on the enzymatic breakdown of seaweed-derived agar has recently gained attention due to the progress in green technologies for marine biomass utilization. The enzymes known as agarases catalyze the cleavage of glycosidic bonds within the polysaccharide. In this study, a new β-agarase, Aga2, was identified from <I>Cellulophaga omnivescoria</I> W5C. Aga2 is one of four putative agarases from the W5C genome, and it belongs to the glycoside hydrolase 16 family. It was shown to be exclusive to the <I>Cellulophaga</I> genus. Agarase activity assays showed that Aga2 is an endolytic-type β-agarase that produces tetrameric and hexameric neoagaro-oligosaccharides, with optimum activity at 45°C and pH 8.0. Zinc ions slightly enhanced its activity while manganese ions had inhibitory effects even at very low concentrations. Aga2 has a K<SUB>m</SUB> of 2.59mgmL<SUP>−1</SUP> and V<SUB>max</SUB> of 275.48Umg<SUP>−1</SUP>. The K<SUB>cat</SUB> is 1.73×10<SUP>2</SUP> s<SUP>−1</SUP>, while the K<SUB>cat</SUB>/K<SUB>m</SUB> is 8.04×10<SUP>6</SUP> s<SUP>−1</SUP> M<SUP>−1</SUP>. Aga2 also showed good thermostability at 45°C and above, and retained >90% of its activity after repeated freeze-thaw cycles. Bioinformatic analysis of its amino acid sequence revealed that intrinsic properties of the protein (e.g. presence of certain dipeptides and the relative volume occupied by aliphatic amino acids) and tertiary structural elements (e.g. presence of salt bridges, hydrophobic interactions and H-bonding) contributed to its thermostability.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Endolytic β-agarase Aga2 was identified from <I>Cellulophaga omnivescoria</I> W5C. </LI> <LI> Aga2 optimum activity was at 45°C, pH 8.0, with NA4 and NA6 as major products. </LI> <LI> Aga2 was thermostable at high temperatures as well as after many freeze-thaw cycles. </LI> <LI> Intrinsic properties and tertiary structural elements confer the thermostability. </LI> </UL> </P>

        Methodological Study on the Evaluation of Face Mask Use Scale among Public Adult: Cross-Language and Psychometric Testing

        Purpose: This study aimed to establish the translation adequacy and examine the psychometric properties of FaceMask Use Scale (FMUS). Methods: This methodological study employed a cross-sectional design with repeatedmeasures. Phase 1 examined the equivalence and relevance of English and Chinese versions of FMUS. Phase2 examined the internal consistency, stability and construct validity. Different sample batches (213 universitystudents and 971 general public) were used appropriately for psychometric testing. The 2-phase data were collectedbetween January and April 2017. Results: In Phase 1, the semantic equivalence and relevance (item- and scale-levelcontent-validity-index=100%) was satisfactory. Furthermore, from 133 paired test-retest responses, the quadraticweighted kappa (.53~.73, p<.001) and Intraclass Correlation Coefficient (ICC=.81) between the English andChinese version of FMUS were satisfactory. In Phase 2, FMUS demonstrated satisfactory internal consistency(Cronbach’s ⍺=.80~.81; corrected item-total correlation coefficients=.46~.67) and two-week test-retest stability(ICC=.84). The known-groups method (t=3.08, p<.001), exploratory (71.10% of total variance in two-factor model)and confirmatory factory analysis (x2/df=4.02, Root Mean Square Residual=.03, Root Mean Square Error ofApproximation=.06, Goodness of Fit Index=.99, Comparative Fit Index=.99) were all satisfactory for establishingthe construct validity. Conclusion: The FMUS has an equivalence Chinese and English versions, satisfactoryreliability and validity for measuring the practice of face mask use. This poses clinical and research implications forthose community health nurses who works on respiratory protection. Further research should be conducted on the‘negligent practice’ of FMU.
