        SIRT5-related desuccinylation modification of AIFM1 protects against compression-induced intervertebral disc degeneration by regulating mitochondrial homeostasis

        Mitochondrial dysfunction plays a major role in the development of intervertebral disc degeneration (IDD). Sirtuin 5 (SIRT5) participates in the maintenance of mitochondrial homeostasis through its desuccinylase activity. However, it is still unclear whether succinylation or SIRT5 is involved in the impairment of mitochondria and development of IDD induced by excessive mechanical stress. Our 4D label-free quantitative proteomic results showed decreased expression of the desuccinylase SIRT5 in rat nucleus pulposus (NP) tissues under mechanical loading. Overexpression of Sirt5 effectively alleviated, whereas knockdown of Sirt5 aggravated, the apoptosis and dysfunction of NP cells under mechanical stress, consistent with the more severe IDD phenotype of Sirt5 KO mice than wild-type mice that underwent lumbar spine instability (LSI) surgery. Moreover, immunoprecipitation-coupled mass spectrometry (IP-MS) results suggested that AIFM1 was a downstream target of SIRT5, which was verified by a Co-IP assay. We further demonstrated that reduced SIRT5 expression resulted in the increased succinylation of AIFM1, which in turn abolished the interaction between AIFM1 and CHCHD4 and thus led to the reduced electron transfer chain (ETC) complex subunits in NP cells. Reduced ETC complex subunits resulted in mitochondrial dysfunction and the subsequent occurrence of IDD under mechanical stress. Finally, we validated the efficacy of treatments targeting disrupted mitochondrial protein importation by upregulating SIRT5 expression or methylene blue (MB) administration in the compression-induced rat IDD model. In conclusion, our study provides new insights into the occurrence and development of IDD and offers promising therapeutic approaches for IDD.

        Short-term Haze Exposure Predisposes Healthy Volunteers to Nasal Inflammation

        Purpose: This study aimed to investigate the impact of short-term haze exposure on nasal inflammation in healthy volunteers. Methods: Thirty-three healthy university students were assessed for nasal symptoms, nasal patency, upper and lower respiratory tract nitric oxide (NO) as well as inflammatory mediators and neuropeptides in nasal secretions before and after a 5-day haze episode. Peripheral blood mononuclear cells (PBMCs) were stimulated with particulate matter with an aerodynamic diameter of less than 2.5 μm (PM2.5), and cytokines in the supernatants were examined. Results: Mild nasal symptoms were reported by some participants during the haze episode. Objective measures of nasal patency demonstrated that nasal airway resistance was significantly increased from baseline levels, while nasal cavity volume and minimum cross-sectional area were significantly decreased. Similarly, the levels of nasal and exhaled NO, eotaxin, interleukin (IL)-5, chemokine (C-C motif) ligand 17, IL-8, substance P, nerve growth factor and vasoactive intestinal peptides in nasal secretions were significantly increased from baseline values following the haze episode. In contrast, the levels of interferon-γ, IL-10, transforming growth factor-β and neuropeptide Y were significantly decreased. Incubation with 0.1-10 μg/mL PM2.5 significantly increased release of IL-1β, IL-4, IL-5, IL-8 and IL-10 from PBMCs. Conclusions: Short-term haze exposure may lead to nasal inflammation and hypersensitivity in healthy subjects predominantly by Th2 cytokine-mediated immune responses.

      • Error-Correcting Codes Scheme for Remote Control System

        How to rise the reliability of the data transmission is one of the main problem faced by modern digital communication designer. This paper studies the error-correcting codes being suitable for the channel existing both the random error and the burst error. A new error control scheme is given. The scheme is a concatenated coding system using an interleaved Reed-Solomon code with symbols over GF (2⁴) as the outer code and a Vitcrbi-decoded convolutional code as the inner code. As a result of the computer simulation, it is proved that the concatenated coding system has a output at a very low bit error rate (BER) and can correct a lot of compound error pattern. It is suitable for the serious disturb channel existing both the random error and the burst error. This scheme will be adopted for a remote control system

        Rapid Expression of Bm46 in Bombyx mori Cell Lines, Larvae and Pupae

        In this study, ORF 46 of Bombyx mod nucleopolyhedrovirus(Bm46) fused with EGFP was expressed in Bombyx mod cell lines, larvae and pupae by BmNPV Bacmid system. Bm46 and EGFP were cloned into donor plasmid pFastBacHTb, which was transformed to competent DH10B cells containing helper and BmNPV bacmid by site-specific transposition. Recombinant bacmid was used to transfected BmN-4 cells to produce the recombinant baculovirus vBm-Bm46-EGFP. Recombination virus was injected into silkworm larvae and pupae. The expression of the fusion protein was monitored by examining green fluorescence using a fluorescent microscope. Intense fluorescence in cells and silkworm was observed at 4 days post-infection, indicating the Bm46-EGFP fusion gene was expressed successfully.

        Nonlinear Finite Element Modeling of Novel Partially Connected Buckling-Restrained Steel Plate Shear Walls

        It has been demonstrated that the buckling-restrained steel plate shear wall (SPSW) is an effi cient and economic lateral loadresisting system exhibiting high performance on initial stiff ness, ductility, shear resistance, and energy dissipation capacity. In present study, a novel partially connected buckling-restrained SPSW is presented to reduce the stiff ness requirement for the vertical boundary elements. Meanwhile, nonlinear fi nite element (FE) analysis is performed to evaluate the behavior of the proposed shear wall system so that a large expense of conducting additional test can be saved. The experimental results from the literature and the test conducted by the authors are used to establish the validation of FE models. Based on the validated FE models, a further extensive parametric study is carried out to investigate the eff ect of initial imperfection, stiff ness of boundary elements, slenderness ratio (Height/Thickness) of the infi ll panel, aspect ratio (Height/Width) of the infi ll panel, RC cover panel thickness and bolt spacing on the behavior of the partially connected buckling-restrained SPSW.

      • Preventive Effect of Actinidia Valvata Dunn Extract on N-methyl-N'-nitro-N-nitrosoguanidine-induced Gastrointestinal Cancer in Rats

        Purpose: This study was conducted to assess the preventive effect of Actinidia valvata Dunn (AVD) extract on an animal model of gastrointestinal carcinogenesis on the basis of changes in tumor incidence, cell proliferation, and apoptosis. Materials and Methods: Seventy-five male Wistar rats were divided into five different treatment groups with 15 rats in each group. Group I was given normal feed, whereas Groups II to IV were treated with 10% sodium chloride in the first six weeks and 100ug/mL of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in drinking water for 24 weeks. Group II was then given normal feed, whereas Group III was given AVD extract (0.24g/kg/day) for 12 weeks. Group IV was given AVD extract from the first week to the 36th week, whereas Group V was treated with AVD extract alone for 36 weeks. All rats were sacrificed at the end of the 36-week experiment and assessed for the presence of gastrointestinal tumors. The occurrence of cancer was evaluated by histology. Bax, Bcl-2, Caspase-3, and cyclinD1 were determined by immunohistochemical staining and Western blotting. Results: The incidences of gastric cancer were 0% in Group I, 73.3% in Group II, 33.3% in Group III, 26.7% in Group IV, and 0% in Group V. Bcl-2 and cyclinD1 expression was decreased in AVD extract treated groups, whereas Bax and Caspase-3 expression was increased. Comparison with group II revealed significant differences (p<0.01). Conclusions: AVD extract exhibits an obvious preventive effect on gastrointestinal carcinogenesis induced by MNNG in rats through the regulation of cell proliferation and apoptosis.

        Double Line Voltage Synthesis Strategy for Three-to-Five Phase Direct Matrix Converters

        This paper proposes a double line voltage synthesis (DLVS) strategy for three-to-five phase direct matrix converters. In the proposed strategy, the input and expected output voltages are divided into 6 segments and 10 segments, respectively. In addition, in order to obtain the maximum voltage transfer ratio (VTR), the input line voltages and "source key" should be selected reasonably according to different combinations of input and output segments. Then, the corresponding duty ratios are calculated to determine the switch sequences in different segment combinations. The output voltages and currents are still sinusoidal and symmetrical with little lower order harmonics under unbalanced or distorted input voltages by using this strategy. In addition, the common mode voltage (CMV) can be suppressed by rearranging some of the switching states. This strategy is analyzed and studied by a simulation model established in MATLAB/Simulink and an experimental platform, which is controlled by a DSP and FPGA. Simulation and experimental results verify the feasibility and validity of the proposed DLVS strategy.

      • Species specificity of human RPA in simian virus 40 DNA replication lies in T-antigen-dependent RNA primer synthesis

        Replication protein A (RPA) is a three-subunit protein complex with multiple functions in DNA replication. Previous study indicated that human RPA (h-RPA) could not be replaced by Schizosaccharomyces pombe RPA (sp-RPA) in simian virus 40 (SV40) replication, suggesting that h-RPA may have a specific function in SV40 DNA replication, we prepared heterologous RPAs containing the mixture of human and S. pombe subunits and compared these preparations for various enzmatic activities. Heterologous RPAs containing two human subunits supported SV40 DNA replication, whereas those containing only one human subunit poorly supported DNA replication, suggesting that RPA complex requires at least two human subunits to support its function in SV40 DNA replication. All heterologous RPAs effectively supported single-stranded(ss)DNA binding activity and an elongation of a primed DNa template catalyzed by DNA polymerase(pol) α and δ. A strong correlation between SV40 DNA replication activity and primer synthesis by pol α-primase complex was observed among the heterologous RPAs. Furthermore, T-ag showed a strong interaction with 70- and 34-kDa subunits from human, but poorly interacted with their S.pombe counterparts, indicating that the specificity of h-RPA is due to its role in RNa primer synthesis. In the SV40 replication reaction, the addition of increasing amounts of sp-RPA in the presence of fixed amount of h-RPA significantly reduced overall DNA synthesis, but increased the size of lagging strand, supporting a specific role for h-RPA in RNA primer synthesis. Together, these results suggest that the specificity of h-RPA in SV40 replication lies in T-ag-dependent RNA primer synthesis.

        Studies on the Hereditary Properties of SSR Marker in Silkworm (Bombyx mori L.)

        Two BC1 group, c108×(p50×c108) and p50×(p50×c108), one group of F2 progeny, (p50×c108) F2, and 3 SSR markers, F10539, F10626 and F10618 were used to test the hereditary properties of SSR markers in silkworm. F10539, F10626 were proved to be linkage, and F10618 was proved to be independent to those two markers. According to Mendel`s law, the recombinant value between F10539, F10626 was calculated in all of these groups, and they were 8.55% (c108BC1), 8.02% (p50BC1) and 7.81% (F2) respectively. There was dominant difference among the crossing-over value using paired-samples tests by SPSS 10.0 software. This research proved that SSR markers were co-dominant in B. mori too, and F2 progeny could be used to construct SSR linkage map although B. mori lacked of crossing over in females.
