The plant Schisandra chinensis contains a phytoestrogens, a type of naturally occurring estrogens which have multiple functions in a number of biological processes. To investigate the correlation between phytoestrogens and epigenetic modification, especially the effect of phytoestrogens on DNA methylation, sexually healthy female mice were used as an animal model in the present study. Briefly, the total RNA and protein were isolated from the ovary of mice after 7-day oral administration of Schisandra chinensis extract (SCE), while distilled water was given to the animals in the control group. Real-time PCR, Western blotting, and enzyme activity assays were performed to examine the effect of the extract of S. chinensis on Dnmt1 transcription and activity. A promoter assay was further conducted in MCF cells (ER positive) to explore also the influence of SCE on Dnmt1 transcriptional activity. The results revealed that the mRNA and protein expression levels of mouse Dnmt1 were both significantly downregulated in the treated group. The transcription of Dnmt1 was suppressed by SCE and in the E2-added group also. Meanwhile the numbers of oocytes at different stages were increased in the treated group when compared by histological analyses with those in the control group. Taken together, the results indicated that, similarly to the action of estrogen, phytoestrogens affected Dnmt1 transcription in mammals, regulating the related gene expression and cell differentiation. The findings of our examination provided also basic data and understanding for the correlation between phytoestrogens and epigenetic modification.
A Study on the Fashion Images according to the Role Types of the Chinese First Ladies

The purpose of this study is to analyze types of roles and fashion of the Chinese first ladies and provide basic data for fashion image-making of first ladies. As for the research methods, the concepts, role types, and fashion of the first ladies were considered by referring to the precedent studies and relevant literature. Then, the fashion images according to role types were analyzed by collecting the photos of Chinese first ladies from the period of the Republic of China until today. The results were as follows: First, when the roles of Chinese first ladies were categorized based on ceremonial, political, and policy roles, Zhuo Lin and Wang Yeping played a ceremonial role; Wang Guangmei, Liu Yongqing, and Peng Liyuan played a political role; and Soong Qingling, Soong Mayling, and Jiang Qing played a policy role while they were first ladies. While the first ladies of the Republic of China often wore classic Chinese styles like traditional qipao as well as western clothes which contained a western influence, the first ladies of the People’s Republic of China wore more colorful clothes and more accessories in the later period, and also often wore clothes that combined traditional and western elements. Second, first ladies who played a ceremonial role showed mannish and classic images; those who played a political role showed elegant, classic, and diverse images according to T.P.O; and those who played a policy role showed feminine, modern, and mannish images. Fashion images of the first ladies varied according to the level of political involvement, the sociocultural background of China at that time, and their personality and preference.
Therapeutic effects of systemic photodynamic therapy in a leukemia animal model using A20 cells.

<P>Photodynamic therapy (PDT) is attracting attention because of its noticeable inhibitory effects on the growth of dermatological and other solid tumors. Here, we studied the use of PDT in systemic diseases such as leukemia, lymphoma, and metastatic cancer, for which tumor formation areas cannot be clearly compartmentalized. We developed a systemic PDT method and examined its effect in a leukemia mouse model. Growth inhibition of A20 cells (H-2(d), murine B-lymphoma/leukemia, and Balb/c origin) induced by PDT/Photodithazine was evaluated by EZ-Cytox assay. After PDT, changes in cell morphology were assessed by light microscopy. Induction of apoptosis, as well as changes in the cell cycle, were assessed by fluorescence-activated cell sorting (FACS) analysis. A20 cells were injected into Balb/c mice through the tail veins, and PDT was performed. A total of 10 mg kg(-1) body weight of Photodithazine concentration was injected intravenously. After 5 min, micro photofibers (diameter, 200 μm) were inserted into the tail veins and irradiated at 1,200 J with a laser. PDT inhibited growth of A20 cells and resulted in marked morphological changes. PDT also induced apoptosis and G1 arrest. In a leukemia mouse model, systemic PDT increased the survival rate (p < 0.01). This is the first report of the effects of systemic PDT in a leukemia animal model. PDT has been applied only locally in most cases, for example to solid tumors. This study provides experimental evidence that systemic PDT could effectively be applied to systemic and spread tumors, for which tumor formation areas cannot clearly be determined.</P>

Airborne particulate matter (PM) causes oxidative stress and inflammation and could chronically harm the human respiratory and circulatory systems. The protective effect of Chrysanthemum zawadskii Herbich var. latilobum (Maxim.) Kitamura (CZL) extract in PM10-induced lung injury model was studied herein. The mechanism of PM10-induced inflammation was also investigated using human alveolar epithelial A549 cells and BALB/c mice. Histological assessment showed that CZL administration significantly attenuated PM10-induced pathological injury, inflammatory cell infiltration and lung edema. Phosphorylation of AKT, ERK and P65 was increased by PM10 exposure was reduced by CZL treatment in both in vivo and in vitro models. In addition, CZL treatment inhibited the expression of iNOS and COX-2 levels and the secretion of pro-inflammatory cytokines (TNF-α, IL-1β and IL-6) in PM10 challenged lung cells and mice. Based on the results of this study, CZL alleviated lung inflammation caused by PM10 exposure, and could be applied as a potent therapeutic candidate for the treatment measure of lung inflammatory diseases.

<P>The correct identification of the geographical origin of deer antlers is essential to quality control, as its positive physiological effects correlate with chemical components. In this study, we applied both genomics and metabolomics to the origin-identification of 101 samples from Canada, New Zealand, and Korea. The genomics identified deer species in each country but failed to categorize all the samples, due to the presence of identical species in different countries. For identical species, NMR-based metabolomics gave clean separations, compounds specific to each country were identified, and the validity was confirmed by prediction analysis. As the genomics provided unambiguous read-outs for different species, and the metabolomics cleanly distinguished among identical species from different countries, their combined use could be a robust method for origin-identification even in difficult cases.
