        SPINK1 promotes cell growth and metastasis of lung adenocarcinoma and acts as a novel prognostic biomarker

        Serine protease inhibitor Kazal type 1 (SPINK1) plays a role in protecting the pancreas against premature activation of trypsinogen and is involved in cancer progression. SPINK1 promoted LAC cells growth, migration, and invasion. Mechanistically, we found that SPINK1 promoted LAC cells migration and invasion via up-regulating matrix metalloproteinase 12 (MMP12). We observed that SPINK1 expression was only up-regulated in lung adenocarcinoma (LAC) tissues, and was an independent prognostic factor for poor survival. Our results indicate that SPINK1 might be a potential biomarker for LAC that promotes progression by MMP12. [BMB Reports 2018; 51(12): 648-653]

        Imbalance of Innate and Adaptive Immunity in Esophageal Achalasia

        Background/AimsPrevious studies reveal that immune-mediated neuroinflammation plays a key role in the etiology of esophageal achalasia. However, the understanding of leucocyte phenotype and proportion is limited. This study aim to evaluate the phenotypes of leukocytes and peripheral blood mononuclear cells transcriptomes in esophageal achalasia. MethodsWe performed high-dimensional flow cytometry to identified subsets of peripheral leukocytes, and further validated in lower esophageal sphincter histologically. RNA sequencing was applied to investigate the transcriptional changes in peripheral blood mononuclear cells of patients with achalasia. Cell-type Identification by Estimating Relative Subsets of RNA Transcripts (CIBERSORT) was used for estimating the immune cell types. A differential gene expression analysis was performed and the differential expressed genes were subjected to gene ontology, Kyoto Encyclopedia of Genes and Genomes network, protein-protein interaction network construction. ResultsAn imbalance between innate and adaptive immune cells occurred in achalasia. Specifically, neutrophils and CD8+ T cells increased both in peripheral blood and lower esophageal sphincter in achalasia. Eosinophils decreased in peripheral blood but massively infiltrated in lower esophageal sphincter. CIBERSORT analysis of peripheral blood mononuclear cells RNA sequencing displayed an increased prevalence of CD8+ T cells. 170 dysregulated genes were identified in achalasia, which were enriched in immune cells migration, immune response, etc. Proton pump inhibitor analysis revealed the intersections and gained 7 hub genes in achalasia, which were IL-6, Toll-like receptor 2, IL-1β, tumor necrosis factor, complement C3, and complement C1q A chain. ConclusionPatients with achalasia exhibited an imbalance of systematic innate and adaptive immunity, which may play an important role in the development of achalasia.

        Required Active Earth Pressure for 3D ERSS Following a Modified Mohr-Coulomb Strength Criterion

        Exploiting the linear Mohr-Coulomb strength criterion, the stability computation for earth retaining structures (ERSs) by estimating the active earth pressure has been mainly conducted by setting the plane strain 2D. In this research, the modified MC strength criterion with reduced tensile strength is put into use for the stability analysis of ERSs employing a 3D multi-cone collapse analysis mechanism. After computing the coefficient of active earth pressure, the optimal results are captured from a combination of the genetic algorithm and particle swarm optimization. Research findings are validated through comparison, while the influences of soil tensile strength threshold and 3D geometrical features over the stability measures and critical failure modes of ERSs are explored by parametric assessment. It is revealed in this work that the linear MC strength criterion derives conservative estimates of the active earth pressure. The tensile strength threshold of soil has a pronounced effect on both the objective solutions and failure shapes, especially for the ERSs under a greater dimensionless cohesion c/H. This work provides a straightforward approach to perform a 3D stability analysis of ERSs considering a tensile strength cutoff, without a pre-assumed distribution of the rupture angle.

        Unknown Input Observer Design for Takagi-Sugeno Fuzzy Stochastic System

        This paper deals with the unknown input observer design for Takagi-Sugeno (T-S) fuzzy models subjected to measurement noise and stochastic noise. The method applies the singular system theory by considering the measurement noise as an augmented system state, and then an unknown input observer based on the techniques of singular systems is developed to estimate both the system states and measurement noise simultaneously. Under a necessary assumption, the error dynamic system of the observer is free from the unknown inputs. And the observer gain matrix is determined by means of minimum covariance matrix of state residual. Two simulation examples are given to illustrate the correctness and effectiveness of the proposed method.

        A Colorimetric and Fluorescent pH Probe for Extremely Acidic Conditions and its Application in pH Test Paper

        Here, we report a highly water-soluble colorimetric and fluorescent pH probe that works in the pH range 0.2–3.0. The probe showed a rapid response, high stability, and excellent reversibility to acidity. Moreover, the fluorescence of probe was not influenced by the existence of high concentration of cations. The pKa of probe was calculated to be 2.25 ± 0.06. Furthermore, the pH test papers coated with the probe exhibited a distinct color change in acidic conditions.

        A Novel Facile and Green Synthesis Protocol to Prepare High Strength Regenerated Silk Fibroin/SiO2 Composite Fiber

        In this work, regenerated silk fibroin (RSF) and silicon dioxide (SiO2) composite fiber was successfully extrudedby wet spinning method. The effect of SiO2 addition on structure of the composite fiber at microscopic level is studied, whichsubsequently correlated to the mechanical performance. The best concentration ratio for composite fiber is identified byscreening SiO2 concentration from 0.025 w/w% to 0.5 w/w%. The experimental results revealed that the SiO2 at a lowconcentration of 0.1 w/w% was well distributed. The breaking stress, breaking strain and Young’s modulus at 0.1 w/w% SiO2addition of the RSF fibers increased considerably compared to the neat RSF fibers from 243±3 to 458±21 MPa, 51±4 % to54±7 % and 6.34±0.55 to 11.69±1.12 GPa, respectively. To the best of our knowledge, this is the first report for RSF/SiO2composite fiber. We believed the insight provided in this report which looks into the structural evolution should be beneficialto the future design and building of other advanced functional fibers.

        STABILITY ANALYSIS UPON HIGH-PRESSURE COMMON RAIL FUEL INJECTION SYSTEM UNDER MULTIPLE INJECTION MODES

        Fuel injection stability of high-pressure common rail fuel injection system (HPCRFIS) under multiple injection modes is a recognized technical problem. To explain it essentially, a numerical model based on bond graph method, which provides a standardized series of symbol system to describe various components in different physical fields as well as interactive effect analysis tool for different energy categories, was used to investigate the problem. As HPCRFIS is a typical nonlinear system with multi-physical coupling, it is very difficult to analyze system stability directly. Therefore, the system was linearized at typical moments. The system matrix for each typical moment was obtained thereby. To investigate the influential mechanism of system stability deeply, the variations of rank and eigenvalues distributions of state matrices were analyzed. The results show that the rank of state matrices changes abruptly when the needle or control valve moves. In terms of system stability, the oscillation characteristics are subject to the movement of needle or control valve. The ending of the former injection causes the system to show damping oscillation characteristics before the latter injection starts.The motion of control valve makes more contribution to system stability than that of needle, which could be concluded by Lyapunov stability criterion.

        Hydration, Compressive Strength and Durability of Eco-friendly Cement Mortars Containing Recycled Brick Powder and Metakaolin

        Grinding brick waste into powder for use as a supplementary cementitious material has shown to be a promising recycling method for the reuse of construction and demolition waste.However, the strength and durability of concrete containing recycled brick powder (RBP) will significantly decrease due to the poor pozzolanic activity of RBP, limiting its wider applications. This study attempted to overcome these defects through metakaolin (MK) addition. The hydration, compressive strength, durability, and environmental impact of eco-friendly mortars containing RBP and MK were investigated. The results showed that MK promoted the early-stage hydration of cement, creating a suitable hydration environment for the hydration of RBP at a later age. As a result, the volume fractions of low-density calcium silicate hydrate (C-S-H) and high-density C-S-H increased according to the nanoindentation test results. In addition, the strength, chloride penetration resistance, and reinforcement bar corrosion resistance of the blended mortars obviously improved due to the synergistic effect of RBP and MK. Lastly, we achieved CO2 emissions that were 22.1 – 30.2% lower and energy consumption that was 21.4 – 27.0% lower by incorporating 20% RBP and 5 – 15% MK.

        Changpingibacter yushuensis gen. nov., sp. nov., isolated from fluvial sediment in Qinghai Tibet Plateau of China

        Two facultatively anaerobic, short rod-shaped, non-motile, Gram-stain-positive, unknown bacterial strains (JY-X040T and JY-X174) were isolated from fluvial sediments of Tongtian River in Yushu Tibetan Autonomous Prefecture, Qinghai province, China. Cells formed translucent, gray, round and convex colonies, with a diameter of less than 0.5 mm after 5 days of incubation at 30°C on brain heart infusion-5% sheep blood agar. The 16S rRNA gene sequence similarity between strain JY-X040T and Fudania jinshanensis 313T is 93.87%. In the four phylogenetic trees constructed based on the 16S rRNA gene and 423 core genes, the two isolates form an independent branch, phylogenetically closest to F. jinshanensis 313T, but could not be classified as a member of the genus Fudania or any other genus of the family Arcanobacteriaceae. The DNA G + C content of strain JY-X040T was 57.8%. Calculation results of average nucleotide identity, digital DNADNA hybridization value and amino acid identity between strain JY-X040T and F. jinshanensis 313T are 69.9%, 22.9%, and 64.1%. The major cellular fatty acids were C16:0 (23%) and C18:1ω9c (22%). The cell-wall peptidoglycan type was A5α (L-Lys-L-Ala-L-Lys-D-Glu). The polar lipids comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside and four unidentified components. The whole-cell sugars contained rhamnose and ribose. MK-10(H4) was the sole respiratory quinone. The minimum inhibitory concentration of streptomycin was 32 μg/ml. All physiological, biochemical, chemotaxonomic and genomic characteristics support that strains JY-X040T and JY-X174 represent members of a novel species in a new genus, Changpingibacter yushuensis gen. nov., sp. nov. The type strain is JY-X040T (GDMCC 1.1996T = KCTC 49514T).

        Isolation and characterization of tick-borne Roseomonas haemaphysalidis sp. nov. and rodent-borne Roseomonas marmotae sp. nov.

        Four novel Gram-negative, mesophilic, aerobic, motile, and cocci-shaped strains were isolated from tick samples (strains 546T and 573) and respiratory tracts of marmots (strains 1318T and 1311). The 16S rRNA gene sequencing revealed that strains 546T and 573 were 97.8% identical to Roseomonas wenyumeiae Z23T, whereas strains 1311 and 1318T were 98.3% identical to Roseomonas ludipueritiae DSM 14915T. In addition, a 98.0% identity was observed between strains 546T and 1318T. Phylogenetic and phylogenomic analyses revealed that strains 546T and 573 clustered with R. wenyumeiae Z23T, whereas strains 1311 and 1318T grouped with R. ludipueritiae DSM 14915T. The average nucleotide identity between our isolates and members of the genus Roseomonas was below 95%. The genomic G+C content of strains 546T and 1318T was 70.9% and 69.3%, respectively. Diphosphatidylglycerol (DPG) and phosphatidylethanolamine (PE) were the major polar lipids, with Q-10 as the predominant respiratory quinone. According to all genotypic, phenotypic, phylogenetic, and phylogenomic analyses, the four strains represent two novel species of the genus Roseomonas, for which the names Roseomonas haemaphysalidis sp. nov. and Roseomonas marmotae sp. nov. are proposed, with 546T (= GDMCC 1.1780T = JCM 34187T) and 1318T (= GDMCC 1.1781T = JCM 34188T) as type strains, respectively.
