        Production and Characterization of Monoclonal Antibodies Against Primo Vascular System of Rat

        Background: The primo vascular system (PVS) has been difficult to detect due to its small diameter and translucent features of the threadlike network. Thus, contrast-enhancing dyes including Alcian blue, Trypan blue and Janus green B had to be used for finding and taking out PVS from rat and mouse. Objective: Generation of monoclonal antibodies (mAbs) against PVS of rat was intended to use as a detector for PVS and a biological tool for functional study of PVS. Materials and methods: Primo vessel (PV) and Primo node (PN) were isolated from organ surfaces of rat and then their proteins were isolated and injected into mouse as an immunogen. The classical traditional method was applied for production of mAbs against PVS. The various techniques, such as cell fusion, screening of hybridoma, ELISA, Western blotting (WB), immunofluorescence microscopy (IF), and limiting dilution, were used to generate mAbs against PVS. Results: Among 16 mAbs generated, 4 representative mAbs were characterized with their specificities in ELISA, WB, and IF. α-rPVS-m1-1 and α-rPVS-m4-6 had strong binding affinities to PVS in both ELISA and WB but did not show specificities in IF at all. On the contrary, α-rPVS-m3-2 and α-rPVS-m3-4 almost did not respond in WB but had strong binding affinities in ELISA and specificities in IF. Two mAbs stained predominantly at extra cellular matrix and cell membrane of PVS of rat in IF, and they were able to discriminate PVS from blood vessel (BV) and lymphatic vessel (LV). Conclusions: 4 representative mAbs against PVS of rat were characterized by ELISA, WB, and IF. α-rPVS-m3-2 and α-rPVS-m3-4, which had strong specificities in IF, can be used as a tool in discriminating PVS from other similar tissuesand in elucidate biological function of PVS.

        TsMIP6 enhances the tolerance of transgenic rice to salt stress and interacts with target proteins

        Aquaporins (AQPs), a large family of channel proteins in plants, play an important role in regulating the balance of osmotic potential in cells. We isolated an AQP gene, TsMIP6, from the halophyte Thellungiella salsuginea and functionally characterized it in transgenic rice (Oryza sativa). This gene belongs to a subfamily of tonoplast intrinsic proteins and is localized at the plasma membrane. Real-time PCR showed that expression of TsMIP6 in shoots or roots of T. salsuginea was markedly induced by salinity, whereas its ectopic expression in ‘Kitaake’ lines of rice significantly increased plant tolerance to salt stress. Physiological data suggested that TsMIP6 is involved in regulating ion homeostasis and water channel activity in salt-stressed transgenic rice. Heterologous expression analysis indicated that TsMIP6 specifically interacts with a member of the glycoside hydrolase family 64 protein #617 in yeast cells. This suggests that the relationship between TsMIP6 and #617 has a crucial role in mediating osmotic balance in plant cells. Moreover, TsMIP6 might help to modulate the transport of some neutral molecules and may function through a pathway regulating solute equilibrium to maintain osmotic potential.

      • 잠재적인 당뇨병 생체지표로서의 일반타액단백질(CSP1)

        Saliva is a bodily fluid secreted by three pairs of major salivary glands and by many of minor salivary gland. The human common salivary protein 1(CSP1) is one of variety of molecules in saliva and its function remains to be determined. The saliva CSP1 levels of 17 DM patients and 9 normal adults were quantified by ELISA system. As a probe, the mAb-CSP1 was used for Western blotting of human saliva and immunohistochemistry of various human tissues. The study showed that the saliva CSP1 concentration of diabetes mellitus(DM) patients was much higher than of healthy controls. The salivary gland was the only tissue stained with mAb-CSP1 by immunohistochemistry among the various human tissues. Quantification of CSP1 concentration by ELISA showed that the median values of DM patients and healthy adults were 4.65(3.36-6.90)μg/ml and 3.74(2.86-5.02)μg/ml, respectively. Student t-test results indicated that there was a statistically significant difference between the two groups(p<0.026). Therefore, the significant difference of CSP1 levels between the two groups indicated that CSP1 is a potential biomarker for detection or screenig of DM patients.

        Gonorrhea, one of the most common adult diseases of human society, is caused by infection with gonococcus (Neisseria gonorrhoea) through sexual contact between humans and humans, such as sexual intercourse. Gonorrhea causes inflammation of the urogenital system in both males and females, and if not treated at the right time, it can cause a large aftereffect such as infertility or blindness. Therefore, in recent years, the importance of early diagnosis that accurately diagnoses and prescribes at an appropriate time is emphasized. The purpose of this study was to produce monoclonal antibody (mAb) against to N. gonorrhoea and to use for quick diagnosis of whether people were infected with N. gonorrhoea. Spleen from immunized mouse was extracted after N. gonorrhoea was injected as an immunogen. The spleen cell was fused with myeloma SP2 cell, and then hybridoma cells producing mAb against N. gonorrhoea specifically were generated by classical method for mAb. The generated 33 mAbs will be utilized for research material as well as for diagnostic reagent for diseases.

        Indications of the sedimentary environments by the sedimentary characteristics and trace elements of Ordovician Majiagou Formation, Northern Shaanxi salt basin

        The abundant geochemical information on elements in the autogenous sedimentary rocks can help reconstruct the paleoenvironment of the stratigraphic sedimentation period. Based on the analysis of sedimentary characteristics of Ordovician Majiagou Formation and the trace elements in limestone, dolomite and gypsum rocks in Northern Shaanxi salt basin, the relationship between the distribution of trace elements of Majiagou Formation and the sedimentary environment is discussed. The results display that Majiagou Formation is mainly composed of the sedimentation of carbonate rocks and evaporites, and the subfacies of open platform and restricted platform are developed. The trace element analysis shows that the deposition period of the Majiagou Formation is dominated by dry hot climate, and there are alternating dry and wet seasons in climate evolution. In middle Ordovician, the sedimentary environment of Majiagou Formation was undergoing the transformation of oxygen enrichment-oxygen deficiency-anoxic environment.

      • A New Technology of Comprehensive Islanding Detection

        With the widely application of the distributed generation technology, micro grid islanding detection technology has been greatly developed. A new type of islanding detection technique is proposed in this paper. The new detection technology has realized the rapid, efficient, stable and reliable islanding detection, and achieved the zero non detection zone. At the same time, which put money into limit in minimum range. This paper presents the principle of islanding comprehensive detection technology. This paper designs and describes the method to realize the new islanding detection technology. The new technology makes up for a series of shortcomings,such as the non-detection zone (NDZ) is larger of the local testing technology. And it also overcomes a great many shortcomings of the remote detection technology, such as a big investment and too single.

        Characterization of fermented pomegranate juice: ACE inhibitory activity under in vitro digestion, antioxidant capacity, phenolics composition, chemical properties and sensory evaluation

        Consuming pomegranate juice (PJ) is beneficial for hypertensive regulation because of the phenolic compounds in PJ and their inhibitory activity on angiotensin-I-converting enzyme (ACE). To better utilize bioactive function of food, microorganism fermentation has been adopted to alter phenolic metabolism. This study confirms that even under in vitro digestion, fermented PJ (FPJ) maintains higher ACE inhibitory activity than that of PJ. The main phenolic compounds in PJ were compared either under fermentation or in vitro digestion. This study finds that fermentation promotes antioxidant capacity of PJ. The chemical properties of FPJ are evaluated and the corresponding relationship with bioactivities is analyzed. A sensory evaluation comparison is conducted between FPJ and PJ, furnishing interesting information for consumers. This study highlights the relationship between ACE inhibitory activity of PJ and phenolic composition under fermentation and in vitro digestion, providing novel insights for diet regulation of phenolic-rich FPJ in ACE inhibition therapy.

        LINC00319 promotes cancer stem cell-like properties in laryngeal squamous cell carcinoma via E2F1-mediated upregulation of HMGB3

        Laryngeal squamous cell carcinoma (LSCC) is one of the most common subtypes of head and neck malignancies worldwide. Long intervening/intergenic noncoding RNAs (LINCRNAs) have been recently implicated in various biological processes that take place in the setting of laryngeal cancer, but the regulatory role of LINC00319 in LSCC remains largely unknown. The current study aimed to elucidate the regulatory effect of LINC00319 on the development and progression of LSCC via high-mobility group box 3 (HMGB3). Microarray-based analysis was initially conducted to identify differentially expressed long noncoding RNAs, after which the expression of LINC00319 and HMGB3 in LSCC tissues and cells was determined accordingly. CD133 + CD144 + TU177 cells were subsequently isolated and transfected with LINC00319 overexpression vector (oe-LINC00319), short hairpin RNA (sh)-LINC00319, sh-HMGB3, sh-E2F transcription factor 1 (E2F1), and oe-E2F1, as well as their corresponding controls. The proliferative, invasion, self-renewal, and tumorigenic abilities of CD133 + CD144 + TU177 cells were then evaluated. Our in vitro findings were further confirmed following subcutaneous injection of cells expressing the corresponding plasmids into nude mice. LINC00319 and HMGB3 expressions were elevated in LSCC cells and tissues. LINC00319 increased HMGB3 expression by recruiting E2F1. Furthermore, the stimulatory role of LINC00319 on the proliferation, invasion, self-renewal ability, and tumorigenicity of CD133 + CD144 + TU177 cells was achieved by upregulating HMGB3 via recruitment of E2F1. The in vitro findings were also confirmed by in vivo experiments. Taken together, these data show that downregulating LINC00319 in CD133 + CD144 + TU177 cells may serve as a potential anticancer regimen by inhibiting the proliferation and invasion of cancer stem cells in LSCC.

        Effects of non-uniform inflow on the spatial and temporal distribution of impeller excitation in axial flow pumps in water tunnels

        The effect of non-uniform inflow on the spatial and temporal distribution of impeller excitation is investigated. The experiments are first conducted to verify the hydraulic performance of the axial flow pump and to provide a reference for the accuracy of the numerical simulations. Subsequently, the effect of non-uniform inflow on the spatial and temporal distribution of the flow field and impeller load is investigated. The results indicate that the non-uniform inflow modifies the velocity distribution at the impeller inlet, resulting in alterations in the attack angle and subsequently affecting the radial and circumferential distribution of the impeller load. The extreme difference in the head between the different blades can reach up to 36 % of the total head. Furthermore, velocity pulsation and attack pulsation exhibit more pronounced fluctuations in the low-speed region, and there exists a strong correlation between the amplitude of unsteady forces and the magnitude of the impeller load.
