Hyaluronic acid (HA) is a linear high-molecular-weight polysaccharide with useful biomedical applications. Streptococcus zooepidemicus, a typical HA-producing bacterium, requires an animal-derived nitrogen source such as tryptone, peptone or sheep blood as a nutrient. Sixteen non-animal-derived (NAD) nitrogen sources were tested as a replacement for the expensive animal-derived nitrogen sources, which may have safety issues. Among the sixteen tested NAD nitrogen sources, a yeast-derived nitrogen source (YE 0251) showed the highest HA productivity, which was equivalent to the control HA production medium containing tryptone in a 5-L jar and in 3,000-L industrial fermentations. In the 3,000-L fermentation, YE 0251 increased cell mass (dry cell weight) and HA production by 11% and 8%, respectively, compared with the control HA production medium. The fmal specific volumetric productivity (0.41 g/L h) was improved by about 70% after reducing the fermentation time from 20 h to 12 h, compared to the conventional production medium.

Background: Recombinant IFN-beta products have been approved for the treatment of several diseases. Recently, Inbionet, Co. has successfully developed rhIFN-beta product, Genebeta. This study was performed in a purpose of confirming the biological activity of Genebeta by comparison with another recombinant IFN-beta, Beneserin which was already approved. Methods: In vitro antiporliferative activity, regulation of MHC class I expression, antiviral activity, regulation of natural killer cell cytotoxicity were studied for functional comparison of these two rhIFN-beta products. Results: Proliferation assay data showed that two recombinant IFN-beta inhibited T cell and Daudi cell proliferation to a similar extent. When antiviral activity was compared, there was no significant difference between them. The treatment of these two rhIFN-beta also resulted in a similar level of upregulation of class I expression on the surface of A549 cells. Cytotoxicity experiment against tumor cells revealed that both of rhIFN-beta could enhance NK cell cytotoxicity to comparable level. Conclusion: All comparison data indicated that rhIFN-beta products, Genebeta and Beneserin have comparable potentials as a biological response modifier.

We have studied the effect of the hydrogen ion doping on the free surface of back channel etched a Si:H TFTs. The hydrogen ion doping on the back channel etched TFT reduces the off-state photo leakage current by one order of magnitude. The reduction of the photo-leakage current is due to the creation of defect states at the free surface by hydrogen ion doping. After hydrogen ion doping on the free surface, the field effect mobility change little, but the threshold voltage increases, which is due to the state creation in a-Si:H by ionized H ions. Furthermore, the off-state leakage current under frontlight illumination can be reduced by 2 orders magnitude by hydrogen ion doping on the back channel of a Si:H TFT.

In order to vitrify the combustible dry active waste (DAW) generated from Korean Nuclear Power Plants, a glass formulation development based on waste composition was performed. A borosilicate glass, DG-2, was formulated to vitrify the DAW in an induction cold crucible melter (CCM). The processability, product performance, and volume reduction effect of the candidate glass were evaluated using a computer code and were measured experimentally in the laboratory and CCM. The glass viscosity and electrical conductivity as the process parameters were in the desired ranges. Start-up and maintaining glass melt of the candidate glass were favorable in the CCM. The product of the glass product such as chemical durability, phase stability, and density was satisfactory. The vitrification process using the candidate glass was also evaluated assuming that it was operated as economically as possible.

The antigenic potential of CFA-001, cefazolin, a cephalosporin derivative produced by an enzymatic semisynthesis, was determined in Hartley guinea pigs. A battery of tests employed consisted of active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA), and indirect hemagglutination test (IHA). The results were as follows: 1) In ASA, no signs attributable to anaphylaxis was observed in guinea pigs sensitized with CFA-001, whereas OVA-sensitized animals induced severe anaphylactic symptoms; 2) guinea pigs did not produce antibodies against CFA-001 when sensitized with or without Freund's complete adjuvant (FCA) in homologous PCA tests. Meanwhile, antibodies against ovalbumin (OVA) were clearly detected; 3) No CFA-001-specific hemagglutination was observed in the IHA using sera obtained from CFA-001-sensitized guinea pigs. These results suggest that CFA-001 has no antigenicity potential in guinea pigs.

Background: Recent evidence demonstrates that high doses of epoetin-alpha(EPO-α) can be administrated at extended intervals, despite its relatively shortserum half-life. However, no prospective randomized trials on the effects ofextended dosing intervals of EPO-α compared with darbepoetin-alpha (DA-α) havebeen performed. This study was designed to investigate whether a single biweekly(Q2W) administration of a high dose of EPO-α is as effective as DA-α for anemia inchronic kidney disease (CKD) patients not receiving dialysis. Methods: Sixty non-dialysis CKD patients were equally randomized to either Q2Wsubcutaneous EPO-α (10,000 unit) or DA-α (50 μg) therapy groups for the first6 weeks. After a 6-week washout period, the participants of the EPO-α and DA-αtreatment groups switched to the alternate regimen for 6 weeks. The meanhemoglobin (Hb) levels after erythropoiesis stimulating agent (ESA) therapy andpercentage change in Hb levels from baseline to the end of the study were analyzed. Results: The mean Hb levels of postESA therapy increased significantly comparedwith those of preESA therapy in both ESA regimens. The percentage increase in Hblevels and erythropoietin resistance index did not show a significant differencebetween the different ESA regimens. No difference was observed between theregimens regarding mean Hb levels after ESA therapy. Additionally, there were noserious adverse effects leading to withdrawal from treatment. Conclusion: Biweekly high doses of EPO-α therapy may be equally as effective asQ2W DA-α therapy in maintaining target Hb levels in non-dialysis CKD patients.

<P>A–D–A conjugated polymer, PTPD-TVT, containing thienopyrroledione and thiophene–vinylene–thiophene (TVT) units was synthesized as an electron donor for organic photovoltaic devices. It possesses a small bandgap and has excellent coplanarity and high hole mobility. To further enhance the interchain interactions between the polymer chains, a selenophene–vinylene–selenophene (SVS) unit was also introduced and copolymerized to form the PTPD-SVS polymer. Devices made from PTPD-TVT and PTPD-SVS have rather promising power conversion efficiencies (PCEs) of 4.87 and 5.74%, respectively. The higher PCE value for solar cells based on PTPD-SVS was attributed to an enhanced carrier mobility resulting from stronger interchain aggregation in the BHJ active layer. Twenty-three fungal strains were isolated from meju that had originated from the Sunchang province, the famous location for making fermented soybean foods in Korea. The restriction fragment length polymorphism (RFLP) of the internal transcribed spacer (ITS) region of the rDNA (ITS-RFLP) was applied to differentiate the isolated fungal strains. First, the ITS region by polymerase chain reaction (PCR) with specific primers was amplified and then cleaved the products with different restriction enzymes. Cleavage of the amplified fragments with the restriction enzymes AluI, HaeIII, HhaI, and TaqI revealed extensive polymorphisms. The ITS-RFLP results highly correlated with ITS sequence analysis. All of the 23 fungal strains were classified into 5 groups by ITS-RFLP analysis. Aspergillus oryzae was the major fungal strain isolated from Sunchang meju (12 out of 23), while Aspergillus fumigatus was the next most frequently isolated strain (7 out of 23). In contrast, it was found that Fusarium asiaticum, Aspergillus sydowii, and Arthrinium sp. were the minor fungal strains in meju.
